Formation of the Mouse Internal Capsule and Cerebral Peduncle: A Pioneering Role for Striatonigral Axons as Revealed in Isl1 Conditional Mutants.

The projection neurons of the striatum, the principal nucleus of the basal ganglia, belong to one of the following two major pathways: the striatopallidal (indirect) pathway or the striatonigral (direct) pathway. Striatonigral axons project long distances and encounter ascending tracts (thalamocortical) while coursing alongside descending tracts (corticofugal) as they extend through the internal capsule and cerebral peduncle. These observations suggest that striatal circuitry may help to guide their trajectories. To investigate the developmental contributions of striatonigral axons to internal

Sleep Disorders in Pediatric Patients Affected by Neurofibromatosis Type 1: Reports of a Questionnaire and an Apple Watch Sleep Assessment.

Introduction: Sleep is a fundamental biological function critical for physical and mental health. Chronic sleep disturbances can significantly impair cognitive, emotional, and social functioning, leading to deficits in attention, alertness, and executive function, alongside increased irritability, anxiety, and depression. For pediatric patients, such disturbances pose additional concerns, potentially disrupting developmental processes and quality of life for both children and their families. Objectives: Emerging evidence suggests a correlation between neurofibromatosis type 1 (NF1) and an incr

De novo unbalanced translocation (4p duplication/8p deletion) in a patient with autism, OCD, and overgrowth syndrome.

Chromosomal abnormalities, such as unbalanced translocations and copy number variants (CNVs), are found in autism spectrum disorders (ASDs) [Sanders et al. (2011) Neuron 70: 863-885]. Many chromosomal abnormalities, including sub microscopic genomic deletions and duplications, are missed by G-banded karyotyping or Fragile X screening alone and are picked up by chromosomal microarrays [Shen et al. (2010) Pediatrics 125: e727-735]. Translocations involving chromosomes 4 and 8 are possibly the second most frequent translocation in humans and are often undetected in routine cytogenetics [Giglio et

Association of anxiety and ODD/CD in children with and without ADHD.

The goal of this study is to examine levels of oppositional defiant disorder (ODD) and conduct disorder (CD) in four groups of children: attention-deficit/hyperactivity disorder (ADHD) only, anxiety only, ADHD and anxiety, and controls (i.e., non-ADHD youth). Although children with ADHD exhibit more ODD and CD than non-ADHD youth, it is unknown if anxiety is associated with increased or decreased ODD and CD in children with ADHD. We examined parent and teacher ratings of ODD and CD from the Disruptive Behavior Disorder Rating Scale in 203 school age children (ages 6-9); 70% were male, and 47%

Intrauterine Growth and Offspring Neurodevelopmental Traits: A Mendelian Randomization Analysis of the Norwegian Mother, Father and Child Cohort Study (MoBa).

IMPORTANCE: Conventional epidemiological analyses have suggested that lower birth weight is associated with later neurodevelopmental difficulties; however, it is unclear whether this association is causal. OBJECTIVE: To investigate the relationship between intrauterine growth and offspring neurodevelopmental difficulties. DESIGN, SETTING, AND PARTICIPANTS: MoBa is a population-based pregnancy cohort that recruited pregnant women from June 1999 to December 2008 included approximately 114 500 children, 95 200 mothers, and 75 200 fathers. Observational associations between birth weight and neur

Using a Developmental Approach to Investigate Behavioral, Neurodevelopmental, and Depressive Irritability Types.

OBJECTIVE: Irritability is highly heterogeneous and a common challenge in youth clinical services. Albeit transdiagnostic, the diagnostic manuals conceptualize severe irritability differently: the ICD-11 primarily recognizes it as behavioral (oppositional defiant disorder specifier), and the DSM-5-TR, as depressive (core to disruptive mood dysregulation disorder). Irritability is also highly prevalent in, and genetically linked to, neurodevelopmental conditions. It is unclear whether irritability represents a unitary construct or multiple different types. We examined whether distinct behaviora

Increased Risk of Obesity and Metabolic Dysregulation Following 12 Months of Second-Generation Antipsychotic Treatment in Children: A Prospective Cohort Study.

OBJECTIVE: To determine the risk of developing obesity and related metabolic complications in children following long-term treatment with risperidone or quetiapine. METHODS: This was a 1-year naturalistic longitudinal study conducted between February 2009 and March 2012. A total of 130 children aged 2 to 18 years without prior exposure to second-generation antipsychotics (SGAs) were enrolled at initiation of treatment with either risperidone or quetiapine. Metabolic parameters were measured at baseline and months 6 and 12. Data of 37 participants (20 treated with risperidone and 17 treated wi

The selective D3-Receptor antagonist VK4-116 effectively treats behavioral inflexibility in rats caused by self-administration and withdrawal from cocaine.

Chronic psychostimulant use can cause long lasting changes to neural and cognitive function that persist even after long periods of abstinence. As cocaine users transition from drug use to abstinence, a parallel transition from hyperactivity to hypoactivity has been found in orbitofrontal-striatal glucose metabolism, and striatal D2/D3 receptor activity. Targeting these changes pharmacologically, using highly selective dopamine D3 receptor (D3R) antagonists and partial agonists, has shown significant promise in reducing drug-taking, and attenuating relapse in animal models of cocaine and opioi

Prenatal Alcohol Exposure and Mitochondrial Function in the Brain.

