Extraction/Leaching of Metal-Containing Additives from Polyvinyl Chloride, Ethyl Vinyl Acetate, and Polypropylene Bags and Infusion Sets into Infusion Solutions.

Flexible medical devices are primarily made of plasticized polyvinyl chloride (PVC). In recent times, to avoid undesired migration of the PVC plasticizers, ethyl vinyl acetate (EVA) and polypropylene (PP) has replaced PVC. Nevertheless, other additives are necessary to generate useful polymeric materials. Metallic species present in such additives can also leach out into the infusion solutions. The migration of barium (Ba), cadmium (Cd), lead (Pb), tin (Sn), and zinc (Zn) from devices made from PVC, EVA, and PP was evaluated. Bags and infusion sets were decomposed and their metallic contents a…

Stability of the inner polyaniline solid contact layer in all-solid-state K+-selective electrodes based on plasticized poly(vinyl chloride).

A simple and powerful method based on UV-visible spectroscopy is presented for studying the stability of the inner electrically conducting polyaniline (PANI) solid contact (SC) layer in all-solid-state ion-selective electrodes (ISE). The influence of the plasticized poly(vinyl chloride) (PVC) membrane (ISM) composition and the pH of the sample solution on the stability of the solid contact is reported. PANI is used as a model compound in this study, but the method presented is universal and can be applied to different types and combinations of SCs and ISMs. It provides a tool for finding the b…

Mutation analysis of K-ras-2 in liver angiosarcoma and adjacent nonneoplastic liver tissue from patients occupationally exposed to vinyl chloride.

Vinyl chloride (VC) is a potent liver carcinogen that induces angiosarcomas in humans and animals. Recent evidence shows that liver tumors from patients with VC exposure may have a specific K-ras mutation pattern. This study was performed to determine the status of K-ras-2 in liver angiosarcomas (LAS) from workers occupationally exposed to VC. We examined the presence of K-ras-2 mutations in 15 LAS from patients with known exposure to VC (median exposure: 8,260 ppm [range 3,900- 21,000 ppm]]. In all cases, other risk factors for the development of LAS were excluded. Direct DNA sequencing after…

Evidence for the mutagenic potential of the vinyl chloride induced adduct, N2, 3-etheno-deoxyguanosine, using a site-directed kinetic assay.

N2,3-Ethenoguanine (epsilon G) is a product of vinyl chloride reaction with DNA in vivo and of its ultimate metabolite, chloroacetaldehyde, in vitro. The synthesis of the very labile 5'-triphosphate of N2,3-etheno-deoxyguanosine (epsilon dGuo) has made it possible to study the base pairing properties of this derivative placed opposite a defined normal base in a 25-base oligonucleotide template. The kinetic parameters, Km and Vmax were determined from elongation of a [32P]5'-end labeled primer annealed one base prior to the designated template base, epsilon G.T pairs, which would be mutagenic, …

Assessment of a groundwater contamination with vinyl chloride (VC) and precursor volatile organic compounds (VOC) by use of a geographical information system (GIS).

Regarding the health effects of volatile organic compounds (VOC) and their decomposition products (particularly vinyl chloride (VC)) under chronic low-dose exposure, VOC groundwater contaminations are seen to be an ongoing public health issue. This article presents results of a long-term investigation surveying VOC and VC groundwater contamination upstream of a large groundwater works in Cologne, Germany. For 10 years a contaminated aquifer has been monitored for different VOC and for VC. In total, 255 samples have been taken to assess both the 3-dimensional distribution and the temporal dynam…

HPRT mutant frequencies and detection of large deletions by multiplex-PCR in human lymphocytes of vinyl chloride exposed and non-exposed populations.

The frequency of hypoxanthine guanine phosphoribosyltransferase (HPRT) mutations was determined in human T-lymphocytes isolated from peripheral blood of three populations: (1) 24 employees occupationally exposed to vinyl chloride monomer; (2) 23 healthy non-exposed matched control individuals and (3) 41 regular blood donors. In addition, mutant clones of all studied groups were examined by multiplex-PCR for visible abnormalities of the gene (large deletions). Surprisingly, the mutation frequency of all three investigated populations was approximately the same (7-8 x 10(-6)). However, great dif…

Co-immobilization of Ciprofloxacin and Chlorhexidine as a Broad-Spectrum Antimicrobial Dual-Drug Coating for Poly(vinyl chloride) (PVC)-Based Endotracheal Tubes.

