Network Abnormalities in Ischemic Stroke: A Meta-analysis of Resting-State Functional Connectivity.

Aberrant large-scale resting-state functional connectivity (rsFC) has been frequently documented in ischemic stroke. However, it remains unclear about the altered patterns of within- and across-network connectivity. The purpose of this meta-analysis was to identify the altered rsFC in patients with ischemic stroke relative to healthy controls, as well as to reveal longitudinal changes of network dysfunctions across acute, subacute, and chronic phases. A total of 24 studies were identified as eligible for inclusion in the present meta-analysis. These studies included 269 foci observed in 58 con

Reduced local functional connectivity correlates with atypical performances in children with autism spectrum disorder.

To characterize local functional connectivity (FC) differences in children with autism spectrum disorder (ASD) compared to typically developed (TD) children, and to analyze the correlation between local FC and the atypical behavior in autistic children. Thirty children with ASD and 25 TD children were recruited. Participants underwent rs-fMRI scans, and regional homogeneity (ReHo) of specific brain regions was measured. Performance was assessed using the Autism Behavior Checklist (ABC) and the Gesell Development Diagnosis Scale (GDDS). Children with ASD demonstrated reduced ReHo in the right o

Electroencephalographic Functional Connectivity Patterns in Children With Tourette Syndrome and Attention-Deficit/Hyperactivity Disorder.

BACKGROUND: Tourette syndrome (TS) and attention-deficit/hyperactivity disorder (ADHD) often co-occur and are linked to emotional and behavioral difficulties. However, their shared and distinct neural underpinnings, particularly in terms of functional connectivity, remain unclear. Here, we assessed how functional connectivity differs across TS and ADHD as well as its association with emotional and behavioral difficulties. METHODS: Resting-state electroencephalography (EEG) was recorded from 137 children with TS (n = 51), ADHD (n = 24), or TS + ADHD (n = 29) or from typically developing contro

Whole Brain Monosynaptic Input of Distinct Neurons in the Globus Pallidus Externa.

OBJECTIVE: The globus pallidus externa (GPe) is involved in mediating physiological functions and contains two types of neurons: Forkhead box protein P2-expressing (GPeFoxP2) neurons which inhibit motor, and parvalbumin-expressing (GPePV) neurons which improve motor. The functional complexity of the GPe, directly linked to its neuronal heterogeneity, necessitates exploring the neuroanatomical circuits of its distinct neuron types as a foundation for functional research. METHODS: In this study, we employed specific, modified rabies viruses and adeno-associated viruses to investigate the monosy

Detection of Threshold-Level Stimuli Modulated by Temporal Predictions of the Cerebellum.

The cerebellum has the reputation of being a primitive part of the brain that mostly is involved in motor coordination and motor control. Older lesion studies and more recent electrophysiological studies have, however, indicated that it is involved in temporal perception and temporal expectation building. An outstanding question is whether this temporal expectation building cerebellar activity has functional relevance. In this study, we collected magnetoencephalographic data from 30 healthy participants performing a detection task on at-threshold stimulation that was presented at the end of a

Decoding state-dependent cortical-cerebellar cellular functional connectivity in the mouse brain.

The cortex and cerebellum form multi-synaptic reciprocal connections. We investigate the functional connectivity between single spiking cerebellar neurons and the population activity of the mouse dorsal cortex using mesoscale imaging. Cortical representations of individual cerebellar neurons vary significantly across different brain states but are drawn from a common set of cortical networks. These cortical-cerebellar connectivity features are observed in mossy fibers and Purkinje cells as well as neurons in different cerebellar lobules, albeit with variations across cell types and regions. Probabilistic coverage of the frontal aslant tract in young adults: Insights into individual variability, lateralization, and language functions.

The frontal aslant tract (FAT) is a crucial neural pathway of language and speech, but little is known about its connectivity and segmentation differences across populations. In this study, we investigate the probabilistic coverage of the FAT in a large sample of 1065 young adults. Our primary goal was to reveal individual variability and lateralization of FAT and its structure-function correlations in language processing. The study utilized diffusion MRI data from 1065 subjects obtained from the Human Connectome Project. Automated tractography using DSI Studio software was employed to map whi

Distinct Thalamo-Subcortical Circuits Underlie Painful Behavior and Depression-Like Behavior Following Nerve Injury.

