Adhesion strength differential of human ligament fibroblasts to collagen types I and III.

Fibroblasts embedded in the amorphous healing tissue matrix of ligaments migrate into damaged sites during the inflammatory process that precedes the formation of new connective tissue after ligament injury. Cell motility involved in this migration is strongly influenced by cellular adhesion to proteins of the extracellular matrix. The adhesion mechanism of interest in this study is the attachment of fibroblasts from the anterior cruciate and medial collateral ligaments to types I and III collagen, two fibrillar collagens secreted by fibroblasts during tissue repair. Types I and III collagen c

Localization of types I and III collagen and fibronectin in the developing mouse palatal shelves.

The detailed distribution of types I and III collagen and fibronectin was studied in the developing mouse palatal shelf using a double immunofluorescence technique. Just prior to palatal shelf elevation, type I collagen showed denser localization on the nasal side than on the oral side of the middle hard palate region. Such a distribution extended as far as the posterior hard palate region but not to the soft palate region. In type III collagen and fibronectin, this uneven distribution was not so distinct even in the middle hard palate region during the same period. The correspondence of the m

Presence of type I and VI collagen mRNAs in endothelial cells in cutaneous neurofibromas.

In situ hybridization and peroxidase anti-peroxidase immunodetection were used in the same tissue sections to elucidate the spatial distribution of collagen gene expression in cutaneous neurofibromas, particularly in relation to blood vessels; the latter structures were identified by the presence of factor VIII-related antigen. The data indicate a clear relationship between the vascular structures and sites of locally elevated expression of type I and VI collagen genes. Specifically, some, but not all, blood vessels were surrounded by stromal cells highly active in expressing pro alpha 1(I) an

Immunohistochemical localization of collagen types I, II, III, and IV in pterygium tissues.

Immunohistochemical localization of collagen types I, II, III, and IV in the pterygium was demonstrated by type-specific antibodies. The stromata of pterygium and normal conjunctival tissues contained collagen types I, II, and III, while the normal corneal stroma showed collagen types I and III but not collagen type II. Collagen type IV was located in the epithelial and capillary endothelial basement membranes of pterygium and normal conjunctival tissues. These results suggest that collagens in pterygial tissues are derived from those in the conjunctival tissues.

Collagen remodeling and increased type I collagen density following non-ablative monopolar radiofrequency for vulvar laxity: A prospective pilot study.

INTRODUCTION: Vulvar laxity is associated with alterations in dermal extracellular matrix structure, particularly collagen organization, resulting in reduced tissue firmness and elasticity. Energy-based technologies have been proposed as minimally invasive approaches for tissue remodeling. The aim of this study was to evaluate the clinical and histological effects of a single session of non-ablative monopolar radiofrequency on vulvar laxity, with emphasis on collagen remodeling. METHODS: This prospective study included nine women aged 30-60 years with vulvar laxity. Vulvar biopsies were obtai

Introduction of the human pro alpha 1(I) collagen gene into pro alpha 1(I)-deficient Mov-13 mouse cells leads to formation of functional mouse-human hybrid type I collagen.

The Mov-13 mouse strain carries a retroviral insertion in the pro alpha 1(I) collagen gene that prevents transcription of the gene. Cell lines derived from homozygous embryos do not express type I collagen although normal amounts of pro alpha 2 mRNA are synthesized. We have introduced genomic clones of either the human or mouse pro alpha 1(I) collagen gene into homozygous cell lines to assess whether the human or mouse pro alpha 1(I) chains can associate with the endogenous mouse pro alpha 2(I) chain to form stable type I collagen. The human gene under control of the simian virus 40 promoter w

Type-I Collagen Polypeptide-Based Composite Nanofiber Membranes for Fast and Efficient Bone Regeneration.

The clinical treatment of bone defects includes allogeneic bone transplantation and autologous bone transplantation. However, they all have their own limitations, and the scope of application is limited. In recent years, bone tissue engineering scaffolds based on a variety of materials have been well developed and achieved good bone regeneration ability. However, most scaffold materials always face problems such as high biotoxicity, leading to inflammation and poor bioactivity, which limits the bone regeneration effect and prolongs the bone regeneration time. In our work, we prepared hydroxyap

Molecular mechanisms and clinical manifestations of rare genetic disorders associated with type I collagen.

Type I collagen is an important structural protein of bone, skin, tendon, ligament and other connective tissues. It is initially synthesized as a precursor form, procollagen, consisting of two identical pro-α1(I) and one proα2(I) chains, encoded by COL1A1 and COL1A2, respectively. The N- and C- terminal propeptides of procollagen are cleavage by N-proteinase and C-proteinase correspondingly, to form the central triple helix structure with Gly-X-Y repeat units. Mutations of COL1A1 and COL1A2 genes are associated with osteogenesis imperfecta, some types of Ehlers-Danlos syndrome, Caffey diseases

Pulmonary and diaphragmatic pathology in collagen type I α1 mutant mice with osteogenesis imperfecta.