The endotracheal tube (ETT) affords support for intubated patients, but the increasing incidence of ventilator-associated pneumonia (VAP) is jeopardizing its application. ETT surfaces promote (poly)microbial colonization and biofilm formation, with a heavy burden for VAP. Devising safe, broad-spectrum antimicrobial materials to tackle the ETT bioburden is needful. Herein, we immobilized ciprofloxacin (CIP) and/or chlorhexidine (CHX), through polydopamine (pDA)-based functionalization, onto poly(vinyl chloride) (PVC) surfaces. These surfaces were characterized regarding physicochemical properti…

Association between methylation of DNA damage response-related genes and DNA damage in hepatocytes of rats following subchronic exposure to vinyl chloride.

In the present study, we investigated the association between methylation of DNA damage response-related genes such as cyclin-dependent kinase inhibitor (CDKN)2A, Ras association (RalGDS/AF-6) domain family member (RASSF)1A, O6-methylguanine DNA methyltransferase (MGMT), Kirsten rat sarcoma viral oncogene homolog (KRAS), and spleen-associated tyrosine kinase (SYK) and DNA damage in hepatocytes of rats following subchronic exposure to vinyl chloride (VC). Sixty-four healthy rats were randomly divided into three VC exposure groups (5, 25, and 125 mg/kg) and an untreated negative control group (n…

A new LC-MS/MS method for the quantification of endogenous and vinyl chloride-induced 7-(2-Oxoethyl)guanine in sprague-dawley rats.

Vinyl chloride (VC) is an industrial chemical that is known to be carcinogenic to animals and humans. VC primarily induces hepatic angiosarcomas following high exposures (≥50 ppm). VC is also found in Superfund sites at ppb concentrations as a result of microbial metabolism of trichloroethylene and perchloroethylene. Here, we report a new sensitive LC-MS/MS method to analyze the major DNA adduct formed by VC, 7-(2-oxoethylguanine) (7-OEG). We used this method to analyze tissue DNA from both adult and weanling rats exposed to 1100 ppm [(13)C(2)]-VC for 5 days. After neutral thermal hydrolysis, …

Effects of vinyl chloride exposure alone and in combination with trypan blue--applied systematically during all thirds of pregnancy on the fetuses of CFY rats.

Vinyl chloride (VC) has been shown to be present in the fetal and maternal blood as well as in the amniotic fluid after the exposition of pregnant CFY rats to VC at an atmospheric concentration of 5500, 18 000 or 33 000 mg/m3 (approximately 2000, 7000 or 12 000 ppm) for 2.5 h on the 18th day of pregnancy, indicating the permeability of the placenta to the agent. Teratological investigation of the offspring of pregnant rats exposed continuously to VC at an atmospheric concentration of 4000 mg/m3 air (1500 ppm) during the first, second or last third of pregnancy has shown that VC has no teratolo…

Analysis of plasticizers in poly(vinyl chloride) medical devices for infusion and artificial nutrition: comparison and optimization of the extraction procedures, a pre-migration test step.

Medical devices (MDs) for infusion and enteral and parenteral nutrition are essentially made of plasticized polyvinyl chloride (PVC). The first step in assessing patient exposure to these plasticizers, as well as ensuring that the MDs are free from di(2-ethylhexyl) phthalate (DEHP), consists of identifying and quantifying the plasticizers present and, consequently, determining which ones are likely to migrate into the patient's body. We compared three different extraction methods using 0.1 g of plasticized PVC: Soxhlet extraction in diethyl ether and ethyl acetate, polymer dissolution, and roo…

A strategy to prepare internally-plasticized poly(vinyl chloride) by grafting castor oil onto the PVC chain with three different isocyanates as intermediate bridges.

In this work, three types of internally-plasticized poly(vinyl chloride) materials (PVC-H-C, PVC-TH-C, PVC-IP-C) were prepared by grafting castor oil onto the PVC chain with three different isocyanates as intermediate bridges, respectively. The three different isocyanates were hexamethylene diisocyanate (HDI), trimer of HDI (THDI), and isophorone diisocyanate (IPDI). This method does not need any castor oil pretreatment. The effects of different isocyanates on the plasticizing ability of the internally-plasticized PVC and the thermal stability of the whole material were discussed. The grafting…

Evaluation of thiodiglycolic acid as a urinary biomarker for exposure to vinyl chloride monomer: Development and analytical validation of a colorimetric method based on metal-organic frameworks.

