[The influence of ethrane on the electro- and pentylenetetrazol-convulsions in mice (author's transl)].

The influence of Ethrane upon electro shock and pentylenetetrazol-induced convulsions was studied in mice. Similar to other inhalation anaesthetics and to barbiturates Ethrane exerts a powerful anticonvulsive activity. There is a dose response relationship between the rate of electro shock (20 mA/0.2 sec) induced convulsions and inspiratory Ethrane concentration, convulsions being completely prevented by 1.5 vol.-percent of Ethrane. The LD(50) of pentylentetrazol increases five fold under a 2 vol.-percent Ethrane anaesthesia.

Oxilorphan (l-N-cyclopropylmethyl-3,14-dihydroxymorphinan): a new synthetic narcotic antagonist.

Oxilorphan is a fully synthetic morphinan derivative containing the 14-hydroxy group characteristics of naloxone and naltrexone. As a narcotic antagonist, oxilorphan was 4 times more potent than dl-cyclazocine, equipotent to naloxone and about one-fourth as potent as naltrexone parenterally. Duration studies in rodents were inconclusive, but in antagonism of morphine analgesia and miosis in the dog, oxilorphan was longer acting than naloxone and equivalent to dl-cyclazocine. Oxilorphan was inactive in the conventional animal thermal analgesic tests. However, oxilone did exhibit relatively weak

Neural computations of threat in the aftermath of combat trauma.

By combining computational, morphological, and functional analyses, this study relates latent markers of associative threat learning to overt post-traumatic stress disorder (PTSD) symptoms in combat veterans. Using reversal learning, we found that symptomatic veterans showed greater physiological adjustment to cues that did not predict what they had expected, indicating greater sensitivity to prediction errors for negative outcomes. This exaggerated weighting of prediction errors shapes the dynamic learning rate (associability) and value of threat predictive cues. The degree to which the stria

Levetiracetam combined with ACEA, highly selective cannabinoid CB1 receptor agonist changes neurogenesis in mouse brain.

The aim of the study was to evaluate the impact of second generation antiepileptic drug levetiracetam (LEV) with arachidonyl-2'-chloroethylamide (ACEA) on proliferating neural precursor cells in mouse brain. Additionally, we established the relationship between treatment with ACEA in combination with LEV and hippocampal neurogenesis in mouse brain. All experiments were performed on male CB57/BL mice injected i.p. with LEV (10 mg/kg), ACEA (10 mg/kg) and PMSF (30 mg/kg) for 10 days. Experiments were provided in two stages: stage 1- an acute response of proliferating neural precursor cells to AC

Behavioral tagging in infant rats.

Recent studies have shown that exposure to a novel environment may stabilize the persistence of weak memories, a phenomenon often attributed to a process referred to as "behavioral tagging." While this phenomenon has been repeatedly demonstrated in adult animals, no studies to date have examined whether it occurs in infant animals, which is surprising given that infants exhibit an impaired ability to form long-term memories (LTMs). In the present study, infant (i.e., postnatal day (P) 17) rats were placed in a context and repeatedly shocked. Infant rats given brief open field exposure 1 h, but

Sex differences in the burying behavior test in middle-aged rats: effects of diazepam.

The full behavioral profile displayed during the burying behavior test was studied in middle aged (11-14 months) males, females with irregular estrous cycles, and females in persistent diestrus, with and without diazepam (0.5-2.0mg/kg). Ambulation and motor coordination were also tested to discern behavioral changes from general motor alterations. Without diazepam treatment, middle-aged males showed longer burying behavior latencies, more prod explorations and less freezing than both groups of females. Untreated middle aged males also showed less cumulative burying and more immobility compared

Glucocorticoid mechanisms may contribute to ECT-induced retrograde amnesia.

RATIONALE: Cortisol levels rise sharply immediately after electroconvulsive therapy (ECT); the resultant stimulation of steroid receptors in the hippocampus may be beneficial or harmful to cognition, depending on the magnitude of the stimulation. Steroid mechanisms may therefore modulate ECT-induced amnesia. OBJECTIVES: Using mifepristone (a glucocorticoid receptor antagonist) as a chemical probe, we sought to examine steroid mechanisms in an animal model of ECT-induced retrograde amnesia. MATERIALS AND METHODS: Adult, male Wistar rats (n = 68) trained in a step-through passive-avoidance tas

Multiple modes of action potential initiation and propagation in mitral cell primary dendrite.

