Haplotype Blocks Are Associated With Rapid Local Adaptation to Environmental Shifts in Wild Barley.

Genomic mechanisms of local adaptation must be highly responsive in geographic regions where climate is changing rapidly. The Levant region is a critical biodiversity hotspot and the distribution edge for many species, including the wild ancestor of domesticated barley. This region is under an accelerated desertification process, thus enforcing a rapid genomic response to the projected environmental changes. To elucidate the genomic basis of rapid local adaptation, we studied wild barley populations using an ecological-genetic sampling design that decouples environmental variation from demogra

CsNIP2 and CsNIP16 facilitated Al-F transport and hyperaccumulation in tea plants (Camellia sinensis).

Although Al-F complexes (e.g., AlF2+, AlF2+) exist in the tea apoplast and symplast, their root uptake mechanisms and translocation to mature leaves remain elusive. Here, we identified two plasma membrane-localized Nodulin 26-like Intrinsic Proteins (NIPs), CsNIP2 and CsNIP16, as crucial mediators of Al-F complex transport. CsNIP2 is predominantly expressed in roots and enriched in the phloem-epidermic part tissues, whereas CsNIP16 is predominantly expressed in stems and abundantly in xylem tissues. Heterologous expression in yeast revealed that both transporters facilitate Al-F uptake, signif

Loss of function of STAY-GREEN 1 enhances cold tolerance in tomato (Solanum lycopersicum L.) associated with jasmonic acid signaling and metabolic reprogramming.

Photosynthetic machinery reprogramming is essential for Cold Stress Tolerance (CST) in plants. STAY-GREEN 1 (SlSGR1) plays vital roles in chlorophyll breakdown and leaf senescence, but its function in tomato cold acclimation has not been clarified. Here, we constructed two inbred tomato lines (T048, T069) carrying SlSGR1 homozygous knockout mutants via CRISPR/Cas9 editing, and conducted integrated physiological, biochemical, metabolomic, and transcriptome analyses to dissect the role of SlSGR1 in regulating cold tolerance. Loss of function of SlSGR1 triggered extensive physiological, metabolic

Sweet Sorghum Microspores' Fractionalization, Individual Isolation, and Whole-Genome Amplification.

Sweet sorghum is one of the important crops that has been widely reported to be recalcitrant to genetic manipulation endeavors. It can serve as biofuel, food, and food per production systems toward energy sources as well as human and animal sustenance. To deploy biotechnology tools in advancement of this crop of such agronomic importance, genetic and molecular investigation for understanding is necessary. Underdeveloped cell walls are a significant physiochemical state for microspores per their developmental stages, which makes it ideal for DNA isolation, being readily lysed as natural protopl

Transcriptome and metabolome analysis of Cycas bifida response to low-temperature stress.

BACKGROUND: Cycas bifida is an endemic species distributed in limited areas of Yunnan and Guangxi, with a notably small population size. Under the context of global climate warming, this species may require introduction to colder habitats. Previous studies have confirmed its weak cold tolerance. To thoroughly investigate the molecular and metabolic mechanisms underlying its response to low temperature, this study employed low-temperature stress treatments combined with transcriptome sequencing and metabolomics technologies for systematic comparative analysis. RESULTS: Through multi-omics anal

Adenoviruses in Wild Bog Turtles (Glyptemys muhlenbergii) of the Northeastern USA.

Bog turtles (Glyptemys muhlenbergii) are critically endangered semiaquatic turtles inhabiting fens and bogs along the east coast of the USA. Threats to their survival include habitat loss from development, roads, altered hydrology, ecological succession, proliferation of invasive plants, poaching, and infectious disease. In response to an increase in bog turtle mortality and concerns about their health, we assessed bog turtle health and pathogen prevalence across five USA states-New York, Pennsylvania, New Jersey, Delaware, and Maryland-between 2014 and 2022. Adenoviruses, specifically barthad

Agrochemical contaminants in six species of edible insects from Uganda and Kenya.

Edible insects are currently promoted worldwide as an alternative animal protein source, but they are mostly still harvested from the wild where they are predisposed to contamination with agrochemicals. This study analysed six species of edible insects (Ruspolia differens, Rhynchophorus phoenicis, Schistocerca gregaria, Oryctes sp, Pachnoda ephippiata and Acanthoplus sp) collected from different habitats and/or reared in the laboratory in Kenya and Uganda for safety from agrochemical contaminants using liquid chromatography tandem mass spectrometry. The residue levels were statistically compar

A Survey of Recent Patents in Engineering Technology for the Screening, Separation and Processing of Eggshell.

The chicken egg is a well-known complete food of human daily consumption which serves as a cost-effective, high-quality nutrient resource. About 30% of table eggs are directed to breaker plants in developed countries, leading to the generation of substantial eggshell (ES) waste, which is increasingly explored for potential value-added applications. The number of patents describing ES-based applications has increased dramatically in recent years. This review provides insight into the most recent patents published between 2015 and 2020, with focus on different engineering technologies for the sc

Hybrid incompatibilities in interspecific crosses between tetraploid wheat and its wild diploid relative Aegilops umbellulata.

