Genomic and chromosomal Instability: Hidden drivers of Glioblastoma aggressiveness.

Glioblastoma is the most common and aggressive primary brain tumor in adults, characterized by rapid proliferation, diffuse infiltration, and poor clinical outcomes despite multimodal therapy. A hallmark of GBM biology is its remarkable genomic (GI) and chromosomal instability (CIN), which act as fundamental engines of tumor evolution. These processes fuel extensive intratumoral heterogeneity, drive the emergence of therapy-resistant clones, and contribute to immune evasion and disease recurrence. GI arises from replication stress, oxidative damage, and impaired DNA damage response pathways, w

Age specificity in chronic stress: psychometric properties and factorial structure for higher age of the short version of the Trier Inventory of Chronic Stress.

BACKGROUND: There are many ways, tests, and questionnaires to measure chronic stress. One of them is the Trier Inventory for Chronic Stress (TICS). Meanwhile, there are three different forms of this particular questionnaire: the original version of the TICS with 57 items, the 12-item short screening scale of the TICS (Short Screening Scale for Chronic Stress, SSCS), and a newer short form with only 9 items. METHODS: The TICS-9 was answered by N = 765 healthy participants aged 60 to 93 years. The sample was selected by random-route sampling. Confirmatory factor analyses applying Robust Maximum

Towards a standard toolbox for compatibility testing of pediatric drug products: verification of standard vehicles for fruit juice, apple sauce, yogurt, and pudding through comparative biorelevant dissolution testing using pediatric furosemide mini-tablets.

In vitro compatibility assessments are a valuable tool for the early identification of potential drug-vehicle interactions. To standardize such evaluations, a set of standard vehicles, also referred to as simVehicles (simJuice, simApplesauce, simYogurt, and simPudding) because they simulate the key physicochemical properties of commonly used pediatric dosing vehicles, was previously developed using a Design of Experiments (DoE) approach to replicate these properties while ensuring reproducibility under controlled conditions. The present study aimed to validate selected simVehicles through comp

Deciphering interventional dynamical causality via Kolmogorov-Arnold networks.

Identifying causal relationships from observational time-series data is a central challenge in understanding complex dynamical systems. However, few existing methods attempt to capture the intuitive human understanding of causality: that an intervention on one variable leads to a subsequent change in another. To bridge this gap, this paper introduces a causal discovery method named Intervened TaylorKAN (ITK), which simulates controlled experiments within reconstructed systems. Specifically, we leverage the recently emerging Kolmogorov-Arnold Networks (KANs) with Taylor series as activation fun

Dual SOT Switching Modes in a Single Device Geometry for Neuromorphic Computing.

Neuromorphic computing aims to replicate the brain's efficient processing through artificial neurons and synapses, requiring binary and multilevel switching. We present a PtMn/(Co/Pd)4/Ta device that uniquely enables dual spin-orbit torque (SOT) switching modes─binary and multilevel (analog)─within the same geometry and stack structure, eliminating the need for device modifications. Binary SOT switching is achieved via domain wall nucleation and propagation at moderate current levels (∼65 mA), while multilevel switching occurs via domain nucleation mode without significant propagation after a

SIP2 is the master transcription factor of Plasmodium merozoite formation.

Malaria, one of the most serious infectious diseases worldwide, is caused by the proliferation of Plasmodium parasites through repeated cycles of intraerythrocytic development. The parasite replicates via schizogony in host erythrocytes, producing multiple progeny merozoites that invade new erythrocytes. Although merozoite formation is the most crucial step in malaria pathogenesis, its molecular mechanism remains unclear. SIP2 is an AP2 transcription factor expressed during schizogony and is particularly conserved among erythrocyte-infecting apicomplexan parasites. Here, we reveal that SIP2 in

Integrative Computational Approaches for Understanding Drug Resistance in HIV-1 Protease Subtype C.

Acquired immunodeficiency syndrome (AIDS) is a chronic disease condition caused by the human immunodeficiency virus (HIV). The widespread availability of highly active antiretroviral therapies has helped to control HIV. There are ten FDA-approved protease inhibitors (PIs) that are used as part of antiretroviral therapies in HIV treatment. Importantly, all these drugs are designed and developed against the protease (PR) from HIV subtype B. On the other hand, HIV-1 PR subtype C, which is the most dominant strain in countries including South Africa and India, has shown resistance to PIs due to it

A dual-target real-time PCR for proactive detection of Mpox variants.

In 2022, cases of Monkeypox virus (MPXV) in California contained a mutation in the TNF receptor gene (GR2G) that rendered the virus undetectable using a widely adopted public health diagnostic qPCR assay. This underscored the need for a dual-target PCR approach and prompted validation of a second target by the BCCDC Public Health Laboratory. In addition to the GR2G target validated in the original qPCR assay (and duplexed with the endogenous target human β-globin (HBG)), GP113 (OPG128) was identified and validated using both clinical samples and MPXV DNA controls. Mutations in GR2G and GP113 (

Mechanism-guided engineering of a minimal biological particle for genome editing.

The widespread application of genome editing to treat and cure disease requires the delivery of genome editors into the nucleus of target cells. Enveloped delivery vehicles (EDVs) are engineered virally derived particles capable of packaging and delivering CRISPR-Cas9 ribonucleoproteins (RNPs). However, the presence of lentiviral genome encapsulation and replication proteins in EDVs has obscured the underlying delivery mechanism and precluded particle optimization. Here, we show that Cas9 RNP nuclear delivery is independent of the native lentiviral capsid structure. Instead, EDV-mediated genom

Human Ovarian Surface Epithelium Organoids as a Platform to Study Tissue Regeneration.

