Dynamic contrast enhanced MRI in prostate cancer.

Angiogenesis is an integral part of benign prostatic hyperplasia (BPH), is associated with prostatic intraepithelial neoplasia (PIN) and is key to the growth and for metastasis of prostate cancer. Dynamic contrast enhanced magnetic resonance imaging (DCE-MRI) using small molecular weight gadolinium chelates enables non-invasive imaging characterization of tissue vascularity. Depending on the technique used, data reflecting tissue perfusion, microvessel permeability surface area product, and extracellular leakage space can be obtained. Two dynamic MRI techniques (T2*-weighted or susceptibility

Inhibition of human nasopharyngeal carcinoma growth and metastasis in mice by adenovirus-associated virus-mediated expression of human endostatin.

Nasopharyngeal carcinoma (NPC) is a highly malignant and frequently metastasized tumor. Endostatin has been shown to inhibit NPC growth, but its efficacy against NPC metastasis has not been shown in vivo. Here, we established a NPC metastasis model in mice by transplanting EBV-positive NPC cells, C666-1, in the livers of nude mice and observed lung metastasis. Furthermore, we showed that tail vein injection of recombinant adeno-associated virus encoding human endostatin (rAAV-hEndo) significantly prolonged the median survival rate of NPC metastasis-bearing mice (from 22 to 37 days, P < 0.01).

The transport barrier in intraperitoneal therapy.

The peritoneal cavity is important in clinical medicine because of its use as a portal of entry for drugs utilized in regional chemotherapy and as a means of dialysis for anephric patients. The barrier between the therapeutic solution in the cavity and the plasma does not correspond to the classic semipermeable membrane but instead is a complex structure of cells, extracellular matrix, and blood microvessels in the surrounding tissue. New research on the nature of the capillary barrier and on the orderly array of extracellular matrix molecules has provided insights into the physiological basis…

Smooth muscle cells and the pathogenesis of cerebral microvascular disease ("angiomyopathies").

Many forms of human cerebral microvascular disease result from abnormal proliferation and/or degeneration of smooth muscle cells (SMC) in the vessel wall of arteries and arterioles. Human cerebral microvessel-derived smooth muscle cells (MV-SMC) in culture can be used to study the pathogenesis of microvascular disease. Primary cultures were established from nonneoplastic human brain specimens surgically resected and characterized as to their growth properties and phenotype. The cultures have been used to study various factors that may be relevant in the pathogenesis of microangiopathies, in pa

Protein and angiogenic dose-response expression of phVEGF-A(165) gene in rat myocardium.

Therapeutic myocardial angiogenesis by means of transient overexpression of angiogenic growth factors is a potential treatment modality for severe ischemic heart disease. This study was undertaken in the rat to examine effects of phVEGF-A(165) myocardial transfection in terms of dose-response as regards the number of hVEGF-A expressing cells on one hand and on the other angiogenesis. Non-surgical echocardiography-guided intramyocardial injection of phVEGF-A(165) was done into normoxic or hypoxic (10% O(2)) rats. Cardiomyocytes expressing VEGF-A protein, capillary morphology and density were de

Role of leukocyte-endothelium adhesion in affecting recovery from ischemic episodes.

The role of the inflammatory process in the onset of sickle cell crisis has not been fully elucidated, although there is evidence for leukocytosis and enhanced leukocyte to endothelium adhesion. The elevated LEA typical of inflammation may impede recovery from transient ischemic episodes by increasing the resistance to blood flow, which in the case of sickle cell disease may exacerbate the "vicious cycle" of HbS deoxygenation which leads to microvascular stasis. Recent studies on the mechanics of white blood cell margination and adhesion in postcapillary venules of laboratory animals, either i

Cerebrovascular phenotype analysis in Gucy1a3 loss-of-function mice: insights into moyamoya disease susceptibility.

BACKGROUND: Moyamoya disease (MMD) is characterized by progressive stenosis of the internal carotid artery and compensatory moyamoya angioge … MD. Instead, it induced a unique cerebrovascular phenotype marked by "rarefaction" of both cortical microvessels and leptomeningeal networks. Whether large‑vessel pathology develops in aged mice remains to be determined. These findings suggest that GUCY1A3 loss‑of‑function may contribute to MMD susceptibility primarily by compromising small‑vessel integrity, n …

TET2-driven demethylation of ANG promotes angiogenesis and malignant transformation in oligodendroglioma.

Oligodendrogliomas (OG) are indolent yet inevitably progressive gliomas in which angiogenesis is tightly linked to malignant transformation, but the underlying epigenetic mechanisms remain unclear. By integrating public datasets of primary and malignant transformed OG specimens, patient-derived cell lines and orthotopic xenograft models, we identify angiogenin (ANG) as a key pro-angiogenic regulator whose high expression correlates with increased microvessel density, enhanced proliferation and poor overall survival. Promoter methylation analyses reveal that ANG is heavily methylated in low-gra

Dynamic characterization of vascular response and treatment in oral traumatic ulcer in mice via photoacoustic imaging.

