The role of efferocytosis in ischemic stroke and insights from retinopathy.

Efferocytosis, the phagocytic removal of dead cells by microglia and macrophages, was recently recognized as a critical process in the resolution of inflammation and tissue repair following ischemic stroke. In this review, we examine the pathways involved in efferocytosis after stroke, including the recognition and engulfment of apoptotic cells and the modulation of inflammatory responses. We elaborate on the pathways regulating efferocytosis and discuss how dysregulated efferocytosis can contribute to secondary injury and neurological deficits. Furthermore, we explore valuable mechanisms glea

Endocytic Pathways Unveil the Role of Syndecans in the Seeding and Spreading of Pathological Protein Aggregates: Insights into Neurodegenerative Disorders.

Alzheimer's disease and other neurodegenerative disorders are characterized by the accumulation of misfolded proteins, such as amyloid-beta, tau, and α-synuclein, which disrupt neuronal function and contribute to cognitive decline. Heparan sulfate proteoglycans, particularly syndecans, play a pivotal role in the seeding, aggregation, and spreading of toxic protein aggregates through endocytic pathways. Among these, syndecan-3 is particularly critical in regulating the internalization of misfolded proteins, facilitating their propagation in a prion-like manner. This review examines the mechanis

Membrane-associated glycoconjugates in parasitic protozoa: The central role of lipophosphoglycan in Leishmania.

A key event in Leishmania pathogenesis is the interaction between parasite surface molecules and host immune cells. Among these, lipophosphoglycan (LPG), the dominant surface glycoconjugate of Leishmania promastigotes, plays a central role in establishing infection and evading immune responses. Beyond mediating adhesion and protection, LPG actively modulates host membrane receptors and intracellular signaling pathways. This chapter explores the multifaceted strategies by which LPG alters host cell membrane organization to promote parasite survival. By engaging various surface receptors, LPG fa

Oxidative Stress in Cataract Formation: Is There a Treatment Approach on the Horizon?

Cataracts, a leading cause of blindness worldwide, are closely linked to oxidative stress-induced damage to lens epithelial cells (LECs). Key factors contributing to cataract formation include aging, arterial hypertension, and diabetes mellitus. Given the high global prevalence of cataracts, the burden of cataract-related visual impairment is substantial, highlighting the need for pharmacological strategies to supplement surgical interventions. Understanding the molecular pathways involved in oxidative stress during cataract development may offer valuable insights for designing novel therapeut

The signaling pathways involved in non-coding RNA regulation during osteogenic differentiation of periodontal tissue-derived cells in the field of periodontitis.

Periodontitis is a prevalent oral disease caused by chronic inflammation of the periodontal tissues surrounding the teeth, which can lead to bone loss, tooth loosening, and even tooth loss. This inflammation has a negative impact on the osteogenic differentiation capacity of periodontal tissue-derived cells. Non-coding RNAs (ncRNAs) are a class of RNA molecules that do not encode proteins but can regulate various physiological processes. In this review, we summarized the critical signaling pathways that ncRNAs modulate in osteogenic differentiation of periodontal tissue-derived cells, such as

Persulfides, at the crossroads between hydrogen sulfide and thiols.

Persulfides (RSSH/RSS-) can be formed in protein and non-protein thiols (RSH) through several different pathways, some of which are dependent on hydrogen sulfide (H2S/HS-). In addition to their roles in biosynthetic processes, persulfides are possible transducers of physiological effects of H2S through the modification of critical cysteines. Persulfides have a very rich biological chemistry that is currently under investigation. They are more nucleophilic and acidic than thiols and, unlike thiols, they can also be electrophilic. They are especially good one-electron reductants. Methods to dete

Artemisinin Bioactivity and Resistance in Malaria Parasites.

Artemisinin is the most widely-used compound against malaria and plays a critical role in the treatment of malaria worldwide. Resistance to artemisinin emerged about a decade ago in Southeast Asia and it is paramount to prevent its spread or emergence in Africa. Artemisinin has a complex mode of action and can cause widespread injury to many components of the parasite. In this review, we outline the different metabolic pathways affected by artemisinin, including the unfolded protein response, protein polyubiquitination, proteasome, phosphatidylinositol-3-kinase, and the eukaryotic translation

Hemophagocytic lymphohistiocytosis and other hemophagocytic disorders.

