Vitamin-D-dependent calcium-binding-protein and parvalbumin occur in bones and teeth.

Vit-D-Dependent calcium binding protein and parvalbumin have both been detected in ameloblasts and calcified cartilage by immunohistochemical techniques. These two Ca2+ binding proteins may play a crucial role in the local accumulation of Ca2+ ions during the process of mineralization. The mechanisms underlying the deposition of inorganic substances in bone and teeth during physiologic calcification are still the object of intense debate [1, 2]. The hypotheses concerning the factors controlling the initiation of mineralization can be subdivided into three large categories: enzymatic (or non-en

Relationship between cellular retinoic acid-binding protein and histology of human lung tumors.

Human lung tumors from 28 patients were investigated by sucrose density analysis for the presence of cellular retinoic acid-binding proteins. In the tumor-free lung tissue and in most of the 20 squamous cell carcinoma and in 1 small cell carcinoma specimens tested no specific binding of retinoic acid was detectable, whereas these binding proteins were observed in 5 adeno- and 1 large cell carcinoma. The amount of retinoic acid-binding protein was slightly increased in 1 bronchial adenoma considered to be a semimalignant tumor. These findings suggest that the occurrence of retinoic acid-binding

Plasma parathyroid hormone levels and intestinal calcium binding protein in magnesium deficient rats.

Rats were pair fed a magnesium deficient or control diet. Plasma parathyroid hormone (PTH) was estimated by radioimmunoassay using synthetic 1-34 PTH and intestinal calcium-binding protein (CaBP) was quantified directly by RIA in proximal duodenum, distal ileum and medium jejunum. In magnesium depleted rats, plasma magnesium levels were significantly decreased, a fall in plasma phosphate paralleled the decrease in plasma magnesium and plasma calcium levels were significantly increased after 14 days of magnesium deficiency. A significant rise in plasma PTH was observed on day 7 and 14 after mag

Organic anion-binding protein in rat liver: drug induction and its physiologic consequence.

The administration of phenobarbital to rats enhanced hepatic uptake of an organic anion, bromsulphalein, in vivo and simultaneously increased the amount of Y, a hepatic cytoplasmic organic anion-binding protein. This study supports the postulate that Y is a major determinant in the selective hepatic uptake of certain organic anions from plasma. Induction of Y may contribute to the enhanced hepatic uptake and metabolism of various organic anions (drugs, hormones, etc.) produced by phenobarbital and other agents.

Binding of Glycans to the SARS CoV-2 Spike Protein, an Open Question: NMR Data on Binding Site Localization, Affinity, and Selectivity.

We have used NMR experiments to explore the binding of selected glycans and glycomimetics to the SARS CoV-2 spike glycoprotein (S-protein) and to its receptor binding domain (RBD). STD NMR experiments confirm the binding of sialoglycans to the S-protein of the prototypic Wuhan strain virus and yield dissociation constants in the millimolar range. The absence of STD effects for sialoglycans in the presence of the Omicron/BA.1 S-protein reflects a loss of binding as a result of S-protein evolution. Likewise, no STD effects are observed for the deletion mutant Δ143-145 of the Wuhan S-protein, thu

The Duffy binding protein (PkDBPαII) of Plasmodium knowlesi from Peninsular Malaysia and Malaysian Borneo show different binding activity level to human erythrocytes.

BACKGROUND: The zoonotic Plasmodium knowlesi is a major cause of human malaria in Malaysia. This parasite uses the Duffy binding protein (PkDBPαII) to interact with the Duffy antigen receptor for chemokines (DARC) receptor on human and macaque erythrocytes to initiate invasion. Previous studies on P. knowlesi have reported distinct Peninsular Malaysia and Malaysian Borneo PkDBPαII haplotypes. In the present study, the differential binding activity of these haplotypes with human and macaque (Macaca fascicularis) erythrocytes was investigated. METHODS: The PkDBPαII of Peninsular Malaysia and Ma

Androgen regulation of the messenger RNA encoding diazepam-binding inhibitor/acyl-CoA-binding protein in the human prostatic adenocarcinoma cell line LNCaP.

