Noncovalent interactions (NCIs) are critical elements of molecular recognition in a wide variety of chemical contexts. While NCIs have been studied extensively for closed-shell molecules and ions, very little is understood about the structures and properties of NCIs involving free radical intermediates. In this report, we describe a detailed mechanistic study of the enantioselective radical hydroamination of alkenes with sulfonamides and present evidence suggesting that the basis for asymmetric induction in this process arises from attractive NCIs between a neutral sulfonamidyl radical interme

Although evidence has mounted in recent years for the biosynthetic relevance of [4 + 2] cycloaddition reactions, other cycloadditions have received much less attention. Herein we used density functional theory (DFT) calculations to assess the viability of nitrone-alkene (3 + 2) cycloaddition reactions proposed to occur during the biosynthesis of several alkaloid natural products (flueggines and virosaines). The results of our calculations indicate that these reactions have low enough intrinsic barriers and diastereoselectivity that they can proceed without enzymatic intervention.

We describe the implementation of an iron catalyzed cyano-fluoroalkylation of unactivated alkenes that leverages inexpensive trifluoroacetic acid (CF3CO2H) and related carboxylic acids (CFnRCO2H) as radical fluoroalkyl sources, together with tosyl cyanide (TsCN) as a low-toxicity cyanation reagent. Notably, the mild and redox-neutral conditions of the method are compatible with a broad range of substrates, including alkenes containing biologically relevant motifs (40 examples, up to 83% yield). The resulting β-fluoroalkylated nitriles can be diversely derivatized to afford other valuable fluor

A visible-light-driven metal-free method for the arylation of alkenes with aryl iodides and formate salt has been developed. The reaction exhibited a broad substrate scope for both α,α-disubstituted alkenes and diversely substituted aryl iodides. Combined experimental and computational mechanistic studies highlighted the essential role of the carbon dioxide radical anion (CO2˙-) in the transformation. This mild photocatalytic strategy provides practical access to valuable arylated alkene products.

While energy transfer photocatalysis has recently been used in the synthesis of multisubstituted polycyclic alkanes as benzene bioisosteres, the use of highly reactive noble-metal and organo-photocatalysts often leads to rapid generation of diradical intermediates, triggering polymerization side reactions and limiting functional-group tolerance. Here we show that heteroleptic copper(I)-BINAP complexes containing bidentate (pyrazolyl)pyridine ligands act as effective triplet photosensitizers for cycloadditions between bicyclo[1.1.0]butanes and alkenes. These copper catalysts, with their extende

Herein, we report a visible-light-driven 1,2-alkylpyridylation of alkenes via the deacylation of unactivated ketones, which exhibits broad substrate compatibility with various alkenes, dihydroquinazolinone derivatives, and substituted 4-cyanopyridines. Gram-scale synthesis and late-stage modification of bioactive molecule derivatives were successfully achieved. Mechanistic studies confirm a radical pathway, and a plausible photoredox mechanism is proposed.

α,β-Unsaturated aldehydes are ubiquitous and versatile synthetic building blocks in organic synthesis. Nevertheless, the direct construction of such skeletons via site-selective β-formylation of readily available alkenes still remains challenging and largely unexplored. Herein, we disclose a visible-light photoredox-catalyzed selective β-formylation strategy of various alkenes, employing bench-stable α-chloro-N-methoxyphthalimide as a practical formyl radical precursor. This mild photoredox protocol delivers diverse α,β-unsaturated aldehydes with broad substrate generality toward electron-rich

We report a photoredox-catalyzed three-component sulfonaminoalkynylation of alkenes with N-aminopyridine salts and potassium alkynyltrifluoroborate salts. This aminoalkylation reaction underwent a radial/polar crossover mechanism, which was distinguished from the previous reports. A variety of β-alkynylated sulfonamides were obtained in moderate to excellent yields. The versatility of this method was further evidenced by its successful application in modifying biological molecules in advanced stages of development.

The direct aminative difunctionalization of alkenes offers a powerful strategy for synthesizing valuable amine-containing compounds. However, achieving this transformation enantioselectively remains a significant challenge in both synthetic chemistry and biocatalysis. In this study, we engineered a nonheme iron enzyme, quercetin 2,3-dioxygenase from Bacillus subtilis (BsQueD), to catalyze three distinct alkene aminofunctionalization reactions with high efficiency and enantiocontrol. Through directed evolution, we developed an optimized BsQueD variant capable of producing a wide array of chiral

Herein, we report a ligand-controlled, amide-assisted, nickel-catalyzed regiodivergent hydroacylation of unactivated alkenes with readily available acyl chlorides. This strategy enables the efficient and selective synthesis of amino ketones with diverse regioselectivities, including α- and β-migratory insertion, as well as Markovnikov-type addition, all governed by ligand tuning. The reaction proceeds under mild conditions, exhibits broad functional group tolerance, and offers a straightforward and cost-effective approach to structurally diverse branched ketones.

