Naloxone reversal of ischemic arrhythmia is stereospecific and suggests role of endogenous opioid peptides in ischemic heart disease.

The effects of the stereoisomers of naloxone during myocardial ischemia were studied. (-)-Naloxone (but not the (+)-isomer naloxone) attenuated the ischemia-induced cardiac arrhythmias, hypotension, and bradycardia that result from coronary artery occlusion in anesthetized rats. From these findings, it may be inferred that endogenous opioid peptides may play a role in the pathophysiology of myocardial ischemia. It is also suggested that naloxone may have therapeutic value in the prevention and treatment of ischemic heart disease.

Aerobic fitness and opioidergic inhibition of cardiovascular stress reactivity.

The role of endogenous opioids in aerobic fitness-induced decrements in cardiovascular stress reactivity was examined by comparing the effects of opioid antagonism with naltrexone on responses to stress in young adults with high versus low levels of aerobic fitness. Two hundred forty subjects were given an activity questionnaire and males with the highest (Fit) and lowest (Nonfit) aerobic activity profiles were recruited for maximal oxygen consumption (VO2max) treadmill testing and psychological stress testing (final sample N = 28). Heart rate and blood pressures were measured during performan

Physiologic and plasma hormone correlates of survival in endotoxic dogs: effects of opiate antagonists.

Two groups of awake dogs were given an iv bolus of endotoxin (3.0 mg/kg from Salmonella typhimurium); one group (n = 9) was pretreated with either naloxone (2.0 mg/kg iv bolus with 1.7 mg/kg.h; n = 6) or naltrexone (2.0 mg/kg iv bolus with repeat bolus of 1.0 mg/kg at 1, 3 and 5 h; n = 3) and the second group (n = 10) received no opiate antagonist. All of nine dogs that were pretreated with an antagonist survived for 24 h, compared to only five of ten dogs that were not pretreated. Survival correlated with improved BP (mean of 91 vs. 61 mm Hg) and cardiac output (3.9 vs. 2.45 L/min) measured d

Effects of naloxone on vasopressin secretion in conscious rats: evidence for inhibitory role of endogenous opioid peptides in vasopressin secretion.

The effects of naloxone, an opioid antagonist, on arginine vasopressin (AVP) secretion were examined in conscious unrestrained rats under both basal and stimulated conditions. Intravenous injection of naloxone in a dose of 0.1 mg/kg did not significantly affect the basal plasma AVP level. However, 0.5 or 2.5 mg/kg naloxone significantly raised the basal AVP level in euhydrated rats. Naloxone (0.5 mg/kg) significantly enhanced AVP secretion after 72-h water deprivation. However, the enhancement was more prominent in euhydrated rats than in dehydrated rats. Pretreatment with naloxone (0.5 mg/kg)

Polycystic ovarian disease: animal models.

The reproductive systems of human beings and other vertebrates are grossly similar. In the ovary particularly, the biochemical and physiologic processes are identical not only in the formation of germ cells, the development of primordial follicles and their subsequent growth to Graafian follicles, and eventual ovulation but also in anatomic structure. In a noncarcinogenic human ovary, hypersecretion of androgen causes PCOD. Such hypersecretion may result from a nonpulsatile, constant elevated level of circulating LH or a disturbance in the action of neurotransmitters in the hypothalamus. In st

Motor neuron excitability: the Hoffmann reflex following exercise of low and high intensity.

The current exercise literature lacks physiological evidence for a stress reduction induced by high intensity aerobic work. The purpose of this study was to investigate the effectiveness of high intensity aerobic work on stress reduction and to evaluate the currently tenable hypotheses in view of the data on the response to high intensity work. The subjects participated in a control trial and two 20-min treadmill exercise trials at low and high (40% and 75% VO2max) intensity. The H- and M-wave responses to artificial stimulation of the tibial nerve (Angel and Hofmann, 1963) were used to assess

Opiate-induced seizures: a study of mu and delta specific mechanisms.

Two groups of experiments were conducted to determine if morphine- and enkephalin-induced seizures are specifically mediated by the mu and delta receptor, respectively. In the first experiments, designed to assess the ontogeny of mu- or delta-seizures, rats from 6 h to 85 days of age received implanted cortical and depth electrodes as well as an indwelling cannula in the lateral ventricle. Various amounts of the mu-receptor agonists, morphine and morphiceptin, and the delta agonists, D-Ala2-D-Leu5-enkephalin (DADL) and Tyr-D-Ser-Gly-Phe-Leu-Thr (DSLET), were then administered intracerebroventr

The pulmonary effects of opiate blockade in septic shock.

