We present a study of fibrinogen levels in 133 patients who were prone to develop disseminated intravascular coagulation as a result of an underlying complication of pregnancy such as abruptio placentae, pregnancy-induced hypertension, missed abortion, septic abortion, intrauterine fetal death, vesicular mole and amniotic fluid embolism. Dentine permeability measurements were made each hour for 6 h in tooth cavities prepared in control dogs or dogs which had been pretreated with a purified fraction of snake venom to deplete their plasma of fibrinogen. The control dogs exhibited a progressive fall in dentine permeability (approximately 50 per cent reduction in 6 h); the permeability in the fibrinogen-depleted dogs only fell 15 per cent in 6 h. The results implicate plasma proteins, particularly high molecular-weight proteins such as fibrinogen, as agents which reduce dentine permeability in vivo.

Mean plasma fibrinogen levels were determined in 133 normal calves, bulls, non-pregnant and pregnant cows. These were 508, 505, 660, and 581 mg per 100 ml of plasma respectively. The levels in 233 sick cows were often greatly increased. This appeared to be related to inflammation and tissue destruction. Lower than normal levels were sometimes seen in liver disease and terminal states.

BACKGROUND: Congenital fibrinogen deficiency is a rare inherited coagulation disorder characterized by partial or total lack of plasma fibrinogen (hypo- or afibrinogenemia), or expression of dysfunctional fibrinogen (dysfibrinogenemia), which may cause plasma level reduction (hypodysfibrinogenemia).
