Spermidine ameliorates homosalate-induced mouse oocyte defects via regulation of mitochondria and cytoskeleton.

Homosalate (HMS) is an organic ultraviolet filter widely used in personal care products. Although its endocrine-disrupting properties are recognized, its impact on oocytes remains poorly characterized. Through network pharmacology, Smart-seq2 sequencing, and in vitro models, our findings indicate that HMS exposure delayed meiotic progression at metaphase I (MI) in mouse oocytes. HMS treatment led to persistent oxidative stress, DNA damage, and early apoptosis. It also impaired mitochondrial function. Treated oocytes exhibited decreased membrane potential (ΔΨm), reduced ATP production, and down

Anti-Inflammatory Effects of Artocarpin-Loaded Chitosan Microparticles on Inflammatory Chondrocytes.

Artocarpin is a promising bioactive compound with anti-inflammatory potential; however, its limited aqueous solubility may restrict its direct biological application. Therefore, artocarpin-loaded chitosan microparticles (CSPs/AE) were prepared as a biocompatible carrier to improve artocarpin delivery. This study aimed to evaluate the cytocompatibility, antioxidant activity, and anti-inflammatory effects of CSPs/AE in lipopolysaccharide (LPS)-stimulated chondrocytes. CSPs/AE maintained high cell viability (>80% at 0.125 and 0.5 mg/mL) and showed significantly lower cytotoxicity than free artoca

Effects of mechanical loading on cranial suture mesenchymal stem cell proliferation.

Sutures are the primary sites of cranial bone growth, allowing the skull to accommodate the growing brain. External mechanical stimulus has been shown to delay suture fusion and induce tissue remodelling. Recent studies suggest that in vivo cyclic loading increased proliferation of mesenchymal stem cells (MSC) in the coronal suture. The overall aim of this study was to understand how many loading sessions (exposure-response) and how long after loading (time-course) did MSC proliferation increase in the coronal suture. In the exposure-response analysis, mice underwent 1, 3, or 5 loading session

Magnetic-Assisted Fractionation of Bone Marrow Cells into Subsets Differing in CD45 Expression Levels, Surface Phenotypes and Functional Properties.

Cells of higher organisms express numerous cell surface proteins, and their spectrum and level of expression are directly related to cells' functions. The technology of mass cell selection based on the surface protein expression levels may be highly important both for basic research and cell therapy applications. We have previously developed a method of magnetic selection of cells differing in surface marker expression levels, which we term here MACS-MEL (Magnetic-Assisted Cell Selection by Marker Expression Levels). The method demonstrated its effectiveness in the artificial model system, nam

Intraoperative Analgesic Requirements in Dogs Receiving Retromammillary or Erector Spinae Plane Blocks for Thoracolumbar or Lumbar Hemilaminectomy: A Single-Center Retrospective Cohort Study.

Thoracolumbar and lumbar hemilaminectomy in dogs requires effective perioperative analgesia. The erector spinae plane (ESP) and retromammillary (RM) blocks are ultrasound-guided regional anesthesia techniques targeting the dorsal branches of spinal nerves, but comparative clinical data are lacking. This retrospective cohort study compared perioperative analgesic requirements and complication rates between ESP (n = 62) and RM (n = 207) blocks in client-owned dogs undergoing hemilaminectomy. The primary outcome was intraoperative opioid rescue analgesia; secondary outcomes included rescue analge

Preclinical pharmacokinetic modeling and clinical translation of SIG001, a novel anti-cancer therapeutic antibody targeting on cancer-derived sialylated IgG.

INTRODUCTION: Sialylated immunoglobulin G (SIA-IgG) has emerged as a novel therapeutic target selectively overexpressed in epithelial-derived malignancies. The humanized monoclonal antibody SIG001 binds specifically to tumor-associated SIA-IgG and is regarded as a promising first-in-class candidate for the broad-spectrum treatment of epithelial cancers. METHODS: The pharmacokinetic (PK) behavior of SIG001 was evaluated in both cynomolgus monkeys and mice following intravenous administration. Utilizing modeling approaches, we projected the first-in-human (FIH) starting dose and the human pharm

Prenatal Δ9-tetrahydrocannabinol exposure rewires glucocorticoid-hippocampal crosstalk and impairs memory under stress in adolescent rat offspring.

BACKGROUND: Stress is a major determinant of psychopathology, with coping and cognitive impairment arising from abnormal glucocorticoid activity and brain-derived neurotrophic factor (BDNF)-dependent neuroplasticity in the hippocampus. The endocannabinoid system modulates stress and hippocampal functioning since early development, thus its early-life disruption may program vulnerability to stress-related cognitive deficits. METHODS: We investigated whether prenatal exposure to tetrahydrocannabinol (pTHC; 2 mg/kg, gestational days 5-20) alters spatial learning and memory in adolescent offsprin

Neural correlates of insomnia and hyperarousal.