Sepsis remains the most common associated factor in acute respiratory failure (ARF). Endogenous opiates are known to have both respiratory and cardiovascular depressant effects. Because there is a high level of circulating endogenous opiates in sepsis, the possible role of opioids in the ARF syndrome seen in sepsis was studied. Sixteen piglets were infused with an LD100 dose (7.5 X 10(10) organisms/kg) of live Escherichia coli (Type 09-41). The pigs were hemodynamically monitored. Serial blood samples were taken for arterial blood gases and lactate. Serial lung biopsies were taken for determin

Brain regulation of gastric acid secretion in rats by neurogastrointestinal peptides.

Brain alteration of catecholaminergic, serotoninergic, dopaminergic, gabaergic and cholinergic pathways by intracisternal injection of agonists, antagonists or specific neurotoxic drugs did not significantly affect gastric acid secretion in 2 hr pylorus-ligated rats. In contrast, several neuropeptides were found to be very potent in influencing gastric secretion when administered intracisternally but not when given intravenously. Bombesin-like peptides, opioid peptides and arginine vasopressin acted within the brain to inhibit gastric acid secretion through yet unknown neurohumoral pathways, w

The hypothalamus: anatomy and functions.

The neuroanatomy of the human hypothalamus is reviewed with special interest focused on its neuroendocrine role. The magnocellular neurons in the supraoptic and paraventricular nuclei are the site of synthesis of the nonapeptides antidiuretic hormone and oxytocin and their carriers, the neurophysins. They are in close relation with the posterior lobe of the pituitary which contains their axonal neurosecretory endings. The parvocellular neurons are scattered around the third ventricle, from the preoptic area towards the infundibulum. They control the adenohypophysis by the releasing hormones fo

Enkephalin effects on blood pressure, heart rate, and baroreceptor reflex.

The cardiovascular effects of opioid peptides have been studied. Leucine-enkephalin (Leu-ENK) produced blood pressure (BP) increases following administration into the lateral brain ventricles (i.v.t.), into the cisterna magna (i.c.i.), and following intravenous (i.v.) administration. Heart rate (HR) increases were observed following all routes of administration (threshold for BP and HR effects at 0.3 nmole, maximum at 360 nmoles). The cardiovascular effects were independent of generalized seizures, which may occur at higher doses of enkephalins (ENK). D-alanine-enkephalin (D-Ala-ENK) attenuate

Physiotherapy approaches for pain control in patients who are critically ill.

Pain is a significant challenge in critical care settings, affecting patient outcomes, recovery time, and quality of life. While pharmacological interventions remain the cornerstone of pain management in intensive care units (ICUs), they are associated with numerous adverse effects including respiratory depression, delirium, and prolonged ICU stays. To examine evidence-based physiotherapy approaches that can effectively complement traditional pain management strategies in patients who are critically ill. We conducted a comprehensive literature review of physiotherapy modalities used for pain c

The role of the endogenous opioid system in modulating orthodontic-induced neurogenic inflammation of the dental pulp: A comprehensive review of the literature.

BACKGROUND: Orthodontic forces may lead to neurogenic inflammation in the dental pulp by triggering the release of somatosensory neuropeptides such as Substance P (SP), Calcitonin gene-related peptide (CGRP) and Neurokinin A (NKA). In the vast majority of patients, acute symptoms are not triggered, probably due to the control of the neurogenic inflammatory process exerted by endogenous opioid systems. OBJECTIVE: This review aimed to assess the cellular and molecular mechanisms through which the endogenous opioid system modulates the orthodontic-induced neurogenic inflammation of the dental pu

Dynamic analysis of protracted withdrawal symptoms, neurotransmitters, cytokine content and psychological status of heroin addicts in Yunnan Province.

BACKGROUND: Yunnan Province, adjacent to the "Golden Triangle," a notorious drug trafficking region, faces a severe public health challenge due to high rates of heroin addiction. Despite existing interventions, protracted withdrawal symptoms (PWS) remain a significant barrier to recovery and a primary cause of relapse. This study investigates the dynamic changes in neurotransmitter and cytokine levels and their correlation with PWS and psychological status in heroin addicts undergoing detoxification. METHODS: A prospective cohort study was conducted with 136 voluntary heroin detoxification pa

Mechanisms of action of ethyl acetate fractions of Liparis nervosa (Thunb.) Lindl. as potential central anti-nociceptive agents.

Opioids/non-steroidal anti-inflammatory drugs are used to alleviate pain; however, they are expensive and can have adverse effects, especially when used over extended periods. Therefore, there is immense demand for innovative, non-addictive analgesics. Here, we report a novel plant-derived central anti-nociceptive agent, Liparis nervosa (Thunb.) Lindl. (LN), validated in animal pain models. Ethyl acetate fractions of L. nervosa (EALN) exhibited central anti-nociceptive activity in hot plate, tail immersion, formalin-induced paw oedema, and acetic acid-induced abdominal writhing tests. The chem