The ovarian surface epithelium (OSE), the outermost layer of the ovary, undergoes rupture during each ovulation and plays a crucial role in ovarian wound healing while restoring ovarian integrity. Additionally, the OSE may serve as the source of epithelial ovarian cancers. Although the OSE regenerative properties have been well studied in mice, understanding the precise mechanism of tissue repair in the human ovary remains hampered by limited access to human ovaries and suitable in vitro culture protocols. Tissue-specific organoids, miniaturized in vitro models replicating both structural and

Congenital chromoanagenesis in the routine postnatal chromosomal microarray analyses.

Chromosomal microarray analyses (CMA) have greatly increased both the yield and diagnostic accuracy of postnatal analysis; it has been used as a first-tier cytogenetic test in patients with intellectual disability, autism spectrum disorder, and multiple congenital abnormalities. During the last 15 years, we performed CMA in approximately 8,000 patients with neurodevelopmental and/or congenital disorders, of which 13 (0.16%) genetically catastrophic complex chromosomal rearrangements were identified. These ultrarare rearrangements showed clustering of breakpoints, characteristic of chromoanagen

An additively manufactured locking fixation system for potential application in patient-specific implants.

This study introduces a novel technique to implement a locking hole system into AM patient-specific implants without the need of additional post-processing steps such as mechanical machining. This has the potential to decrease the time and cost of manufacturing these implants, providing surgeons with an additional option, that is better suited in cases where the underlying bone is already weakened or bone porosis is an inherent risk. A commercially available locking system was chosen and replicated using high-resolution X-ray CT. A biocompatible material, 316L stainless steel was used to print

Contribution of Resident Stem Cells to Liver and Biliary Tree Regeneration in Human Diseases.

Two distinct stem/progenitor cell populations of biliary origin have been identified in the adult liver and biliary tree. Hepatic Stem/progenitor Cells (HpSCs) are bipotent progenitor cells located within the canals of Hering and can be differentiated into mature hepatocytes and cholangiocytes; Biliary Tree Stem/progenitor Cells (BTSCs) are multipotent stem cells located within the peribiliary glands of large intrahepatic and extrahepatic bile ducts and able to differentiate into hepatic and pancreatic lineages. HpSCs and BTSCs are endowed in a specialized niche constituted by supporting cells

Diminished medial prefrontal cortex activation during the recollection of stressful events is an acquired characteristic of PTSD.

BACKGROUND: Previous research has shown relatively diminished medial prefrontal cortex activation and heightened psychophysiological responses during the recollection of personal events in post-traumatic stress disorder (PTSD), but the origin of these abnormalities is unknown. Twin studies provide the opportunity to determine whether such abnormalities reflect familial vulnerabilities, result from trauma exposure, or are acquired characteristics of PTSD. METHODS: In this case-control twin study, 26 male identical twin pairs (12 PTSD; 14 non-PTSD) discordant for PTSD and combat exposure recall

Validating the gambling functional assessment--revised in a United kingdom sample.

The gambling functional assessment-revised (GFA-R) was designed to assess whether the respondent's gambling was maintained by positive reinforcement or escape. The present study attempted to validate the GFA-R's psychometric properties using United Kingdom (UK) university students and to compare the results to those from a sample of American university students. Two hundred seventy four UK students completed the GFA-R, and 153 also completed the South Oaks gambling screen (SOGS). Two hundred one United States (US) university students completed both measures. A confirmatory factor analysis indi

Pancreatic duct ligation after almost complete β-cell loss: exocrine regeneration but no evidence of β-cell regeneration.

There has been great interest in the extent of β-cell regeneration after pancreatic duct ligation (PDL) and whether α- to β-cell conversion might account for β-cell regeneration after near-complete β-cell loss. To assess these questions, we established a PDL-model in adult male rats after almost complete beta-cell depletion achieved by giving a single high dose of streptozocin (STZ) in the fasted state. Because of the resultant severe diabetes, rats were given islet cell transplants to allow long-term follow-up. Although animals were followed up to 10 months, there was no meaningful β-cell reg

From prion diseases to prion-like propagation mechanisms of neurodegenerative diseases.

Prion diseases are fatal neurodegenerative sporadic, inherited, or acquired disorders. In humans, Creutzfeldt-Jakob disease is the most studied prion disease. In animals, the most frequent prion diseases are scrapie in sheep and goat, bovine spongiform encephalopathy in cattle, and the emerging chronic wasting disease in wild and captive deer in North America. The hallmark of prion diseases is the deposition in the brain of PrP(Sc), an abnormal β -sheet-rich form of the cellular prion protein (PrP(C)) (Prusiner 1982). According to the prion hypothesis, PrP(Sc) can trigger the autocatalytic con

Synergistic parasite-pathogen interactions mediated by host immunity can drive the collapse of honeybee colonies.

The health of the honeybee and, indirectly, global crop production are threatened by several biotic and abiotic factors, which play a poorly defined role in the induction of widespread colony losses. Recent descriptive studies suggest that colony losses are often related to the interaction between pathogens and other stress factors, including parasites. Through an integrated analysis of the population and molecular changes associated with the collapse of honeybee colonies infested by the parasitic mite Varroa destructor, we show that this parasite can de-stabilise the within-host dynamics of D

Ligandless dispersive liquid-liquid microextraction for the separation of trace amounts of silver ions in water samples and flame atomic absorption spectrometry determination.

In this article, a new ligandless dispersive liquid-liquid microextraction method has been developed for preconcentration of trace quantities of silver as a prior step to its determination by flame atomic absorption spectrometry. In the proposed approach, carbon tetrachloride and ethanol were used as extraction and dispersive solvents. Several factors that may be affected on the extraction process, like, extraction solvent, disperser solvent, the volume of extraction and disperser solvent, pH of the aqueous solution and extraction time were optimized. Under the optimal conditions, the calibrat