This chapter provides an exhaustive review of the neural substrates associated with insomnia and hyperarousal, considering the chronic disturbance of sleep from the complex perspective of a reciprocal biopsychosocial model. It focuses on several critical abnormalities observed on the structure and function of neuroanatomy, especially the hypersensitive reaction of the amygdala and insular cortex along with reduced reactivity of the prefrontal cortex that results in the development of emotional dysregulation and a continuous feeling of danger. At the circuitry level, it is possible to observe a

Evolutionary dynamics of plant cell wall-degrading enzymes reflects feeding ecology in weevils.

Many herbivorous beetles depend on plant cell wall-degrading enzymes (PCWDEs) to access the nutritious plant cell content. However, the evolution of PCWDEs in weevils, the most diverse lineage of herbivores, remains poorly understood. Using transcriptomic and genomic analyses of 45 weevil species spanning the majority of existing subfamilies, we identified a total of 13 different PCWDE families, revealing high variability between species. Interestingly, the PCWDE repertoires tracked dietary specialization, with convergent reductions in fungivorous taxa. Furthermore, PCWDE gene family dynamics

Bioherbicidal activity and putative mode of action of Dittrichia graveolens against Amaranthus palmeri and A. retroflexus.

BACKGROUND: Challenges in controlling Amaranthus weeds and rising cases of herbicide resistance necessitate the development of alternative weed control strategies. Plant-derived extracts may serve as valuable sources of phytotoxic chemicals. Nevertheless, their species-specific effects on germination and early seedling development require investigation. This study evaluated the allelopathic activity of aqueous extracts of Dittrichia graveolens on Amaranthus palmeri and A. retroflexus. METHODS: Aqueous extracts of D. graveolens were evaluated against A. palmeri and A. retroflexus under control

Mechanistic Pathways of Wound Healing: From Hemostasis to Scar-Free Regeneration.

Wound healing is the critical process regulated through a harmonious coordination of cellular, molecular, pathological, and systemic communications. Normally, healing occurs in four overlapping phases, which are modified and controlled by various growth factor interactions and signaling pathways. Disruption of the tightly coordinated wound-healing cascade, together with aberrant protein signaling, mitochondrial dysfunction, antimicrobial resistance, sustained fibroblast activation, and maladaptive epigenetic reprogramming, drives pathological tissue repair culminating in chronic non-healing wo

Investigating the Impact of Protein Supplementation from Different Sources on Advanced Glycation End Products in Older Adults in Singapore Following a Healthy Dietary Pattern.

Background/Objectives: Protein supplementation may elevate advanced glycation end products (AGEs) through higher dietary AGE (dAGE) content and increase amino acid-driven endogenous formation. Protein supplementation from different sources may differentially affect AGE levels, possibly through differences in amino acid composition, digestion and metabolism, but evidence from randomized controlled trials (RCTs) remains limited. This study aimed to examine the effect of protein supplementation from either casein (animal-based) or soy (plant-based) protein isolates on circulating, skin, and dAGE

Loose-patch recordings reveal differential Kv7-dependent effects on spontaneous excitatory activity in control and epileptic CA1.

INTRODUCTION: The non-inactivating potassium channels Kv7.2 and Kv7.3 contribute to the neuronal resting membrane potential and regulate neuronal excitability. Previous studies have indicated a transcriptional downregulation of these channels in temporal lobe epilepsy, both in patients and in the pilocarpine-induced animal model. However, the functional consequences of this alteration for synaptic activity have not yet been fully elucidated. METHODS: To investigate the role of Kv7 channels in epileptic tissue, we used the pilocarpine model of temporal lobe epilepsy. We performed loose-patch c

Targetome-defined miR-181c signaling from extracellular vesicles governs periodontal MSC fate via RNF150-MAP3K5.

BACKGROUND: In regenerative settings, robust differentiation of human mesenchymal stromal cells (hMSCs) requires precise coupling between post-transcriptional regulation and kinase-driven pathways to achieve optimal therapeutic efficacy. miRNA-mediated regulation is widely implicated in stem cell fate control, yet its mechanistic contribution to lineage commitment remains incompletely defined. METHODS: Multi-conditioned periodontal ligament-derived hMSCs were developed as the regenerative model for alveolar bone. Extracellular vesicle (EV)-encapsulated miRNAs were delineated through microarra

Overexpression of FGFR1 in AgRP neurons protects against obesity and improves glucose homeostasis.

BACKGROUND: The rising prevalence of metabolic disorders highlights the need to identify targetable pathways. Although hypothalamic AgRP neuron signaling influences energy balance, the beneficial role of fibroblast growth factor receptor 1 (FGFR1) within these neurons remains incompletely understood. METHODS: We employed an AAV-mediated CRISPR/Cas9 activation system to achieve AgRP neuron-specific overexpression of FGFR1 in adult mice. Metabolic phenotypes were assessed during high-fat diet (HFD) feeding, and mechanistic insights were gained using GT1-7 hypothalamic cells. RESULTS: FGFR1 ove