Hybrid abnormalities, severe growth abortion and grass-clump dwarfism, were found in the tetraploid wheat/Aegilops umbellulata hybrids, and the gene expression changes were conserved in the hybrids with those in other wheat synthetic hexaploids. Aegilops umbellulata Zhuk., a diploid goatgrass species with a UU genome, has been utilized as a genetic resource for wheat breeding. Here, we examine the reproductive barriers between tetraploid wheat cultivar Langdon (Ldn) and various Ae. umbellulata accessions by conducting interspecific crossings. Through systematic cross experiments, three types o

Tissue culture of Juniperus chinensis: effects of sex and leaf morphology on in vitro regeneration.

BACKGROUND AND AIMS: In tissue culture of Juniperus chinensis, issues such as inadequate sterilization, severe browning and poor rooting have significantly hindered the widespread application of techniques of its asexual propagation. Meanwhile, J. chinensis presents three sexes: female (FP), male (MP) and monoecious (MOP), with leaf morphological differentiation into scale leaves (SL) and spiny leaves (SPL). Variations in sexes and leaf morphogenesis may affect the establishment of tissue culture systems. However, there are currently no studies that have established tissue culture systems by i

The genetics of metal tolerance in vascular plants.

In many parts of the world soils are detrimental to plant growth owing to elevated levels of metal ions, caused either by natural processes or by the result of man's activities. Many plants have evolved ecotypes or varieties that are able to grow more-or-less normally on these soils. This paper reviews our knowledge of the genetics of this phenomenon. The nature of tolerance and the problems of its measurement are discussed. Tolerance is frequently measured by an index produced by comparing growth in a contaminated environment with growth in a control environment. It is argued that this measur

Analysis on Changes and Influencing Factors of the Intestinal Microbiota of Alpine Musk Deer between the Place of Origin and Migration.

In China, the population of wild musk deer, belonging to the family Moschidae, has drastically decreased in recent years owing to human activities and environmental changes. During the 1990s, artificial breeding of Alpine musk deer was conducted in Xinglong Mountain, Gansu Province, China, and their ex situ conservation was explored for over a decade. Ex situ protection is beneficial for expanding the population of animals and maintaining their genetic diversity; however, it can also induce metabolic diseases and parasitic infections and reduce reproductive capacity. The gut microbiota of anim

[Genome-wide identification, expression profiling, and natural allelic variation analysis of the ckx genes in sorghum (Sorghum bicolor)].

Cytokinin (CK) is one of the essential plant hormones, and cytokinin oxidase (CKX) is the key enzyme for CK degradation. Currently, the ckx gene family of rice (Oryza sativa) and maize (Zea mays) have been characterized, while a genome-wide analysis of the ckx gene family in sorghum (Sorghum bicolor) has not been reported. This study aims to identify the Sbckx gene family in sorghum, analyze its evolutionary characteristics and expression patterns, and screen for key genes. A combined approach of bioinformatics analysis and experimental validation was utilized to examine sorghum subjected to a

Comparative transcriptomics reveals hormone signaling and MADS-box genes in divergent development of inflorescences and tendrils in grapevine lateral shoots.

Hormone signaling and MADS-box genes regulate grapevine tendril and inflorescence growth divergence, offering molecular insights for managing tendril growth. Grapevine (Vitis vinifera L.) tendrils and inflorescences are homologous organs; however, their divergent development has important agronomic consequences because excessive tendril growth increases vineyard management costs. To explore the regulatory mechanisms, we compared the inflorescence-prone cultivar 'Einset Seedless' (ENT) with the tendril-prone cultivar 'Pinot Noir' (PN) using anatomical observation, transcriptome analysis of spec

Response of some faba bean genotypes to heat stress under environmental variations.

The faba bean productivity is affected by high temperatures during the growing season. Therefore, faba bean genotypes must be developed to tolerate heat stress. In this study, nine genotypes were assessed over two years and two locations under normal and heat-stress conditions to identify high-yielding, and heat-tolerant genotypes. Seed and straw yields, along with most of the traits under study, showed high genotype diversity and genetic variability across years and locations under in both conditions, according to the combined ANOVA. Seed yield and other traits under investigation were signif

Genetic dissection of inherited cold-stress reveals resilience hotspots in barley.

Cold episodes during early crop development threaten cereal establishment and yield, but it remains unclear whether ancestral cold exposure can generate measurable inherited resilience in barley. We tested this question in a three-generation field experiment using 138 barley accessions and eight third-generation lineage-treatment combinations that separated no-memory, transgenerational, intergenerational and repeated cold-memory classes under control and recurrent cold-stress conditions. Cold stress was imposed by delayed sowing, and establishment, physiological, biochemical, growth, reproduct

Reframing fruit biofortification for sustainable micronutrient security: from crop innovation to nutritional impact.

Micronutrient malnutrition, commonly referred to as "hidden hunger, " remains a persistent global health challenge, particularly in regions with limited dietary diversity. Although agricultural intensification has substantially improved caloric availability, it has not ensured adequate micronutrient density in food systems, highlighting the urgent need for nutrition-sensitive crop improvement strategies. Biofortification has emerged as a sustainable and cost-effective approach to enhance the micronutrient content of food crops through agronomic, genetic, and biotechnological interventions. Whi