The mitral cell primary dendrite plays an important role in transmitting distal olfactory nerve input from olfactory glomerulus to the soma-axon initial segment. To understand how dendritic active properties are involved in this transmission, we have combined dual soma and dendritic patch recordings with computational modeling to analyze action-potential initiation and propagation in the primary dendrite. In response to depolarizing current injection or distal olfactory nerve input, fast Na(+) action potentials were recorded along the entire length of the primary dendritic trunk. With weak-to-

Potentiation of sensory responses in the anterior cingulate cortex following digit amputation in the anaesthetised rat.

The anterior cingulate cortex (ACC) is important for processing different types of information, including sensory inputs. In the present study on anaesthetised rats, we recorded in vivo sensory responses of the ACC to peripheral electrical shocks. Peripheral electrical stimulation at high intensities sufficient to activate nociceptive sensory fibres elicited EPSPs within the ACC. Digit amputation caused long-lasting potentiation of ACC responses to peripheral electrical stimulation. Evoked field EPSPs remained enhanced for at least 120 min after the amputation. Because electrical shocks were d

Expression of antinociception in response to a signal for shock is blocked after selective downregulation of mu-opioid receptors in the rostral ventromedial medulla.

Prior work has shown that release of endogenous ligands for mu-opioid receptors in the rostral ventromedial medulla (RVM) is critical for the modulation of spinal nociceptive reflexes observed during stress. In the present study, we used antisense oligodeoxynucleotides (AS ODN) to suppress synthesis of mu-opioid receptors in the RVM prior to activating descending antinociceptive systems with a signal for foot shock. Five groups of rats with RVM cannulae were trained with paired or unpaired exposures to white noise (WN) and foot shock. Over several days, they received RVM infusions of an AS ODN

Septal vasopressin modulates motility and passive avoidance in pinealectomized rats.

Experiments were performed to investigate the role of central arginine vasopressin (AVP) in an interrelationship with the pineal gland on motility and passive avoidance response in rats. The involvement of the pineal gland in behavioral paradigms was examined using pinealectomized (PE) and pineal-intact (sham-operated and nonoperated) animals. Central administration of 200 pg AVP or 40 ng of the AVP receptor antagonist, d(CH2)sThyr(Et)VAVP (AAVP) was performed into the mediolateral septum by means of microdialysis probes. The blockade of vasopressinergic neurotransmission or neuromodulation in

Changes in brain monoamine metabolism and carbon dioxide induced amnesia in the rat.

The effect of treatment with carbon dioxide (CO2) on the performance of rats 24 hr after receiving a foot shock in a passive avoidance task was studied. Foot shock induced avoidance. Carbon dioxide produced retrograde amnesia for the foot shock induced avoidance response. Changes in brain monoamine metabolism were studied in groups of rats which had been treated with CO2, foot shock or foot shock +CO2. The rats were killed 24 hr after treatment. Changes mainly occurred in the brain stem and hippocampus. In the rats which had received foot shock alone, brain stem and hippocampal serotonin conce

Diaphragmatic pacing for the prevention of sudden unexpected death in epilepsy.

Sudden unexpected death in epilepsy is the leading cause of epilepsy related death. Currently, there are no reliable methods for preventing … be prevented with diaphragmatic pacing. Our approach was to induce respiratory arrest using maximal electroshock seizures in adult, male, C57BL6 mice outfitted with EEG and diaphragmatic electrodes (n = 8 mice). In the experimental group, the diaphragm was stimulated to exogenously induce breathing. In the control group, no stimulation was applied. Breathing a …

Pharmacokinetic and pharmacodynamic analysis of (2S,3S)-sec-butylpropylacetamide (SPD) in rats and pigs-A CNS-active stereoisomer of SPD.

OBJECTIVES: To advance the development of (2S,3S)-sec-butylpropylacetamide (SPD) as a new treatment for acute repetitive seizures (ARS), by studying its pharmacokinetics (PK) in pigs and its PK-pharmacodynamic (PK-PD) correlation in rats. METHODS: Two (2S,3S)-SPD intramuscular formulations (FA and FB ) were administered to pigs and rats and blood samples were withdrawn at different times after dosing. Major PK parameters were estimated in both species. PD analysis was conducted in rats utilizing the maximal-electroshock seizure threshold (MEST) test. Because ARS treatment requires a rapid act…

Electrical stimulation therapies for CNS disorders and pain are mediated by competition between different neuronal networks in the brain.

CNS neuronal networks are known to control normal physiological functions, including locomotion and respiration. Neuronal networks also mediate the pathophysiology of many CNS disorders. Stimulation therapies, including localized brain and vagus nerve stimulation, electroshock, and acupuncture, are proposed to activate "therapeutic" neuronal networks. These therapeutic networks are dormant prior to stimulatory treatments, but when the dormant networks are activated they compete with pathophysiological neuronal networks, disrupting their function. This competition diminishes the disease symptom…
