Enzymatic peptide macrocyclization via indole-N-acylation.

Indole N-acylation is chemically challenging, due to the low nucleophilicity of the indole nitrogen. Although a few similar transformations have been proposed in the biosynthesis of indole-containing natural products, their enzymatic basis remains elusive. Here, we show that BulbE TE is an N-acylindole-forming macrocyclase involved in the biosynthesis of the non-ribosomal cyclopeptide bulbiferamide. BulbE catalyzed macrocyclization not only via the indole nitrogen, but also via a primary amine and an alcohol. The uncommon catalytic residue Cys731 in BulbE TE was indispensable for the nucleophi…

Enamine Acylation Enabled Desymmetrization of Malonic Esters.

Asymmetric enamine alkylation represents a powerful tool for stereoselective C-C bond formation; in contrast, the development of enantioselective enamine acylation remains elusive. Here, we report that a chiral phosphoric acid can render an in-situ-formed enamine to undergo a stereoselective intramolecular α-carbon acylation, providing an alternative approach for the synthesis of useful pyrrolinones and indolinones bearing tetrasubstituted stereocenters. Utilizing an effective integration of the desymmetrization strategy and bifunctional organocatalysis, the first example of enantioselective e…

Ion channel regulation by protein S-acylation.

Protein S-acylation, the reversible covalent fatty-acid modification of cysteine residues, has emerged as a dynamic posttranslational modification (PTM) that controls the diversity, life cycle, and physiological function of numerous ligand- and voltage-gated ion channels. S-acylation is enzymatically mediated by a diverse family of acyltransferases (zDHHCs) and is reversed by acylthioesterases. However, for most ion channels, the dynamics and subcellular localization at which S-acylation and deacylation cycles occur are not known. S-acylation can control the two fundamental determinants of ion…

Enantiodivergent deprotonation/acylation of α-amino nitriles.

Back to 'base'ics: The title reaction of enantioenriched α-ureidonitriles was found to proceed in a highly enantiodivergent manner despite the intermediacy of stereolabile α-nitrile metallocarbanions. Enantiodivergence is dependent upon the base used. For the less basic hexamethyldisilazides (HMDS), deprotonation in which a metal (M) cation is precomplexed with an electrophile is proposed. LDA=lithium diisopropylamide.

Acylation of protein lysines by trichloroethylene oxide.

Stable lysine adducts were formed in proteins following reaction with trichloroethylene (TCE) oxide, the major reactive compound generated by the metabolism of TCE. The order of formation of these adducts is N(6)-formyllysine > N(6)-(dichloroacetyl)lysine >> N(6)-glyoxyllysine, with the ratio being influenced by the particular protein. Protein lysine adducts were also analyzed following the enzymatic oxidation of TCE with several different cytochrome P450 (P450) enzyme systems. The ratio of formyl/dichloroacetyl lysine adducts was influenced by the enzyme system that was used. Chloral and TCE

A light-driven enzymatic enantioselective radical acylation.

Enzymes are recognized as exceptional catalysts for achieving high stereoselectivities1-3, but their ability to control the reactivity and stereoinduction of free radicals lags behind that of chemical catalysts4. Thiamine diphosphate (ThDP)-dependent enzymes5 are well-characterized systems that inspired the development of N-heterocyclic carbenes (NHCs)6-8 but have not yet been proved viable in asymmetric radical transformations. There is a lack of a biocompatible and general radical-generation mechanism, as nature prefers to avoid radicals that may be harmful to biological systems9. Here we re

Detection and Analysis of S-Acylated Proteins via Acyl Resin-Assisted Capture (Acyl-RAC).

Protein palmitoylation is a lipid modification where a palmitoyl group is covalently attached via a thioester linkage to one or more cysteines on a substrate protein. This modification, catalyzed by a group of enzymes named DHHC enzymes after their conserved Asp-His-His-Cys motif, plays a significant role in regulating the localization, stability, and function of a wide range of cellular and viral proteins. By influencing how and where proteins interact within the cell, palmitoylation is essential for various cellular processes, including signaling pathways, membrane dynamics, and protein-prot

A broad inhibitor of acyl-acyl carrier protein synthetases.

The acyl-acyl carrier protein synthetase enzyme enables some bacteria to scavenge free fatty acids from the environment for direct use in lipids. This fatty acid recycling pathway can help pathogens circumvent fatty acid synthase (FAS) inhibition with established antibiotics and those in clinical development. AasS enzymes are surprisingly hard to identify as they show high sequence similarity to other adenylate forming enzymes, and only a handful have been correctly annotated to date. Four recently discovered AasS enzymes from Gram negative bacteria, Chlamydia trachomatis, Neisseria gonorrhoea

CuCl-Promoted β-Acylation of Cyclopropanols with Thioesters.

CuCl-induced cyclopropyl alcohol ring-opening followed by β-acylation with thioester is reported. Cyclopropanols and thioesters with various substitution patterns were successfully subjected to the reaction, and a series of 1,4-dicarbonyl compounds were synthesized. The mechanistic investigation revealed the involvement of Cu(I)-homoenolate, which reacts with the thioester via oxidative addition. Both experimental and computational evidence were found. Operational simplicity, high chemo- and regioselectivity, and good to excellent yields are the core features of the presented approach.

S-acylation of TDP-43: PALMing down aggregation?

S-acylation is well known for regulating protein stability and trafficking. In a recent issue of Molecular Cell, Xu et al.1 reveal a distinct, aggregation-suppressing function of this posttranslational lipid modification: S-acylation of the RNA-binding protein TDP-43 antagonizes poly(ADP-ribose)-driven condensation. Moreover, reduced S-acylation levels are linked to ALS pathogenesis.

Electrostatic effects in acylation of hemoglobin by aspirins.

Carboxylate substituents added to the salicylate ring increase the effectiveness of a variety of aspirins and diaspirins in acylating hemoglobin. Even more effective are a series of monoesters of dicarboxylate derivatives. Bis(5-carbomethoxysalicyl)fumarate and -succinate at 5 mM concentrations modify approximately 100% of the hemoglobin in solution and should alter the aggregation behavior of sickle hemoglobin.

Post-translational acylation of proteins in colorectal cancer.

Accumulating preclinical evidence indicates that protein acylation, a functionally diverse class of post-translational modifications, is implicated in the regulation of protein stability, enzymatic activity, subcellular localization, molecular interactions, and gene transcription. In colorectal cancer (CRC), this modification family has been linked to proliferation, invasion, metastasis, metabolic reprogramming, therapeutic resistance, and immune escape. This review critically summarizes recent advances in five major acylation types associated with CRC: crotonylation, succinylation, lactylatio…

Post-translational acylation of proteins in cardiac hypertrophy.

Acylations are post-translational modifications in which functional groups are attached to amino acids on proteins. Most acylations (acetylation, butyrylation, crotonylation, lactylation, malonylation, propionylation and succinylation) involve lysine but cysteine (palmitoylation) and glycine (myristoylation) residues can also be altered. Acylations have important roles in physiological and pathophysiological processes, including cardiac hypertrophy and related cardiovascular diseases. These post-translational modifications influence chromatin architecture, transcriptional regulation and metabo…