Clinically, chronic pain and depression often coexist in multiple diseases and reciprocally reinforce each other, which greatly escalates the difficulty of treatment. The neural circuit mechanism underlying the chronic pain/depression comorbidity remains unclear. The present study reports that two distinct subregions in the paraventricular thalamus (PVT) play different roles in this pathological process. In the first subregion PVT posterior (PVP), glutamatergic neurons (PVPGlu) send signals to GABAergic neurons (VLPAGGABA) in the ventrolateral periaqueductal gray (VLPAG), which mediates painfu

Peripheral and central innervation pattern of mechanosensory neurons in the trigeminal ganglion.

The trigeminal ganglion (TG) comprises primary sensory neurons responsible for orofacial sensations, subsequently projecting to the trigeminal nuclei in the brainstem. However, the circuit basis of nasal mechanosensation is not well characterized. Here we elucidate the anatomical organization of both peripheral and central projections of the TG. We found that the non-peptidergic nociceptor, MAS-related G protein-coupled receptor member D positive (MrgprD+) neurons in the TG densely innervate the nasal mucosa, whereas the low-threshold mechanoreceptors subtypes rarely innervate the nasal mucosa

Altered interhemispheric connectivity in Huntington's Disease.

Pyramidal cells give rise to the corpus callosum, interhemispheric fibers that constitute the associations between the left and the right hemispheres. These interconnections are the substrates of important neurological functions, such as perception, memory, emotion, and movement control, which are all affected in Huntington's disease (HD). In this study we used directional tract density patterns (dTDPs) to evaluate changes in interhemispheric connectivity in gene-expanded individuals, which included presymptomatic and early symptomatic HD subjects. Our results demonstrated regionally selective

Cytoarchitectonic, receptor distribution and functional connectivity analyses of the macaque frontal lobe.

Based on quantitative cyto- and receptor architectonic analyses, we identified 35 prefrontal areas, including novel subdivisions of Walker's areas 10, 9, 8B, and 46. Statistical analysis of receptor densities revealed regional differences in lateral and ventrolateral prefrontal cortex. Indeed, structural and functional organization of subdivisions encompassing areas 46 and 12 demonstrated significant differences in the interareal levels of α2 receptors. Furthermore, multivariate analysis included receptor fingerprints of previously identified 16 motor areas in the same macaque brains and revea

Structural and functional connectivity of the inferior temporal numeral area.

A growing body of evidence suggests that in adults, there is a spatially consistent "inferior temporal numeral area" (ITNA) in the occipitotemporal cortex that appears to preferentially process Arabic digits relative to non-numerical symbols and objects. However, very little is known about why the ITNA is spatially segregated from regions that process other orthographic stimuli such as letters, and why it is spatially consistent across individuals. In the present study, we used diffusion-weighted imaging and functional magnetic resonance imaging to contrast structural and functional connectivi

Characteristic cortico-cortical connection profile of human precuneus revealed by probabilistic tractography.

It is generally hypothesized that functional connectivity (FC) reflects the underlying structural connectivity (SC). The precuneus is associated with highly integrated cognitive functions. However, our understanding of the structural connections that could underlie them is limited. This study aimed to characterize the cortico-cortical connections by probabilistic tractography. The precuneus corresponds to the five cortical areas (7Am, PCV, 7Pm, 7m, POS2) on the HCP MMP atlas. We first conducted the atlas-based probabilistic tractography. The anterior part (7Am) was strongly connected to the se

Responsiveness variability during anaesthesia relates to inherent differences in brain structure and function of the frontoparietal networks.

