Diversity of specialized metabolites in Phaeoacremonium species revealed by untargeted metabolomics and bioactivity assays.

Fungi belonging to the genus Phaeoacremonium are recognized as cross-kingdom pathogens, that affect both agricultural crops and human health. Here, we report the first comprehensive large-scale chemotaxonomic study based on untargeted MS-based metabolomics of 28 isolates representing 24 Phaeoacremonium species. The chemometric analysis was integrated with in vitro toxicity on human keratinocytes and cucumber (Cucumis sativus) cotyledons, serving as representative models for human and plant hosts, respectively. A targeted LC-MS/MS method validated the production of the naphthoquinones scytalone…

The gynoecious CmWIP1 transcription factor interacts with CmbZIP48 to inhibit carpel development.

In angiosperms, sex determination leads to development of unisexual flowers. In Cucumis melo, development of unisexual male flowers results from the expression of the sex determination gene, CmWIP1, in carpel primordia. To bring new insight on the molecular mechanisms through which CmWIP1 leads to carpel abortion in male flowers, we used the yeast two-hybrid approach to look for CmWIP1-interacting proteins. We found that CmWIP1 physically interacts with an S2 bZIP transcription factor, CmbZIP48. We further determined the region mediating the interaction and showed that it involves the N-termin…

Validation of an ampicillin selection protocol to enrich for mutants of Listeria monocytogenes unable to replicate on fresh produce.

Several outbreaks of listeriosis have implicated fresh produce but genetic factors required for growth of Listeria monocytogenes on produce remain poorly characterized. Based on the fact that β-lactam antibiotics only kill bacterial cells that are growing, we hypothesized that ampicillin selection can enrich for L. monocytogenes mutants unable to grow on produce. For validation, we examined relative recovery of L. monocytogenes strain 2011L-2858 and its cold-sensitive mutant L1E4 following inoculation of cantaloupe rind fragments with 1:1 mixture of the strains and incubation at 4°C with or wi

[Effects of NO3- stress on cucumber seedling growth and magnesium absorbing under suboptimal temperature].

The effects of NO3- stress on the growth and the magnesium absorption of cucumber seedlings were investigated after 1 and 14 d of suboptimal temperature [18 °C/12 °C (day/night)] treatment. The results indicated that the growth, net photosynthetic rate, transpiration rate, Fv/Fm and ΦPSII of cucumber seedlings were significantly inhibited by NO3- stress under suboptimal temperature. The magnesium content of cucumber seedlings was also significantly decreased compared with control treatment, especially in the IV treatment (suboptimal temperature +140 mmol · L(-1) NO3- + 1 mmol · L(-1) Mg2+), an

Distribution and accumulative pattern of tetracyclines and sulfonamides in edible vegetables of cucumber, tomato, and lettuce.

Veterinary antibiotics can be released to environment by the animals' excretions, which thereby poses human health and ecological risks. Six antibiotics (tetracycline, oxytetracycline, chlortetracycline, sulfamethazine, sulfamethoxazole, and sulfadimethoxine) at three concentrations (5, 10, and 20 mg kg(-1) soil) were employed in pots filled with a loamy sand upland soil. Three types of vegetable seedlings, including cucumber (Cucumis sativus), cherry tomato (Solanum lycopersicum), and lettuce (Lactuca sativa), were also cultivated during 45 d in the greenhouse. All antibiotics taken up by tes…

Light quality shapes mesophyll conductance in cucumber leaves in association with coordinated anatomical changes.

Light quality is a pivotal environmental cue shaping leaf anatomy and photosynthetic performance, but how anatomical changes are associated with internal CO2 diffusion remains incompletely understood. Mesophyll conductance, a key determinant of photosynthetic rate, is strongly impacted by anatomical traits, yet the specific structural correlates of mesophyll conductance under varying light spectra remain incompletely understood. We investigated how varying red (R) and blue (B) light ratios affected leaf anatomical traits and how these traits were associated with changes in mesophyll conductanc

Effect of medium composition on in vitro ovary culture of cucumber.

Ovary culture is among the most effective techniques for haploid induction in horticultural crops, including cucumber. The application of doubled haploid technology can significantly accelerate cultivar development by enabling the rapid production of homozygous inbred lines. This study evaluated the effects of culture medium combinations of plant growth hormones (2,4-D, NAA, IAA, IBA, PUT, BAP, KIN, TDZ) on haploid induction in cucumber via ovary culture. Four induction and five regeneration media were tested across four cucumber varieties: Beith Alpha, PTK40, Botanik, and Sardes. The highest

A Novel Epiphytic Clonostachys chloroleuca Strain that Effectively Controls Cucumber Anthracnose.

