Using an adapted form of the picture exchange communication system to increase independent requesting in deafblind adults with learning disabilities.

The current study assessed the effectiveness of an adapted form of the Picture Exchange Communication System (PECS) in increasing independent requesting in deafblind adults with learning disabilities. PECS cards were created to accommodate individual needs, including adaptations such as enlarging photographs and using swelled images which consisted of images created on raised line drawing paper. Training included up to Phase III of PECS and procedures ensuring generalizations across individuals and contexts were included. The effects of the intervention were evaluated using a multiple baseline

Co-occurrence of A1555G and G11778A in a Chinese family with high penetrance of Leber's hereditary optic neuropathy.

Co-occurrence of double pathogenic mtDNA mutations with different claimed pathological roles in one mtDNA is infrequent. It is tentative to believe that each of these pathogenic mutations would have its own deleterious effect. Here we reported one three-generation Chinese family with a high penetrance of LHON (78.6%). Analysis of the complete mitochondrial genome in the proband revealed the presence of the LHON primary mutation G11778A in the NADH dehydrogenase 4 (ND4) gene and a deafness-associated mutation A1555G in the 12S rRNA gene. The other mtDNA variants in this family suggested a haplo

Pathogenic Variants in USH1G/SANS Alter Protein Interaction with Pre-RNA Processing Factors PRPF6 and PRPF31 of the Spliceosome.

Pre-mRNA splicing is an essential process orchestrated by the spliceosome, a dynamic complex assembled stepwise on pre-mRNA. We have previously identified that USH1G protein SANS regulates pre-mRNA splicing by mediating the intranuclear transfer of the spliceosomal U4/U6.U5 tri-snRNP complex. During this process, SANS interacts with the U4/U6 and U5 snRNP-specific proteins PRPF31 and PRPF6 and regulates splicing, which is disturbed by variants of USH1G/SANS causative for human Usher syndrome (USH), the most common form of hereditary deaf-blindness. Here, we aim to gain further insights into th

SANS (USH1G) regulates pre-mRNA splicing by mediating the intra-nuclear transfer of tri-snRNP complexes.

Splicing is catalyzed by the spliceosome, a compositionally dynamic complex assembled stepwise on pre-mRNA. We reveal links between splicing machinery components and the intrinsically disordered ciliopathy protein SANS. Pathogenic mutations in SANS/USH1G lead to Usher syndrome-the most common cause of deaf-blindness. Previously, SANS was shown to function only in the cytosol and primary cilia. Here, we have uncovered molecular links between SANS and pre-mRNA splicing catalyzed by the spliceosome in the nucleus. We show that SANS is found in Cajal bodies and nuclear speckles, where it interacts

What can naturally occurring mutations tell us about Ca(v)1.x channel function?

Voltage-gated Ca²⁺ channels allow for Ca²⁺-dependent intracellular signaling by directly mediating Ca²⁺ ion influx, by physical coupling to intracellular Ca²⁺ release channels or functional coupling to other ion channels such as Ca²⁺ activated potassium channels. L-type Ca²⁺ channels that comprise the family of Ca(v)1 channels are expressed in many electrically excitable tissues and are characterized by their unique sensitivity to dihydropyridines. In this issue, we summarize genetic defects in L-type Ca²⁺ channels and analyze their role in human diseases (Ca²⁺ channelopathies); e.g. mutations

XLA-MTS: a distinct clinical genetic entity characterized by immunodeficiency and neurodevelopmental delay.

BACKGROUND: Mohr-Tranebjærg syndrome (MTS) is a rare X-linked recessive neurodegenerative disorder, typically presenting with progressive sensorineural hearing loss in early childhood, followed by neurological deterioration, dystonia, ataxia, and visual impairment. OBJECTIVE: This study aimed to conduct a comprehensive clinical, genetic, and phenotypic review of MTS, with particular emphasis on mutations in the TIMM8A gene and contiguous deletions involving Bruton tyrosine kinase (BTK), which give rise to an extended phenotype known as XLA-MTS. METHODS: A systematic literature review was per

Genetic diversity of Usher syndrome in Moroccan patients.

Usher syndrome (USH) is a rare genetic disorder accounting for almost 50% of all hereditary deaf-blindness cases. This heterogeneous, autosomal recessive condition is categorized into 3 main clinical subtypes (USH1, USH2 and USH3) and an atypical form of USH based on the severity and age of onset of the hearing loss, the absence or presence of vestibular dysfunction and the age of retinitis pigmentosa (RP) onset. This study aims to characterize the genetic spectrum of Usher syndrome in the Moroccan population. We used targeted-exome, whole exome and Sanger sequencing to identify variants in US

A deafness-blindness syndrome results from ATF6-based disruption of the unfolded protein response.

Sensorineural hearing loss (SNHL) is the most prevalent form of permanent hearing impairment, arising from factors such as aging, exposure to loud noise, disease, ototoxic medications, and genetic mutations. Despite extensive research, effective treatments or cures for SNHL remain elusive. In this issue of the JCI, Lee et al. reveal a link between mutations in ATF6 and SNHL in patients with achromatopsia. The study also shows that Atf6-deficient (Atf6-/-) mice exhibit disorganized stereocilia and age-related loss of outer hair cells. Additionally, the researchers show that Atf6 is critical for

Arts syndrome with a novel missense mutation in the PRPS1 gene: A case report.

Arts syndrome is characterized by early-onset hypotonia, ataxia, intellectual disability, sensorineural hearing impairment, progressive optic atrophy, and a tendency to develop infections. Arts syndrome is an X-linked disorder caused by a loss-of-function mutation in the PRPS1 gene, which encodes phosphoribosylpyrophosphate synthetase 1. Only three families have been reported. Here, we report another family with Arts syndrome. The initial symptoms of the 1-year-old proband were hypotonia and ataxia, worsening recurrent infection-triggered muscle weakness, motor and intellectual developmental d

Horseback riding therapy for a deafblind individual enabled by a haptic interface.

We present a haptic interface to help deafblind people to practice horseback riding as a recreational and therapeutic activity. Horseback riding is a form of therapy which can improve self-esteem and sensation of independence. It has been shown to benefit people with various medical conditions-including autism. However, in the case of deafblind riders, an interpreter must stand by at all times to communicate with the rider by touch. We developed a simple interface that enables deafblind people to enjoy horseback riding while the instructor is remotely providing cues, which improves their indep

Development of the International Classification of Functioning, Disability and Health Core Sets for children and youth with deafblindness: protocol for a multistudy collaboration across regions of WHO.

INTRODUCTION: While the International Classification of Functioning, Disability and Health (ICF) Core Sets have been developed for deafblindness across the lifespan, there are currently no Core Sets specifically designed for children and youth with deafblindness. Children with deafblindness often experience unique challenges not captured in existing Core Sets, requiring a specialised framework to assess their functioning, activity limitations, participation restrictions and environmental factors. This project aims to develop ICF Core Sets specifically for children and youth with deafblindness,

Reproductive dysfunction and decreased GnRH neurogenesis in a mouse model of CHARGE syndrome.

CHARGE is a multiple congenital anomaly disorder and a common cause of pubertal defects, olfactory dysfunction, growth delays, deaf-blindness, balance disorders and congenital heart malformations. Mutations in CHD7, the gene encoding chromodomain helicase DNA binding protein 7, are present in 60-80% of individuals with the CHARGE syndrome. Mutations in CHD7 have also been reported in the Kallmann syndrome (olfactory dysfunction, delayed puberty and hypogonadotropic hypogonadism). CHD7 is a positive regulator of neural stem cell proliferation and olfactory sensory neuron formation in the olfact
