Effects of some antiallergic agents on homologous passive cutaneous anaphylaxis (PCA) in mouse ear were investigated by means of assessing the amount of extravasated dye. Antihistamines (chlorpheniramine and diphenhydramine) and antiserotonins (methysergide and cyproheptadine) suppressed mouse ear PCA significantly. The effects of eugenol, a major component of clove, on anaphylaxis were evaluated in rats. Eugenol inhibited compound 48/80-induced systemic anaphylaxis 100% with a dose of 10 micrograms g-1 body weight (BW). The immunologic effects of ampicillin polymers were studied in mice and rabbits. Polymerized ampicillin--in contrast to monomers--had a suppressive effect on the antibody response to penicilloylated proteins. Unfractionated ampicillin polymer, and high molecular weight fractions were found to have an immunogenic effect in mice, when tested by a haemolytic plaque assay. The immunogenic effect of variously sized polymers did not correspond to their antigenic activity as reflected by passive cutaneous anaphylaxis experiments.

Concentrated aqueous solutions of some β-lactam antibiotics were stored for 14 days. Samples were then subjected to gel filtration and fractions in the high molecular weight regions of the eluate were collected and lyophilized. The yields of materials from solutions of various β-lactam antibiotics were compared and the antigenicity of the materials was assessed by their ability to react with antibodies of benzylpenicilloyl specificity raised in rabbits and tested in guinea-pigs by passive cutaneous anaphylaxis.

We have been able to produce a mouse monoclonal IgE antibody specific to an adult worm antigen extracted from Schistosoma japonicum (Sj). The antibody was able to elicit passive cutaneous anaphylaxis in the rat skin against Sj with the highest titer of 1:256,000 but did not cross-react with S. mansoni antigen. Guinea pigs were sensitized either by intranasal dusting with ragweed (RW) pollen or by intranasal instillation of RW extract and subsequently challenged with a RW extract aerosol. It was relatively easy to determine the microshock time for animals which responded between 0 and 10 minutes. Sera from these sensitive guinea pigs were tittered by passive cutaneous anaphylaxis tests. Data from these two assays showed a semilogarithmic relationship with a correlation coefficient of -0.934.