Anaesthesia combined with functional neuroimaging provides a powerful approach for understanding the brain mechanisms of consciousness. Although propofol is used ubiquitously in clinical interventions that reversibly suppress consciousness, it shows large inter-individual variability, and the brain bases of this variability remain poorly understood. We asked whether three networks key to conscious cognition-the dorsal attention (DAN), executive control (ECN), and default mode (DMN)-underlie responsiveness variability under anaesthesia. Healthy participants (N = 17) were moderately anaesthetize

Functional connectivity between pre-supplementary motor area and inferior parietal lobule associated with impaired motor response inhibition in first-degree relatives of patients with obsessive-compulsive disorder.

Previous studies have suggested that specific fronto-striatal circuits are associated with impaired motor response inhibition in patients with obsessive-compulsive disorder (OCD) and their relatives. However, no study has investigated the underlying resting-state network associated with motor response inhibition in the unaffected first-degree relatives of patients with OCD. We measured motor response inhibition using stop-signal task, and obtained resting-state fMRI in 23 first-degree relatives and 52 healthy control participants. We explored the group differences in the functional network fro

A parabrachial-hypothalamic parallel circuit governs cold defense in mice.

Thermal homeostasis is vital for mammals and is controlled by brain neurocircuits. Yet, the neural pathways responsible for cold defense regulation are still unclear. Here, we found that a pathway from the lateral parabrachial nucleus (LPB) to the dorsomedial hypothalamus (DMH), which runs parallel to the canonical LPB to preoptic area (POA) pathway, is also crucial for cold defense. Together, these pathways make an equivalent and cumulative contribution, forming a parallel circuit. Specifically, activation of the LPB → DMH pathway induced strong cold-defense responses, including increases in …

The role of the primary sensorimotor system in generalized epilepsy: Evidence from the cerebello-cerebral functional integration.

The interaction between cerebellum and cerebrum participates widely in function from motor processing to high-level cognitive and affective processing. Because of the motor symptom, idiopathic generalized epilepsy (IGE) patients with generalized tonic-clonic seizure have been recognized to associate with motor abnormalities, but the functional interaction in the cerebello-cerebral circuit is still poorly understood. Resting-state functional magnetic resonance imaging data were collected for 101 IGE patients and 106 healthy controls. The voxel-based functional connectivity (FC) between cerebral

Alterations of default mode and cingulo-opercular salience network and frontostriatal circuit: A candidate endophenotype of obsessive-compulsive disorder.

Background It is gradually becoming clear that obsessive-compulsive disorder (OCD) patients have aberrant resting-state large-scale intrinsic networks of cingulo-opercular salience (SN), default mode (DMN), and front-parietal network (FPN). However, it remains unknown whether unaffected first-degree relatives of OCD patients have these alterations as a vulnerability marker to the disorder. Methods We performed resting-state functional magnetic resonance imaging (rsfMRI) scans of 47 medication-free OCD patients, 21 unaffected healthy first-degree relatives of OCD patients, and 62 healthy contro

Altered frontoparietal connectivity in patients with obsessive-compulsive disorder during an fMRI cognitive reappraisal task.

Patients with obsessive-compulsive disorder (OCD) present increased brain activity in orbitofrontal and limbic regions when experiencing negative emotions, which could be related to deficits in emotion regulation abilities. 30 OCD patients and 29 healthy controls (HC) performed a cognitive reappraisal functional magnetic resonance imaging (fMRI) task and completed emotion regulation and OCD symptomatology questionnaires. Besides task activation, connectivity was also compared between groups through psychophysiological interaction analysis (PPI), using regions previously reported to be hyperact

Growth hormone deficiency interferes with dynamic brain networks in short children.

PURPOSE: This study aimed to explore the disparities in dynamic brain networks between children with growth hormone deficiency (GHD) and idiopathic short stature (ISS, non-growth hormone deficiency). METHODS: This study enrolled 65 children with GHD and 60 sex- and age-matched children with ISS. Resting-state functional magnetic resonance imaging (rs-fMRI) was performed for all participants to obtain information on dynamic regional homogeneity (dReHo) and functional connectivity (FC) in dynamic (dFC) or static (sFC) state. The rs-fMRI metrics were subsequently compared between the GHD and ISS
