Biological and physiological changes of Biomphalaria glabrata infected by Heterorhabditis bacteriophora.

The present study investigated for the first time the influence of exposure time (24 or 48 h) of Biomphalaria glabrata (Gastropoda: Pulmonata) to Heterorhabditis bacteriophora (Rhabditida: Heterorhabditidae), strain HP88, on some physiological and reproductive parameters of the host mollusk. Throughout the experiment, intense glycogenolysis was observed in both exposed groups, which was more accentuated in the digestive gland. This change was accompanied by a significant increase in the free glucose content in the exposed snails, indicating that H. bacteriophora infection induces the breakdown…

[Hormonal regulation of carbohydrate metabolism in the liver in plague intoxication].

It was shown that "mouse" toxin of Yersinia pestis injected into the rat tail vein (LD100) caused a 2-fold decrease in the glycogen content … control. These data suggest that plague intoxication has no effect on cAMP-dependent regulation of glycogenolysis and gluconeogenesis in the liver. The action mechanism of toxin(s) on carbohydrate metabolism in also unrelated to the decrease in Bmax for alpha 1 adrenergic receptors in liver membranes, whose number increased 1.3 and 1.5 times after 1 and 2 hours …

[Energy metabolism in the perinatal period (author's transl)].

Neonatal hypoglycemia is of frequent occurrence in fasted newborn babies or animals but the origin of this hypoglycemia is not fully understood. Studies performed in newborn rats have shown that liver glycogenolysis and gluconeogenesis occur immediately after birth and that the increase in the activities of key regulatory enzymes (phosphorylase, glycogen synthetase and phosphoenolpyruvate carboxykinase) results probably from the rise of plasma glucagon and the fall of plasma insulin induced by the "stress" of birth. When the liver glycogen stores have been exhausted, i.e. between 6 and 16 hour…

Branched-Chain Amino Acid Catabolism and Cardiopulmonary Function Following Acute Maximal Exercise Testing in Adolescents.

Background: To provide energy for cardiopulmonary function and maintenance of blood glucose, acute aerobic exercise induces lipolysis, fatty acid oxidation (FAO), glycolysis, and glycogenolysis/gluconeogenesis. These adaptations are mediated by increases in cortisol, growth hormone (GH), and catecholamines and facilitated by a decline in insulin. Branched-chain amino acids (BCAA) also undergo catabolism during intense exercise. Here, we investigated the relationship between BCAA catabolism and metrics of cardiopulmonary function in healthy, well-developed, mature adolescent athletes undergoing…

The role of glucose-6-phosphatase activity in glucose homeostasis and its potential for diabetes therapy.

Glucose-6-phosphatase catalytic subunit (G6PC)1 and G6PC2 are crucial for glucose metabolism, regulating processes like glycolysis, gluconeogenesis, and glycogenolysis. Despite their structural and functional similarities, G6PC1 and G6PC2 exhibit distinct tissue-specific expression patterns, G6P hydrolysis kinetics, and physiological functions. This review provides a comprehensive overview of their enzymology and distinct roles in glucose homeostasis. We examine how inactivating mutations in G6PC1 lead to glycogen storage disease, and how elevated G6PC1 and G6PC2 expression can affect the inci…

Continuous glucose monitoring in children with glycogen storage disease type I.

Glycogen storage disease type I (GSD I) is characterized by impaired production of glucose from glycogenolysis and gluconeogenesis resulting in severe fasting hypoglycaemia. The aim of the present study was to examine the efficacy of a continuous subcutaneous glucose monitoring system (CGMS MiniMed), to determine the magnitude and significance of hypoglycaemia in GSD I and to evaluate the efficacy of its dietary treatment. Four children with GSD I were studied over a 72-h period. Results indicated that the values recorded with continuous subcutaneous glucose monitoring were highly correlated w…

Normal activities of AMP-deaminase and adenylate kinase in patients with McArdle disease.

During physical activity in McArdle patients, little or no lactate is released in the skeletal muscle. However, excessive ammonia production … f PGI was significantly higher in patients than in controls suggesting compensation of the impaired glycogenolysis in McArdle. The ratios of activities of AMPD and AK over PGI were significantly lower in patients than in controls. High ammonia production in McArdle patients is not based on enzyme induction of AMPD and AK but possibly due to kinetic activation of …

Mutations in LPIN1 cause recurrent acute myoglobinuria in childhood.