Hemophagocytic disorders result when critical regulatory pathways responsible for the natural termination of immune/inflammatory responses are disrupted or overwhelmed. Hemophagocytic disorders reflect pathologic defects that alter the normal crosstalk between innate and adaptive immune responses, and compromise homeostatic removal of cells that are superfluous or dangerous to the organism. Although hemophagocytic disorders are considered rare, increased awareness of these conditions has led to more frequent diagnoses, more rapid initiation of life-saving treatments, and new insights into the

Therapeutic role of cell signalling pathways and immunomodulatory approaches for recovery of spinal cord injury in dogs and cats.

Spinal cord injury (SCI) can lead to paralysis and other disorders in domestic animals, compromising welfare due to limited treatment techniques and their side effects. Modulating cell signalling pathways, such as the Wnt, BDNF (brain-derived neurotrophic factor)-TrkB and JAK/STAT pathways, plays a potential role in more rapid recovery of spinal cord trauma with fewer side effects. The Wnt signalling pathway plays a critical role in neuronal regeneration and reduction of neuronal apoptosis. The BDNF-TrkB pathway stimulates recovery of spinal cord trauma by modulating synaptic plasticity and re

Effective treatment of psoriasis with narrow-band UVB phototherapy is linked to suppression of the IFN and Th17 pathways.

Narrow-band ultraviolet-B (NB-UVB) phototherapy is an effective treatment for psoriasis. The molecular mechanisms underlying its efficacy are incompletely understood. To identify NB-UVB-induced molecular pathways that may account for its anti-inflammatory efficacy, gene expression profiling was performed using epidermal RNA from lesional and nonlesional skin from patients with psoriasis undergoing NB-UVB therapy. Downregulation of Th17 signaling pathway was observed during NB-UVB therapy in psoriatic epidermis. Strong inhibition of the Th17 pathway by UVB was confirmed in an ex vivo organ cult

Genetics of ventral forebrain development and holoprosencephaly.

The disease holoprosencephaly is the basis of the most common structural anomaly of the developing forebrain in humans. Numerous teratogens when administered during early gastrulation, have been associated with this condition. Recent studies have characterized molecules expressed in the prechordal plate which are critical for normal brain formation. Perturbation of signaling pathways involving these molecules have been shown to cause holoprosencephaly in humans and other organisms.

A Growth Mindset: Expanding the Critical Care Training Pipeline in Anesthesiology.

Critical care demand in the United States continues to rise, driven by an aging population, increasing comorbidities, and longer hospitalizations. Despite ample capacity and need for anesthesiology critical care medicine (ACCM) training, anesthesiologists account for less than 10% of U.S. intensivists, and recent fellowship match data consistently show underfilled ACCM positions. Structural factors, including perceived compensation, workload, and competing clinical demands, contribute to lower resident interest in ACCM. However, these concerns may not reflect current market and practice patter

Selenium: An Element of Life Essential for Thyroid Function.

Selenium (Se), a microelement essential for life, is critical for homeostasis of several critical functions, such as those related to immune-endocrine function and signaling transduction pathways. In particular, Se is critical for the function of the thyroid, and it is particularly abundant in this gland. Unfortunately, Se deficiency is a very common condition worldwide. Supplementation is possible, but as Se has a narrow safety level, toxic levels are close to those normally required for a correct need. Thus, whether the obtaining of optimal selenium concentration is desirable, the risk of da

TGF-β signaling in the ovary: Emerging roles in development and disease.

The TGF-β superfamily plays a pivotal role in a wide array of cellular processes, including cell proliferation, differentiation, apoptosis, and migration. It is also critically involved in ovarian development and the pathogenesis of various diseases. Within the ovary, follicles act as the primary functional units, housing numerous members of the TGF-β superfamily that regulate follicular development and, consequently, overall ovarian function. Dysregulation of the TGF-β signaling pathway is associated with reproductive disorders and the development of ovarian diseases in female mammals, such a
