Assessing the effect of altitude of cultivation on wheat beer quality by a multi-omic approach integrating physico-chemical, volatiles and sensory data.

The cultivation of crops at high altitudes is a promising strategy to face global warming. This study aimed to unravel the effect of altitude of wheat cultivation on wheat beer quality using a multi-omic approach. Common and durum wheat were cultivated at different altitudes and used to produce Blanche craft beers, which were analysed for physico-chemical parameters, volatile organic compounds (VOCs), and sensory profile. These three blocks of data were successfully processed by a sparse multi-block-based approach able to predict altitude of wheat cultivation of beers. VOCs block was the most

Bacterial Tropone Natural Products and Derivatives: Overview of their Biosynthesis, Bioactivities, Ecological Role and Biotechnological Potential.

Tropone natural products are non-benzene aromatic compounds of significant ecological and pharmaceutical interest. Herein, we highlight current knowledge on bacterial tropones and their derivatives such as tropolones, tropodithietic acid, and roseobacticides. Their unusual biosynthesis depends on a universal CoA-bound precursor featuring a seven-membered carbon ring as backbone, which is generated by a side reaction of the phenylacetic acid catabolic pathway. Enzymes encoded by separate gene clusters then further modify this key intermediate by oxidation, CoA-release, or incorporation of sulfu

Hyperammonemia and neonatal cardiac failure.

Severe hyperammonemia (hyperNH3) in neonatal cardiac failure after cardiac surgery is rare. We report a case of a 2470-g female infant born at the week 37 of gestation with complex congenital heart disease (truncus arteriosus type III, interrupted aortic arch and tricuspid valve insufficiency) and hemodynamically non-significant intrahepatic arterio-venous malformation. She developed hyperNH3 (highest NH3 blood level: 467 µmol/L) without severe liver failure (INR of 1.9). The origin of the hyperNH3 was multifactorial including limited capacity of liver detoxification function due to congenital

Ornithine transcarbamylase deficiency.

Two infants, one male and one female, with elevated serum ammonia levels, were shown, based on urine organic acid analysis and DNA studies, to have ornithine transcarbamylase (OTC) deficiency. OTC deficiency is one of the most common urea cycle disorders. Hyperammonemia occurred at 3 days of age in the male infant, and at approximately 7 days of age in the female infant. Administration of sodium benzoate and sodium phenylacetate lowered the serum ammonia level effectively in both cases. Other modalities, including peritoneal dialysis and protein restriction, were also important in the control

A GC/MS-based metabolomic approach for reliable diagnosis of phenylketonuria.

Although the phenylalanine/tyrosine ratio in blood has been the gold standard for diagnosis of phenylketonuria (PKU), the disadvantages of invasive sample collection and false positive error limited the application of this discriminator in the diagnosis of PKU to some extent. The aim of this study was to develop a new standard with high sensitivity and specificity in a less invasive manner for diagnosing PKU. In this study, an improved oximation-silylation method together with GC/MS was utilized to obtain the urinary metabolomic information in 47 PKU patients compared with 47 non-PKU controls.

The glutathione metabolism of the beta-lactam producer filamentous fungus Penicillium chrysogenum.

Glutathione (gamma-L-glutamyl-L-cysteinyl-glycine; GSH) shares structural similarities with the beta-lactam biosynthetic intermediate ACV-tripeptide (delta-(L-alpha-aminoadipyl)-L-cysteinyl-D-valine). Not surprisingly, GSH has been reported to inhibit the beta-lactam biosynthetic machinery quite effectively and, hence, strategies to decrease the intracellular GSH concentrations without influencing negatively the physiological status of idiophasic mycelia would attract industrial interests. Here we present a detailed map of the GSH metabolic network of P. chrysogenum and show a promising way to

Spin-Labeled Diclofenac: Synthesis and Interaction with Lipid Membranes.

Diclofenac is a non-steroidal anti-inflammatory drug (NSAID) from the group of phenylacetic acid derivatives, which has analgesic, anti-inflammatory and antipyretic properties. The interaction of non-steroidal anti-inflammatory drugs with cell membranes can affect their physicochemical properties, which, in turn, can cause a number of side effects in the use of these drugs. Electron paramagnetic resonance (EPR) spectroscopy could be used to study the interaction of diclofenac with a membrane, if its spin-labeled analogs existed. This paper describes the synthesis of spin-labeled diclofenac (di

An in vitro study on the differentiated metabolic mechanism of chloroquine-resistant Plasmodium falciparum using high-resolution metabolomics.

Chloroquine (CQ) is an important drug used therapeutically for treatment of malaria. However, due to limited number of studies on metabolic targets of chloroquine (CQ), it is difficult to attribute mechanisms underlying resistance associated with usage of this drug. The present study aimed to investigate the metabolic signatures of CQ-resistant Plasmodium falciparum (PfDd2) compared to CQ-sensitive Plasmodium falciparum (Pf3D7). Both Pf3D7 and PfDd2 were treated with CQ at 200 nM for 48 hr; thereafter, the harvested red blood cells (RBCs) and media were subjected to microscopy and high-resolut

Ornithine transcarbamylase deficiency: pathogenesis of the cerebral disorder and new prospects for therapy.