Recurrent episodes of life-threatening myoglobinuria in childhood are caused by inborn errors of glycogenolysis, mitochondrial fatty acid beta-oxidation, and oxidative phosphorylation. Nonetheless, approximately half of the patients do not suffer from a defect in any of these pathways. Using homozygosity mapping, we identified six deleterious mutations in the LPIN1 gene in patients who presented at 2-7 years of age with recurrent, massive rhabdomyolysis. The LPIN1 gene encodes the muscle-specific phosphatidic acid phosphatase, a key enzyme in triglyceride and membrane phospholipid biosynthesis…

The rise of blood sugar as an additional parameter in traumatic shock.

Besides the known cardiovascular effects of hemorrhagic shock, regular metabolic consequences can be demonstrated in the rabbit: 1. The rise in blood sugar in hemorrhagic shock increases with the amount of blood withdrawn per unit of time. 2. The characteristic rise in blood sugar lasts longer than the phase of the partial exsanguination. 3. In traumatic shock, the rise in blood sugar indicates the degree of severity of the injury earlier than the fall in blood pressure does. The rise in blood sugar is attributable to a reflex increase in glycogenolysis due to catecholamine secretion elicited …

Hepatic phosphorylase deficiency. Its differentiation from other hepatic glycogenoses.

Two brothers with glycogen storage disease of the liver are reported. The clinical symptoms were hepatomegaly and a slight muscle hypotonia; hypoglycaemic symptoms seldom occurred. There was deficient activity of phosphorylase and normal phosphorylase b kinase activity in the leucocytes and in the liver. Three aspects of carbohydrate metabolism were investigated. Glycogenolysis was studied by glucagon tests, either performed after an overnight fast or postprandially; the ensuing glucose curves were flat or almost flat. Glycolysis was investigated by oral tolerance tests with glucose, galactose…

The Role of Circulating Amino Acids in the Hypothalamic Regulation of Liver Glucose Metabolism.

A pandemic of diabetes and obesity has been developing worldwide in close association with excessive nutrient intake and a sedentary lifesty … P-sensitive potassium channels. The liver then responds by lowering the rate of gluconeogenesis and glycogenolysis, ultimately leading to a net decrease in glucose production and in concentrations of circulating glucose. REVISITING GLUCAGON ACTION IN DIABETES: IS IT ALL BAD?

Traditionally, the islet hormone glucagon has been considered as a counterbalance to insulin, preventing hypoglycemia by promoting glucose release from the liver. This model is compatible both with clinical studies demonstrating that one of the initial endocrine responses to insulin-induced hypoglycemia is a rise in glucagon and with in vitro work demonstrating that glucagon signaling activates glycogenolysis in hepatocytes. This model has been extended to implicate glucagon in diabetogenesis, positing that the increased secretion of glucagon acts as a primary driver of hyperglycemia. However,…

Brain glycogen serves as a critical glucosamine cache required for protein glycosylation.

Glycosylation defects are a hallmark of many nervous system diseases. However, the molecular and metabolic basis for this pathology is not fully understood. In this study, we found that N-linked protein glycosylation in the brain is metabolically channeled to glucosamine metabolism through glycogenolysis. We discovered that glucosamine is an abundant constituent of brain glycogen, which functions as a glucosamine reservoir for multiple glycoconjugates. We demonstrated the enzymatic incorporation of glucosamine into glycogen by glycogen synthase, and the release by glycogen phosphorylase by bio…

Early-onset obesity and food restriction alter hepatocyte metabolism in adult Wistar rats.

CONTEXT: Caloric restriction (CR) is suggested for overweight control. OBJECTIVE: Systemic and liver glucose metabolism in the reduced-litter (RL) rat model under 30% CR was investigated. MATERIALS AND METHODS: Newborn litters were organised in control (G9); RL with free diet (G3L); and RL with CR (G3R). Assessments were made at the age of 90 d. RESULTS: Higher liver glycogen content and changes in systemic glucose handling were found in the RL groups. Hepatocyte glucose metabolism was similar in groups G9 and G3L, but basal glucose production and glycogenolysis were higher, while gluconeog…
