Comparison and correlation of in vitro and in vivo approaches for determining Pseudomonas aeruginosa bacteriophages activity.

The rise of infections caused by multidrug-resistant bacteria has driven the search for alternative therapies, with Pseudomonas aeruginosa representing a major target. Among these alternatives, the use of bacteriophages (phage therapy) employs these viruses that specifically infect bacteria to fight infections. Although phage efficacy has been demonstrated both in vitro and in vivo, few studies have directly correlated these two dimensions. This study aimed to evaluate, compare, and correlate in vitro and in vivo phage activity. For this, three strictly lytic bacteriophages (ph9027, ph1461, an

Epidemiology and antimicrobial resistance analysis of staphylococcus aureus isolated from women, children and neonates in a maternal and child specialty hospital in Southeast China.

BACKGROUND: Staphylococcus aureus (S. aureus) is a common bacterium posing a significant global health threat, with methicillin-resistant S. aureus (MRSA) epidemiology varying widely by region, time period, and population. Moreover, antibiotic management strategies differ for vulnerable groups including neonates, children, and pregnant women. This study investigates the clinical distribution and antimicrobial resistance profiles of S. aureus isolates from a maternal-child specialty hospital, with a particular focus on MRSA epidemiology. METHODS: Herein, we retrospectively enrolled all non-dup

COMPREHENSIVE ASSESSMENT OF BIOFILM FORMATION AND ANTIMICROBIAL RESISTANCE OF STAPHYLOCOCCUS IN PURULENT-INFLAMMATORY DISEASES.

INTRODUCTION: Treatment of biofilms is a priority in purulent surgery. A biofilm consists of 75-85% extracellular polymeric matrix and 15-20% microbial cells. Polysaccharides, proteins, and extracellular DNA within the matrix protect bacteria from adverse environmental factors (pH, antibiotics, phagocytosis). Communication between bacteria occurs through the "quorum sensing" system. Bacteria within biofilms are 100-1000 times more resistant to antibiotics compared to planktonic forms. This is explained by the limited penetration of antibiotics into the matrix and the reduced metabolic activity

The effect of selective decontamination on antimicrobial resistance in intensive care patients: a systematic review and meta-analysis.

Selective Digestive Decontamination (SDD) and Selective Oropharyngeal Decontamination (SOD) are applied to prevent infections amongst Intensive Care Unit (ICU) patients and have proven to reduce mortality and infection rates. However, concerns about the development of antimicrobial resistance persists, specifically after ICU discharge. The aim of this study was to assess the occurrence of antimicrobial resistance during ICU stay and after ICU discharge in patients who had received SDD, or SOD compared to control, explicitly at individual-patient level. A previous systematic review, which inclu

Intestinal spirochetosis in the modern era: a decade-long clinicopathologic study.

OBJECTIVES: Intestinal spirochetosis (IS) is infrequently identified in about 1% of human colonic biopsy specimens. On routine sections, it is diagnosed by identifying a thick band of basophilia on the apical surface of colonic mucosa. Intestinal spirochetosis is caused by Brachyspira pilosicoli and Brachyspira aalborgi, but whether it represents a commensalism or a pathogenic process remains debated. METHODS: We studied 175 cases of IS diagnosed at our institution over a 10-year period. The clinicopathologic features were reviewed using hematoxylin and eosin (H&E)-stained slides, along with

Comparative immunoproteomics of Campylobacter jejuni in acute campylobacteriosis and asymptomatic individuals.

Campylobacter jejuni is a leading cause of bacterial gastroenteritis worldwide; however, the molecular determinants associated with symptomatic infection remain incompletely understood. In this study, an immunoproteomic approach was applied to characterize the antigenic landscape of C. jejuni during acute campylobacteriosis and to identify infection-associated proteins targeted by the human humoral immune response. Sera from patients with laboratory-confirmed acute campylobacteriosis (Group A) and asymptomatic individuals without recent gastrointestinal disease (Group B) were used to immunocap

Hospital-treated infectious diseases and polygenic susceptibility in relation to heart failure and cardiac remodelling: evidence from the UK Biobank and ARIC cohorts.

AIMS: Infectious diseases (IDs) are increasingly recognized as precipitants of heart failure (HF). However, the impact of diverse pathogens and genetic susceptibility on HF and cardiac remodelling remains unclear. METHODS AND RESULTS: We analysed 480 154 UK Biobank (UKB) participants and 9816 Atherosclerosis Risk in Communities (ARIC) participants free of HF at baseline. Hospital-treated IDs were ascertained with 931 ICD-10 codes and modelled as time-varying exposures. Incident HF and HF with preserved (HFpEF) and reduced ejection fraction (HFrEF) were identified through hospital records and

Intrapleural Gentian Violet Irrigation for Refractory Methicillin-Resistant Staphylococcus aureus Empyema: A Case Series.

Methicillin-resistant Staphylococcus aureus (MRSA) empyema remains challenging despite guideline-based management. Gentian violet (GV), an antiseptic dye, has been sporadically used for MRSA infections but not systematically evaluated in pleural disease. We analysed nine consecutive patients with MRSA empyema treated with intrapleural GV irrigation following surgical debridement between 2011 and 2018. All patients successfully completed GV irrigation without procedural complications. Bacterial clearance was achieved within a median of 5 days (IQR, 4-8), which was within the range observed in t

Recent Advances in the Characterization of Uropathogenic Escherichia coli Associated With Resistance and Virulence Mechanisms.

Globally, urinary tract infections (UTIs) constitute a major concern as one of the most frequent bacterial infections and a major public health concern, particularly in developing countries like India. Among the various uropathogens, uropathogenic Escherichia coli (UPEC) is the principal causative agent of UTIs in both community-acquired and hospital-associated settings. UPEC strains are typically identified based on the presence of specific virulence factors (e.g., fimH, pap, sfa, afa) and phylogenetic grouping (mainly groups B2 and D), along with serotyping and molecular methods such as poly

Evidence of cefiderocol-persistence in Pseudomonas aeruginosa in vitro biofilms and in a murine infection model.

BACKGROUND: Persistent and viable but non-culturable (VBNC) Pseudomonas aeruginosa cells hamper the eradication and contribute to the recurrence of biofilm-related infections, especially in cystic fibrosis (CF) patients, often experiencing difficult-to-treat lung infections. The siderophore-cephalosporin cefiderocol, which hijacks bacterial iron-uptake systems, has emerged as a last-resort antibiotic against antibiotic-resistant Gram-negative bacteria and represents a desirable therapeutic option; however, there is still limited information about its impact on bacterial persisters. METHODS: P

Development and validation of a rabbit model of Pseudomonas aeruginosa hyperdynamic septic shock for preclinical drug development.

Translating preclinical findings into clinical success remains a major barrier in the development of antimicrobials targeting Pseudomonas aeruginosa, particularly for severe infections such as sepsis and septic shock. Rodent models frequently fail to predict therapeutic efficacy in clinical sepsis. To address this gap, we developed and validated a rabbit model of hyperdynamic septic shock incorporating intensive care unit (ICU)-style monitoring and supportive care to better reflect human clinical practice. Rabbits challenged intravenously with P. aeruginosa progressed predictably from hyperdyn