Ornithine Transcarbamylase (OTC) is a key urea cycle enzyme. Congenital OTC deficiencies in humans result in hyperammonemia and a spectrum of neurological symptoms including hypotonia, seizures and mental retardation. Neuropathologic evaluation reveals cerebral atrophy, ventricular enlargement and Alzheimer type II astrocytosis. Using an animal model of congenital OTC deficiency, the sparse fur (spf) mouse, recent studies have revealed significant alterations of cholinergic, serotoninergic and glutamatergic neurotransmitter systems. Possible pathophysiologic mechanisms responsible for neuronal

Performance of indigenous microbiota and wild-type Pichia kluyveri in fermentation for quality improvement of Coffea canephora based on chemical and sensory characteristics.

This paper investigated the performance of wild-type Pichia kluyveri inoculation and indigenous microbiota during the fermentation of C. canephora var. Conilon to improve beverage quality. We assessed both the conventional dry-fermentation method and the self-induced anaerobiosis fermentation (SIAF) process over seven days. The highest sensory score occurred after four days of SIAF with P. kluyveri, yielding an increase of 7.1 points (84.4 total points). The conventional method largely preserved the native acid profile, whereas SIAF progressively reduced tartaric and succinic acids while incre

Harnessing OmpR for Enhanced Biocontrol: Characterization of a Novel Engineered Strain of Lysobacter gummosus with Increased Nematicidal Properties.

The development of green alternatives to chemical nematicides is an urgent need in sustainable agriculture. This study constructed an engineered strain, LG209, by overexpressing the transcriptional regulator OmpR, which significantly enhanced nematicidal activity against Meloidogyne incognita J2s and increased secondary metabolite production. From its fermentation broth, 20 compounds were isolated, including a new naphthoquinone derivative. Notably, phenylacetic acid (3), 2-(4-methoxyphenyl)acetic acid (5), and 9H-carbazole (18) exhibited strong nematicidal activity, with LC50 values of 52.34,

Glucocorticoid-induced changes of the gut microbiota and metabolic markers in healthy young men: Outcome of a randomized controlled trial.

Glucocorticoids induce insulin resistance and suppress immunity, but their impact on gut microbiota, which may modulate metabolism and immunity remains under explored. In this 7-day trial, we assess glucocorticoid-induced changes in gut microbiota and metabolic markers in 56 healthy men, randomly assigned to three interventions: oral prednisolone (PO group), intramuscular methylprednisolone acetate (IM group), or saline (CTL group). Shotgun metagenomics reveal that PO glucocorticoid causes shifts in bacterial abundance, increasing Blautia and Collinsella, while decreasing Dysosmobacter welbion

Association between intraspecific variability and penicillin production in industrial strain, Penicillium chrysogenum revealed by RAPD and SRAP markers.

Penicillin represents antibiotic discovered for the first time that prevents bacterial infections. Production of penicillin using penicillin-producing fungi (Penicillium chrysogenum) mainly depends on activity of industrial strain, optimization of culture condition and purification efficiency. To contribute to management of penicillin-producing strains, we conducted phylogenetic study on 27 P. chrysogenum variants originated from industrial strain and selected on media supplemented with penicillin or phenylacetic acid (PAA) using 4 Random Amplified Polymorphic DNA (RAPD) primers and 2 Sequence

Ammonia toxicity and its prevention in inherited defects of the urea cycle.

The urea cycle is the final pathway for removal of surplus nitrogen from the body, and the major route in humans for detoxification of ammonia. The full complement of enzymes is expressed only in liver. Inherited deficiencies of urea cycle enzymes lead to hyperammonaemia, which causes brain damage. Severe defects present with hyperammonaemic crises in neonates. Equally devastating episodes may occur in previously asymptomatic adults with mild defects, most often X-linked ornithine transcarbamylase (OTC) deficiency. Several mechanisms probably contribute to pathogenesis. Treatment aims to reduc

Why did the Fleming strain fail in penicillin industry?

Penicillin, discovered 75 years ago by Sir Alexander Fleming in Penicillium notatum, laid the foundations of modern antibiotic chemotherapy. Early work was carried out on the original Fleming strain, but it was later replaced by overproducing strains of Penicillium chrysogenum, which became the industrial penicillin producers. We show how a C(1357)-->T (A394V) change in the gene encoding PahA in P. chrysogenum may help to explain the drawback of P. notatum. PahA is a cytochrome P450 enzyme involved in the catabolism of phenylacetic acid (PA; a precursor of penicillin G). We expressed the pahA