Vinyl Chloride Monomer (VCM) is an industrial contaminant and carcinogen classified in IARC group 1. Chronic exposure to VCM may lead to serious health problems. Thiodiglycolic acid (TDGA) acts as a biomarker for exposure to VCM; hence, its quantitative estimation in urine provides precise information about exposure level. In this study, the authors propose a newly developed colorimetric assay based on metal-organic framework (MOFs), specifically MIL-100(Fe), acting as a nanozyme. In particular, it shows strong peroxidase-like activity; as such, it generates blue color in response to hydrogen …

The use of multizone models to estimate an airborne chemical contaminant generation and decay profile: occupational exposures of hairdressers to vinyl chloride in hairspray during the 1960s and 1970s.

Vinyl chloride (VC) was used as a propellant in a limited percentage of aerosol hairspray products in the United States from approximately 1967 to 1973. The question has arisen whether occupational exposures of hairdressers to VC-containing hairsprays in hair salons were sufficient to increase the risk for developing hepatic angiosarcoma (HAS). Transient two-zone and steady-state three-zone models were used to estimate the historical airborne concentration of VC for individual hairdressers using hairspray as well as estimated contributions from other hairdressers in the same salon. Concentrati…

Coronary artery disease and cancer mortality in a cohort of workers exposed to vinyl chloride, carbon disulfide, rotating shift work, and o-toluidine at a chemical manufacturing plant.

BACKGROUND: We updated through 2007 the mortality experience of 1,874 workers employed at a New York State chemical manufacturing plant between 1946 and 2006. METHODS: Reassessed exposures to vinyl chloride, carbon disulfide, and shift work and categories of o-toluidine exposure were based on year, department and job title. Standardized mortality ratios (SMR) compared mortality to that of the US population. Internal comparisons used directly standardized rate ratios. RESULTS: Hepatobiliary cancer mortality was elevated among workers ever exposed to vinyl chloride (SMR = 3.80, 95% confidence …

Enhanced removal of cis-1,2-dichloroethene and vinyl chloride in groundwater using ball-milled sulfur- and biochar-modified zero-valent iron: From the laboratory to the field.

Sulfidated zero-valent iron (ZVI) and biochar-supported ZVI have received increasing attention for their potential to dechlorinate trichloroethylene. However, minimal data are available regarding the combined effect of sulfur and biochar ZVI on trichloroethylene byproducts. The primary aim of the current study is to determine whether sulfur- and biochar-modified ZVI (ZVI-BC-S) enhances the removal of cis-1,2-dichloroethene (cDCE) and vinyl chloride (VC) from groundwater. Results show that biochar and sulfur facilitated the milling of ZVI-BC-S into micro- and nanoscale particles and increased F…

1,2-Dichloroethane Exposure Alters the Population Structure, Metabolism, and Kinetics of a Trichloroethene-Dechlorinating Dehalococcoides mccartyi Consortium.

Bioremediation of groundwater contaminated with chlorinated aliphatic hydrocarbons such as perchloroethene and trichloroethene can result in the accumulation of the undesirable intermediate vinyl chloride. Such accumulation can either be due to the absence of specific vinyl chloride respiring Dehalococcoides mccartyi or to the inhibition of such strains by the metabolism of other microorganisms. The fitness of vinyl chloride respiring Dehalococcoides mccartyi subpopulations is particularly uncertain in the presence of chloroethene/chloroethane cocontaminant mixtures, which are commonly found i…

[Acro-osteolysis (acro-osteopathia ulcero-mutilans) in a worker exposed to vapours of synthetics (author's transl)].

The neuro-cutaneo-osseous syndrome of "sporadic" acrosteolysis (of Bureau-Barrière), localized to the ends of the feet, has occasionally been seen in workers engaged in the polymerization of vinyl chloride, A case of the disease is described in a 42-year-old worker exposed to different vapours during the manufacture of plastic products, but he had never worked with vinyl chloride. Outstanding signs were dysproteinaemia, moderate thrombocytopenia, while symptoms of Raynaud's disease as well as scleroderma-like changes in hands and feet (typical of vinyl chloride disease) were absent. The onset …