BackgroundOsteogenesis imperfecta (OI) is most often caused by mutations in type I collagen genes. Respiratory complications have been largely attributed to spine and ribcage deformities. We hypothesized that direct involvement of the pulmonary parenchyma and/or diaphragm by the disease may occur.MethodsIn Col1a1Jrt/+ mice, a model of severe dominant OI, mean linear intercept length (Lm) was used to assess the distal airspace size. Cross-sectional area (CSA) and myosin heavy chain (MyHC) phenotype of the diaphragm muscle fibers, as well as contractile properties, were determined. OI mice were

Tenascin-C promotes migration of hepatic stellate cells and production of type I collagen.

Tenascin-C (TN-C) is an extracellular matrix glycoprotein markedly upregulated during liver fibrosis. The study is performed to explore the role of TN-C during the growth and activation of hepatic stellate cells (HSCs). We found that TN-C was accumulated accompanying with the HSC activation. Our data on cell migration assay revealed that the rTN-C treatment enhanced HSC migration in a dose- and time-dependent manner, but did not influence their proliferation. HSCs transfected with pTARGET-TN-C overexpression vector displayed increased the type I collagen (Col I) production. TN-C overexpression

Increased type I and V collagen expression in uterine leiomyomas during the menstrual cycle.

Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis showed that the ratios of type I and type V collagen expression were significantly increased in the leiomyoma tissues at the protein level, as compared with those in the normal myometrium tissues through the menstrual cycle. These results suggest that increased expression of type I and type V collagen might play a role in the pathogenesis of uterine leiomyoma.

Growth factor-induced angiogenesis in vivo requires specific cleavage of fibrillar type I collagen.

The contribution of specific type I collagen remodeling in angiogenesis was studied in vivo using a quantitative chick embryo assay that measures new blood vessel growth into well-defined fibrillar collagen implants. In response to a combination of basic fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF), a strong angiogenic response was observed, coincident with invasion into the collagen implants of activated fibroblasts, monocytes, heterophils, and endothelial cells. The angiogenic effect was highly dependent on matrix metalloproteinase (MMP) activity, because new

Marfan syndrome: absence of type I or III collagen structural defects in 25 patients.

The structure and metabolism of type I and III collagens were studied in fibroblast cultures and dermis from 25 unrelated patients including 23 with typical Marfan syndrome and two infants with a very severe clinical form of this syndrome. Electrophoretic analysis of collagen alpha-chains, as well as one- and two-dimensional electrophoresis of collagen cyanogen bromide peptides, failed to show any evidence of primary structure defects or overmodification of lysine residues in these collagens. The proportion of hydroxylated prolyl residues in isolated alpha 1(I) chains was also normal. There wa

Abnormal collagen fibril structure in the gravis form (type I) of Ehlers-Danlos syndrome.

The Ehlers-Danlos syndrome (EDS) is a clinically, genetically, and biochemically heterogeneous group of disorders characterized by generalized connective tissue abnormalities. We studied collagen fibrils by electron microscopy in the dermis of nine individuals with clinical findings consistent with type I Ehlers-Danlos syndrome. In all patients the collagen fibrils had an increased mean diameter (13 to 40%) and showed a higher degree of variability in width and shape than collagen fibrils from controls. Approximately 5% of the fibrils were much wider (up to 500 nm. versus 90 nm.), had a highly

A Gly238Ser substitution in the alpha 2 chain of type I collagen results in osteogenesis imperfecta type III.

In general, osteogenesis imperfecta (brittle bone disease) is caused by heterozygous mutations in the genes encoding the alpha 1 or alpha 2 chains of type I collagen (COL1A1 and COL1A2, respectively). In this study we screened these genes in a proband presenting with the severe form (type III) of osteogenesis imperfecta for mutations which might result in the phenotype. Single-strand conformation polymorphism mapping analysis was used to identify a region suspected of harbouring the mutation and subsequent sequence analysis revealed a heterozygous G to A transition in the alpha 2(I) gene of ty

Deposition and selective degradation of structurally-abnormal type I collagen in a collagen matrix produced by osteogenesis imperfecta fibroblasts in vitro.

Collagen matrix deposition and turnover were studied in skin fibroblasts from a control and from a patient with lethal perinatal osteogenesis imperfecta (OI) identified as a Gly667 to Arg substitution in the alpha 1(I) chain. A culture system where ascorbic acid was included to stimulate collagen matrix formation over extended culture periods was used. Serial extraction of the control cell collagen matrix confirmed that a substantial mature crosslinked collagen matrix was formed in the control fibroblast cell layer. In contrast, total collagen deposition by the OI fibroblasts was poor, with th

Thermal stability and folding of the collagen triple helix and the effects of mutations in osteogenesis imperfecta on the triple helix of type I collagen.

Osteogenesis imperfecta (OI) is an inherited disease in which 90% of the cases result from mutations in the 2 genes, pro alpha 1 and pro alpha 2, coding for type I collagen. Type I collagen is a trimeric molecule, (alpha 1)2 alpha 2, which is dominated both structurally and functionally by the 300 nm triple-helical domain. Most OI mutations occur in this domain and almost all point mutations result in the substitution of other amino acids for the obligate glycine which occurs at every third residue. The phenotypic effects of these mutations are frequently attributed in part to alterations in t