Cucumber anthracnose is a destructive disease caused by Colletotrichum orbiculare that affects global cucumber production. Although plant-associated fungi are promising sources of biological control agents, their efficacy against cucumber anthracnose has not been established. In the present study, 108 spore-forming endophytic and epiphytic fungal strains were isolated from the leaves of seven plant species and screened for biocontrol activity against C. orbiculare using detached cucumber cotyledon assays and pot experiments. Three highly suppressive strains (GUSF-44, GUSF-66, and GUSF-153) ide

Inhibitory effects of components from root exudates of Welsh onion against root knot nematodes.

Root-knot nematodes (RKNs; Meloidogyne spp.) are obligate endoparasites that infect many crops and cause severe yield losses. In this research, we studied the effect of Welsh onion, grown as a companion plant, on the resistance of cucumber plants to RKN infection and analyzed the most abundant components of Welsh onion root exudates. The results showed that, when grown with Welsh onion as a companion plant, cucumber roots had 77.0% fewer root knots and egg masses than the control cucumber roots. Welsh onion root exudates were collected and extracted with chloroform, ethyl ether, n-butanol and

Ability of nonpathogenic Fusarium oxysporum strain Fo47 to induce resistance against Pythium ultimum infection in cucumber.

The influence exerted by nonpathogenic Fusarium oxysporum strain Fo47 in triggering cucumber protection against infection by Pythium ultimum was investigated ultrastructurally. Macroscopic and microscopic observations of the pathogen colony in dual cultures revealed that reduction of Pythium growth was associated with marked disorders, including generalized disorganization of the host cytoplasm, retraction of the plasmalemma, and complete loss of the protoplasm. Cytochemical labeling of cellulose with an exoglucanase-gold complex showed that the cellulose component of the host cell walls was s

L-phenylalanine induces cutin and wax biosynthesis in muskmelon fruit epidermis via fatty acid and triterpenoid pathways.

This study explored the effects of preharvest L-Phenylalanine (L-Phe) sprays on cuticle development and associated lipid metabolism in muskmelon (Cucumis melo L.) fruit. Spraying 8 mM L-Phe during fruit growth increased cuticle thickness and the accumulation of hydroxy fatty acids, fatty alcohols, alkanes, and triterpenoids. L-Phe enhanced the activities of key enzymes (FAT, LACS, FAR, SQE, OSC) and upregulated genes involved in fatty acid elongation, hydroxylation, alkane and triterpenoid biosynthesis. It also increased ATP-binding cassette (ABCG) transporter and lipid transfer protein (LTP) …

Survival and transcriptomic response of Salmonella enterica on fresh-cut fruits.

Salmonella enterica is frequently implicated in foodborne disease outbreaks associated with fresh-cut fruits. In the U.S., more than one third of fruit-related outbreaks have been linked to two S. enterica serotypes Newport and Typhimurium. Approximately 80% of fruit-related human salmonellosis cases were associated with tomatoes, cantaloupes and cucumbers. In this study, we investigated the population dynamics of S. Newport and S. Typhimurium on fresh-cut tomato, cantaloupe, cucumber and apple under short-term storage conditions. We further compared the transcriptomic profiles of a S. Newport

Molecular Mechanisms Underlying Cucumber Resistance to Cucumber Green Mottle Mosaic Virus: From Host-Virus Interactions to Precision Breeding Strategies.

Cucumber (Cucumis sativus L.) is an economically important vegetable crop cultivated worldwide, but its sustainable production is increasingly threatened by Cucumber green mottle mosaic virus (CGMMV). CGMMV is a positive-sense single-stranded RNA virus belonging to the genus Tobamovirus and represents a major constraint on cucurbit production because of its efficient seed transmission, mechanical spread, environmental persistence, and broad host range. Infection can cause leaf mosaic and mottling, growth suppression, and severe fruit symptoms, including internal discoloration, water-soaked tis…

Segmented light-dark cycles enhance photosynthetic efficiency and growth in cucumber but inhibit tomato growth through photosynthesis disruption.

Optimizing light environments is essential for maximizing crop productivity and resource-use efficiency in plant factories with artificial lighting. Specifically, the frequency of light-dark cycles within a 24-h period interacts with the intrinsic circadian rhythms of plants, thereby influencing carbon metabolism and photosynthetic efficiency. In this study, we examined the effects of different light-dark cycle frequencies on the growth and photosynthetic activity of cucumber and tomato seedlings. The light-dark cycle frequencies were established as follows: Cycle 1 (16/8 h), 2 (8/4 h), 4 (4/2

Analyses of 37 composts revealed microbial taxa associated with disease suppressiveness.

UNLABELLED: Compost is a valuable amendment for soil and potting substrate when it comes to suppressing soilborne pathogens. However, the effectiveness of different composts varies and cannot yet be predicted. Microbial communities in compost play a key role in disease suppression, and therefore their composition or specific taxa may serve as indicators of suppressive composts. In this study, we investigated 37 composts from 7 commercial compost producers to analyze the association of their bacterial and fungal communities with suppressive activity in three plant-pathogen systems: cress-Globis