Prenatal alcohol exposure (PAE) is recognized as a leading preventable cause of birth defects, giving rise to a continuum of cognitive, behavioral, and physical impairments collectively referred to as Fetal Alcohol Spectrum Disorders (FASD). While PAE affects multiple developing organ systems, the fetal brain is particularly vulnerable, exhibiting enduring structural and functional abnormalities in response to alcohol exposure. Recent research highlights mitochondrial dysfunction as an important mechanism in the pathogenesis of alcohol-related neurodevelopmental deficits. Mitochondria are high

Is Persistent Motor or Vocal Tic Disorder a Milder Form of Tourette Syndrome?

BACKGROUND: Persistent motor or vocal tic disorder (PMVT) has been hypothesized to be a forme fruste of Tourette syndrome (TS). Although the primary diagnostic criterion for PMVT (presence of motor or vocal tics, but not both) is clear, less is known about its clinical presentation. OBJECTIVE: The goals of this study were to compare the prevalence and number of comorbid psychiatric disorders, tic severity, age at tic onset, and family history for TS and PMVT. METHODS: We analyzed data from two independent cohorts using generalized linear equations and confirmed our findings using meta-analys

Feasibility of an Emergency Department-Based Two-Step ADHD Screening Pathway in Primary School-Age Children Presenting with Home Accidents: A Prospective Two-Center Case-Control Study.

Background/Objectives: Emergency department (ED) visits for pediatric home accidents may provide an opportunity to identify children who warrant evaluation for attention-deficit/hyperactivity disorder (ADHD). This study primarily evaluated the feasibility of a two-step ED-based ADHD screening and Child and Adolescent Psychiatry referral pathway. A secondary exploratory objective was to compare the frequency of confirmed ADHD between children presenting with home accidents and non-injured ED controls. Methods: This prospective, two-center case-control study was conducted in two emergency depart

Machine learning-driven blood biomarker profiling and EGCG intervention in fetal alcohol spectrum disorder.

Fetal alcohol spectrum disorder (FASD) is a complex neurodevelopmental condition caused by prenatal alcohol exposure (PAE), often underdiagnosed due to heterogeneous symptoms and diagnostic challenges. This study aimed to identify serum-based biomarkers for early FASD diagnosis and assess the potential of epigallocatechin gallate (EGCG), a natural antioxidant found in green tea, in modulating markers related to FASD. Luminex immunoassays were employed to analyze serum samples from FASD patients, identifying seven predictive biomarkers involved in neuroinflammation and immune dysregulation: IL-

A new era for myelin research in Neurofibromatosis type 1.

Evidence for myelin regulating higher-order brain function and disease is rapidly accumulating; however, defining cellular/molecular mechanisms remains challenging partially due to the dynamic brain physiology involving deep changes during development, aging, and in response to learning and disease. Furthermore, as the etiology of most neurological conditions remains obscure, most research models focus on mimicking symptoms, which limits understanding of their molecular onset and progression. Studying diseases caused by single gene mutations represents an opportunity to understand brain dys/fu

Bisphenol S and Neurological Health: An Integrated Overview of Neurotoxicity and Underlying Mechanisms.

Bisphenol S (BPS), a pervasive contaminant used in consumer and industrial products, has been widely detected in human serum, urine, hair, placenta, and breast milk. Although initially studied mainly as an endocrine disruptor, accumulating evidence indicates that BPS also exerts neurotoxicity by perturbing neuroinflammatory responses, neuroendocrine regulation, and neuronal development. However, its neurotoxic profile and mechanistic basis remain incompletely defined. This review systematically retrieved and synthesized epidemiological, animal, and cellular studies published between 2000 and F

[The projection of autism spectrum disorders in adult life].

Autism Spectrum Disorders (ASDs) consist a group of neurodevelopmental disorders that are usually diagnosed in early childhood but they persist throughout life, although significant changes can happen. The prevalence of the ASDs is estimated to be 1-1.2%. Subjects with the more severe form of the disorder that are usually characterised by the absence of a communicative language and learning difficulties of various severity, are often referred as persons with lower functioning. In the other end of the spectrum we can find subjects with less severe symptomatology, communicative language and at l

The effect of antibiotics on social aversion following early life inflammation.

Epidemiological evidence suggests that exposure to infection during early development increases the risk for neurodevelopmental disorders associated with symptoms such as a decreased desire to engage in social interactions. In animals, disruptions in social behavior can be modelled by administering bacterial mimetics such as liposaccharide (LPS). However, when evaluating social interactions in the laboratory, attention is rarely directed on the reciprocal relationship as a whole, which is important as peers may drive social withdrawal. Previously, we have shown that male adolescent rats treate

Oppositional defiant disorder: information for school nurses.

Oppositional defiant disorder, one of the disruptive behavior disorders, has far-reaching consequences for the individual, family, school, community, and society. Early recognition allows interventions geared toward promotion of prosocial behaviors, possibly halting progression to the more deviant conduct disorder. Awareness of this disorder and associated comorbidities is the first step that enables the school nurse to use assessment techniques and to assist in planning of interventions for the child. Children at risk often show tendencies toward oppositional defiant disorder as early as pres

Coding of Childhood Psychiatric and Neurodevelopmental Disorders in Electronic Health Records of a Large Integrated Health Care System: Validation Study.

BACKGROUND: Mental, emotional, and behavioral disorders are chronic pediatric conditions, and their prevalence has been on the rise over recent decades. Affected children have long-term health sequelae and a decline in health-related quality of life. Due to the lack of a validated database for pharmacoepidemiological research on selected mental, emotional, and behavioral disorders, there is uncertainty in their reported prevalence in the literature. OBJECTIVES: We aimed to evaluate the accuracy of coding related to pediatric mental, emotional, and behavioral disorders in a large integrated he