To study the mechanisms by which androgens intervene in the regulation of growth and differentiation of human prostatic epithelial cells, cDNA clones encoding putative prostate-secreted proteins were characterized and tested as potential markers for androgen action. One of the isolated cDNAs expressed diazepam-binding inhibitor/acyl-CoA-binding protein (DBI/ACBP), suggesting that this polypeptide, that has been implicated in a large number of biochemical processes, is expressed and secreted by prostate cells. As demonstrated by Northern blot analysis, the mRNA encoding DBI/ACBP was expressed i

Tannic acid and chromic chloride-induced binding of protein to red cells: a preliminary study of possible binding sites and reaction mechanisms.

The binding mechanisms and binding sites involved in the tannic acid and chromic chloride-induced binding of protein to red cells were investigated using the binding of IgA paraprotein to red cells as model systems. Inhibition studies of these model systems using amino acid homopolymers and compounds (common as red cell membrane constituents) suggest that the mechanisms involved are similar to those proposed for the conversion of hide or skin collagen to leather, as in commercial tanning. These studies also suggest that tannic acid-induced binding of IgA paraprotein to red cells involves the a

Anti-Müllerian hormone binds heat shock protein glucose-regulated protein 78 in human follicular fluid.

PURPOSE: Anti-Müllerian hormone (AMH) regulates key steps in folliculogenesis, yet unlike other TGF-β family members, no soluble AMH-binding proteins have been identified. We hypothesized that AMH may bind shuttling proteins in human follicular fluid (FF). METHODS: Discarded pooled FF was collected from women undergoing oocyte retrieval at an academic fertility center. AMH-binding soluble proteins in FF were co-immunoprecipitated with AMH, identified by mass spectrometry, and their association was validated by western blot. We characterized the spatiotemporal expression of candidate binding p

Investigation of protein-protein interactions and hotspot region on the NSP7-NSP8 binding site in NSP12 of SARS-CoV-2.

Background: The RNA-dependent RNA polymerase (RdRp) complex, essential in viral transcription and replication, is a key target for antiviral therapeutics. The core unit of RdRp comprises the nonstructural protein NSP12, with NSP7 and two copies of NSP8 (NSP81 and NSP82) binding to NSP12 to enhance its affinity for viral RNA and polymerase activity. Notably, the interfaces between these subunits are highly conserved, simplifying the design of molecules that can disrupt their interaction. Methods: We conducted a detailed quantum biochemical analysis to characterize the interactions within the NS

The Expression of the Cold Shock Protein RNA Binding Motif Protein 3 is Transcriptionally Responsive to Organ Temperature in Mice.

BACKGROUND: Mild hypothermia, i.e. maintenance of organ temperature by up to 8°C lower than body temperature, is a critical strategy for exerting some functions of the cells and organs normally, and is an useful therapy for recovering properly from some diseases, including myocardial infarction, cardiac arrest, brain injury, and ischemic stroke. Nevertheless, there were no focusses so far on organ temperature and potential responses of gene expression to organ temperature in organs of homeothermic animals that survive under normal conditions. OBJECTIVE: The present study aimed to assess organ

The Role of RNA-Binding Protein OsTudor-SN in Post-Transcriptional Regulation of Seed Storage Proteins and Endosperm Development.

Tudor-SN is involved in a myriad of transcriptional and post-transcriptional processes due to its modular structure consisting of 4 tandem SN domains (4SN module) and C-terminal Tsn module consisting of Tudor-partial SN domains. We had previously demonstrated that OsTudor-SN is a key player for transporting storage protein mRNAs to specific ER subdomains in developing rice endosperm. Here, we provide genetic evidence that this multifunctional RBP is required for storage protein expression, seed development and protein body formation. The rice EM1084 line, possessing a nonsynonymous mutation in

Development of a Novel Green Fluorescent Protein-Based Binding Assay to Study the Association of Plakins with Intermediate Filament Proteins.