Primary sulfonamides are inexpensive, naturally occurring, and chemically diverse molecules with significant relevance in drug development. In recent years, late-stage functionalization strategies to generate sulfonamidyl radicals from sulfonamides have emerged as a critical focus in synthetic chemistry. Here, we present a complementary method for activating primary sulfonamides to generate sulfilimines, enabling the efficient synthesis of aminoalcohol and imidazoline derivatives via visible-light photoredox catalysis. This approach featured broad functional group compatibility, operational si

Given the widespread presence of fluoroalkyl functionalities in bioactive molecules, the development of fluoroalkylation reactions with bench-stable and easy-to-use fluoroalkylating reagents is highly desirable. In addition, realization of mono-, di-, tri-, or polyfluoroalkyation usually requires distinct types of fluoroalkylating reagents under different or even harsh reaction conditions, and a universal method to accomplish different hydrofluoroalkylation of alkenes is lacking. Herein, the use of quaternary fluoroalkyl alcohols is reported as the universal fluoroalkylating reagents to readil

Hydrochlorination of alkenes is a practical strategy for accessing organic chlorides. Herein, we report the hydrochlorination of unactivated alkenes via a hydrogen atom transfer/halogen atom transfer process using earth-abundant and biocompatible FeCl3 as a chlorine source under extraordinarily mild reaction conditions. The protocol is easy to operate with notable features such as excellent chemoselectivity, remarkable efficiency, a broad substrate scope, and good functional group tolerance. Importantly, the synthetic utility is highlighted by scaled-up reactions, late-stage derivatizations of

Direct functionalization of C(sp3)-H bonds allows rapid access to valuable products, starting from simple petrochemicals. However, the chemical transformation of non-activated methylene groups remains challenging for organic synthesis. Here, we report a general electrochemical method for the oxidation of C(sp3)-H and C(sp2)-H bonds, in which cyclic alkanes and (cyclic) olefins are converted into cycloaliphatic ketones as well as aliphatic (di)carboxylic acids. This resource-friendly method is based on nitrate salts in a dual role as anodic mediator and supporting electrolyte, which can be reco

In this report, we develop a new strategy for visible-light-induced, copper-catalyzed three-component difluoroalkyl thiocyanidation of alkenes to construct a series of important difluorothiocyanate compounds. The new approach can also be applied to perfluorothiocyanate compounds, even target molecules containing drug/natural product skeletons. Mechanistic studies provide that the copper complex plays a dual role, as both the photoredox catalyst for electron transfer and the cross-coupling catalyst for C-SCN bond formation.

Herein, we report a mild and practical protocol for the α-perchloroalkyl β-heteroarylation of alkenes using available chloroform as the dichloromethyl source via α-amino radical-mediated halogen-atom transfer. Various substrates are compatible under mild reaction conditions, providing the corresponding products in moderate to good yields. This strategy gives an efficient and convenient method for the introduction of chloroalkyl motifs into N-heteroarenes. The control experiment demonstrates that the α-amino radical generated in situ is a key intermediate in the transformation.

Catalytic asymmetric dicarbofunctionalization of tethered alkenes has emerged as a promising tool for producing chiral cyclic molecules; however, it typically relies on aryl-tethered alkenes to form benzene-fused compounds. Herein, we report an enantioselective cross-electrophile divinylation reaction of nonaromatic substrates, 2-bromo-1,6-dienes. The approach thus offers a route to new chiral cyclic architectures, which are key structural motifs found in various biologically active compounds. The reaction proceeds under mild conditions, and the use of chiral t-Bu-pmrox and 3,5-difluoro-pyrox

An atom-economic intermolecular radical addition reaction of acyloxy nitroso compounds to electron-deficient alkenes mediated by visible light is reported. The starting nitroso derivatives are readily prepared by oxidation of the corresponding oximes prepared from ketones and the overall transformation represents an oxidative coupling of a ketone with a Michael acceptor. The cascade proceeds smoothly under mild conditions, providing a series of valuable functionalized oximes in moderate to good yields. Mechanistic studies suggest that these cascades proceed via addition/coupling processes that