BACKGROUND: Dynamic surveillance of vasculature is essential for evaluating the healing of oral ulcer. Existing techniques used in vascular imaging face limitations, such as inadequate spatial resolution, restricted diagnostic depth, and the necessity of exogenous contrast agents. Therefore, this study aimed to use robust photoacoustic imaging (PAI) for the dynamic monitoring of vascular response during healing and the associated treatment process of oral ulcer. METHODS: Kunming mice (male, 8 weeks old, 31-41 g) were treated with 50% acetic acid for 90 s on the tongue mucosa for induction of

Chronic Intermittent Hypobaric Hypoxia Decreases High Blood Pressure by Stabilizing the Vascular Renin-Angiotensin System in Spontaneously Hypertensive Rats.

BACKGROUND AND AIMS: Previous studies have demonstrated the anti-hypertensive effect of chronic intermittent hypobaric hypoxia (CIHH) in hypertensive rats. The present study investigated the anti-hypertensive effect of CIHH in spontaneously hypertensive rats (SHR) and the role of the renin-angiotensin system (RAS) in anti-hypertensive effect of CIHH. METHODS: Fifteen-week-old male SHR and WKY rats were divided into four groups: the SHR without CIHH treatment (SHR-CON), the SHR with CIHH treatment (SHR-CIHH), the WKY without CIHH treatment (WKY-CON), and the WKY with CIHH treatment (WKY-CIHH)

Exosomes secreted by human-induced pluripotent stem cell-derived mesenchymal stem cells attenuate limb ischemia by promoting angiogenesis in mice.

INTRODUCTION: 'Patient-specific' induced pluripotent stem cells (iPSCs) are attractive because they can generate abundant cells without the risk of immune rejection for cell therapy. Studies have shown that iPSC-derived mesenchymal stem cells (iMSCs) possess powerful proliferation, differentiation, and therapeutic effects. Recently, most studies indicate that stem cells exert their therapeutic effect mainly through a paracrine mechanism other than transdifferentiation, and exosomes have emerged as an important paracrine factor for stem cells to reprogram injured cells. The objective of this st

Testing the angiogenic potential of cerebrovascular malformations by use of a rat cornea model: usefulness and novel assessment of changes over time.

OBJECTIVE: This experimental study assessed the usefulness of a rat model of corneal angiogenesis for assessing the angiogenic activity of cerebrovascular malformations (CVMs) over time. It is the first investigation to have focused on dynamic quantification of angiogenesis related to CVMs (i.e., assessing changes over time as opposed to one point in time) and the first to have compared different CVMs and other tissues in this way. METHODS: The CVM specimens were from 15 randomly selected arteriovenous malformation (AVM) cases, 15 randomly selected cavernous malformation cases, and 2 venous a

Characterization of microvascular remodeling in metastatic sentinel lymph nodes via skeletonization framework generated by ultrasound localization microscopy.

Lymphatic spread to the sentinel lymph node (SLN) represents the primary pathway for early-stage breast cancer metastasis. Given that metastatic progression is fundamentally driven by pathological angiogenesis, high-resolution imaging and quantification of SLN microvasculature are critical for assessing tumor spread. In this study, we applied ultrasound localization microscopy (ULM) to a rabbit VX2 breast tumor model and developed a skeletonization-based analytical framework to quantify lymphatic microvascular morphology. This approach extracts vascular centerlines from grayscale ULM images to

Transferrin-Functionalized Liposomes Enhance MAPT-ASO Transport Across a 3D Blood-Brain Barrier Microvascular Network Model.

Tau pathology is a defining hallmark of Alzheimer's disease (AD), closely associated with cognitive decline. Antisense oligonucleotides targeting the tau-encoding gene MAPT (MAPT-ASO) have shown promise in clinical trials, but their therapeutic potential is limited by poor delivery across the blood-brain barrier (BBB). In this study, we developed transferrin (TF)-functionalized liposomes encapsulating MAPT-ASOs and evaluated their transport across a 3D self-assembled microvascular BBB model composed of human brain microvascular endothelial cells, astrocytes, and pericytes embedded in a fibrin

Type-1-to-type-2 transition of brain microvascular pericytes induced by cytokines and disease-associated proteins: Role in neuroinflammation and blood-brain barrier disruption.

While the concept of pericyte heterogeneity in the brain microvasculature is becoming more widely accepted, little is known about how they arise, or their functional contributions to the blood-brain barrier (BBB). We therefore set out to examine the distribution of subtypes of pericytes at the BBB and sought to elucidate some of their functional characteristics by examining their unique mRNA expression patterns. We demonstrate that type-1 pericytes (PC1) that are associated with young healthy brains and BBB homeostasis, can transition into type-2 pericytes (PC2) that are associated with diseas