Protein-protein interactions are fundamental for most biological processes, such as the formation of cellular structures and enzymatic complexes or in signaling pathways. The identification and characterization of protein-protein interactions are therefore essential for understanding the mechanisms and regulation of biological systems. The organization and dynamics of the cytoskeleton, as well as its anchorage to specific sites in the plasma membrane and organelles, are regulated by the plakins. These structurally related proteins anchor different cytoskeletal networks to each other and/or to

Specificity and selectivity evaluations of ligand binding assay of protein therapeutics against concomitant drugs and related endogenous proteins.

Macromolecule drugs designed against specific target proteins/ receptors have been applied in combination therapies, especially for complex and related diseases such as cancer for synergistic efficacy and alleviation of side effects. Protein therapeutics are typically measured using ligand binding assays (LBA). Evaluating the specificity and selectivity of LBA against their target proteins or in instances where concomitantly administered drugs are given was brought up during a conversation at the 3rd American Association of Pharmaceutical Scientists/US Food and Drug Administration Bioanalytica

Novel human plasma proteins, IHRP (acute phase protein) and PHBP (serine protease), which bind to glycosaminoglycans.

A novel human plasma protein was found in the eluate from the dextran sulfate column, which was used for the treatment of the patients with hypercholesteremia to reduce plasma low density lipoprotein. The results of sequence analysis revealed that this protein was a homologue of heavy chains of inter-alpha-trypsin inhibitor (ITI) family, and it was termed IHRP (ITI family heavy chain related protein). IHRP was identified as an acute-phase protein in animals, and slightly increased concentrations in human plasmas were observed in the patients with inflammatory disorders. IHRP bound to actin and

Gene expression of cellular retinol-binding protein I (CRBP I) is affected by dietary proteins in the rat liver.

The effect of dietary proteins and vitamin A status on the gene expression of cellular retinol-binding protein I (CRBP I) was studied in the rat liver. The gene expression was estimated as amounts of transcript (mRNA) by Northern blot analysis using rat CRBP I cDNA. Though vitamin A status is known to positively regulate the gene expression of CRBP I in the extrahepatic tissues, in the present study we observed that the amount of the CRBP I transcript in liver was neither reduced by vitamin A-deficiency, nor affected by replenishment with an excess dose of all-trans retinoic acid. These result

The effect of dietary protein supplementation on insulin-like growth factors (IGFs) and IGF-binding proteins in children with shigellosis.

Nutrient deficiency causes growth failure and decreases serum insulin-like growth factor-I (IGF-I) concentrations. Because IGFBPs modulate the concentrations and availability of IGFs in serum, IGF-binding proteins (IGFBPs) were measured along with IGF-I and IGF-II before and after 21 days of refeeding in 22 undernourished Bangladeshi children (2-4 yr of age) with shigellosis. The effects of a 150 Cal/kg.day diet with a normal protein (6%; n = 10) or high protein (15%; n = 12) content were studied. The results were compared with those of 25 age-matched healthy American children (controls). Body

[Effect of dietary protein and vitamin B group deficiencies on the calcium-binding protein content in the mucosa of the small intestine].

The content of calcium-binding protein (CBP) and calcium-binding activity of small intestinal mucoproteins were examined in growing male rats kept on the diet deficient in proteins and group B vitamins (thiamine, riboflavin and pyridoxine). CBP was isolated by means of electrophoresis in 7.5% polyacrylamide gel. Calcium-binding activity of mucoproteins was explored with the aid of dialysis followed by complexonometric titration. The deficiency of food proteins was shown to lead to substantial changes in the content of vitamin D-dependent CBP and calcium-binding activity of mucoproteins. The de
