Acetazolamide alleviates motion sickness by inhibiting inner ear carbonic anhydrase 2 and reducing endolymph volume.

Motion sickness is common in aerospace, aviation and maritime operations, and travel by vehicles or ships. Existing preventive and therapeutic drugs for motion sickness induce central nervous system (CNS)-related side effects; therefore, there is an urgent need to find new anti-motion sickness targets and to develop novel drugs with reduced adverse effects. In this study, we found that rotational stimulation significantly upregulated carbonic anhydrase 2 (CA2) expression in the inner ears of guinea pigs and mice. Pretreatment with acetazolamide (AZ), an inhibitor of carbonic anhydrase, effecti

On-road evaluation of sickness-less motion planning in automated vehicles: 'Head motion first' helps!

With the rapid advancement of automated driving, motion sickness (MS) has gathered significant attention from both academia and industry. Advanced sensing, decision-making, and control systems in autonomous vehicles offer promising opportunities to mitigate MS in next-generation transportation. Traditionally, objective metrics such as Motion Sickness Dose Value (MSDV) and Motion Sickness Incidence (MSI) are derived directly from vehicle motion states to quantify MS severity. However, these metrics often overlook the filtering or amplification effects of human body dynamics on the motion stimul

The molecular and genetic mechanisms of the vestibular disorders: A mini review.

The vestibular disorders, which generally manifest themselves as nausea, vomiting, and dizziness or vertigo, have troubled many people. Increasing evidence suggests that certain vestibular disorders are hereditary, and gene variants contribute to the onset and susceptibility of these disorders. This review summarizes the latest research progress, especially the molecular and genetic aspects of several common vestibular disorders, including Meniere's disease, vestibular migraine, benign paroxysmal positional vertigo, and motion sickness, with the aim of encouraging more fundamental research and

Effect of low-dose propofol infusion with sevoflurane versus propofol-only total intravenous anesthesia on postoperative nausea and vomiting in high-risk patients: a single-blind randomized controlled clinical trial.

BACKGROUND: Postoperative nausea and vomiting (PONV) is a common complication following general anesthesia, and may lead to delayed recovery and prolonged hospital length of stay. While propofol has been shown to reduce PONV risk, volatile anesthetics like sevoflurane are associated with a higher incidence. OBJECTIVE: This study compares the incidence of PONV within 24 h after surgery between propofol based total intravenous anesthesia (TIVA) and a hybrid technique using low-dose propofol infusion with sevoflurane in patients with a prior history of PONV and/or motion sickness. DESIGN: A prosp

Effects of vestibular habituation training on synaptic plasticity in the medial prefrontal cortex of mice with motion sickness.

BACKGROUND: Motion sickness (MS) is triggered by vestibular conflict, yet how vestibular habituation training (VHT) engages higher-order cortical circuits remains unclear. OBJECTIVE: To test whether VHT modulates medial prefrontal cortex (mPFC) activity and structural plasticity in a rotational MS model. METHODS: Male C57BL/6J mice were assigned to control (CON), motion sickness (MS), or vestibular habituation training (VHT) groups. MS was induced by a single 4-g session (four 1-min bouts; total 4 min), whereas VHT mice received 14 daily sessions (30 × 1-min bouts/day; total 30 min/day). Beh

Cybersickness-induced EEG responses in curved monitor and head-mounted display.

Cybersickness is a common issue in virtual reality (VR), yet addressing it remains challenging due to the lack of quantitative evaluation methods. To quantitatively assess cybersickness, this study examined electroencephalography (EEG) changes and Simulator Sickness Questionnaire (SSQ) scores using two different display types: a curved monitor and a head-mounted display (HMD). Thirty-one male participants (25.0 ± 1.9 years old) were exposed to cybersickness-inducing VR content developed using the Unity engine, featuring pitch, yaw, and roll rotations. The results revealed two key findings. Cybersickness Abatement From Repeated Exposure Generalizes Across Experiences.

Cybersickness, or sickness caused by virtual reality (VR), represents a significant threat to the usability of VR applications. Repeated exposure to the same VR stimulus causes a reduction in cybersickness, referred to as Cybersickness Abatement from Repeated Exposure (CARE). This study examined whether the benefits of CARE generalize across distinct VR contexts, which was operationalized as three distinct games (a climbing game, a puzzle game, and a stealth survival game). Participants played a VR game for up to 20 minutes. Those in the Repeated Exposure condition played one VR game (either a

Effects of anticipatory auditory cues and non-driving related tasks on motion sickness in automated vehicles.

In the context of highly automated vehicles, motion comfort deserves crucial attention for their widespread adoption. Passengers of automated vehicles are prone to motion sickness from diminished cognizance of the driving environment, and from engaging in non-driving related tasks (NDRTs). Awareness about upcoming motion and mental distraction from NDRTs have the potential to mitigate motion sickness. The current study used a motion-based driving simulator where 31 participants performed easy and hard versions of an NDRT in the presence and absence of spatialized anticipatory auditory cues dis

Motion sickness in stop-and-go passenger rides: influence of individual characteristics, contextual, behavioural, and physiological parameters.

Progress in road vehicle automation is tainted by an increased occurrence of passenger motion sickness. Yet, these advances drive the design of car cockpits towards user-centric approaches, such as monitoring passenger well-being. In this framework, the present study aimed to identify robust predictors of motion sickness severity based on data from two experimental studies reproducing a stop-and-go passenger ride. The data from 54 participants tested in a total of 106 identical sessions involving gaze restriction to a visuomotor activity were analysed. Individual characteristics, contextual, b

Cyclizine Toxicity and Misuse in an Adolescent With Eating Disorder and Depression: A Case Report.

Cyclizine is a first-generation H1 antihistamine widely used for the treatment of nausea and motion sickness. In addition to its antiemetic properties, it has anticholinergic and central nervous system effects that may confer psychoactive potential. Although generally regarded as a low-risk medication, increasing evidence suggests a potential for misuse, particularly among adolescents and individuals with mental health disorders or substance-use vulnerabilities. Published data on cyclizine toxicity are limited, and paediatric reports of intentional overdose remain scarce. We report a case of i
