MicroRNAs in cerebral ischemia-induced neurogenesis.

Cerebral ischemia induces neurogenesis, including proliferation and differentiation of neural progenitor cells and migration of newly generated neuroblasts. MicroRNAs (miRNAs) are small noncoding RNAs that decrease gene expression through mRNA destabilization and/or translational repression. Emerging data indicate that miRNAs have a role in mediating processes of proliferation and differentiation of adult neural progenitor cells. This article reviews recent findings on miRNA profile changes in neural progenitor cells after cerebral infarction and the contributions of miRNAs to their ischemia-i

Chronic Intermittent Hypoxia-Induced Neural Injury: Pathophysiology, Neurodegenerative Implications, and Therapeutic Insights.

Obstructive sleep apnea-hypopnea syndrome (OSAHS) is a sleep-related respiratory disorder that poses a global threat to human health. Chronic intermittent hypoxia (CIH) is its main pathological feature. With the advancements in medical research, the study of CIH-induced neural injury has gained increasing attention. Studies have shown that CIH can lead to or aggravate neuroinflammation and apoptosis by increasing blood-brain barrier (BBB) permeability, promoting oxidative stress, activating glial cells, and triggering multiple signaling pathways, ultimately resulting in neural injury. These pr

Effect of bladder outlet obstruction on micturition reflex pathways in the rat.

The effect of outlet obstruction on neural pathways to the bladder was examined six weeks after urethral ligation in rats. Ligated rats exhibited a seven-fold increase in bladder weight, a six-fold increase in volume, and significant increases (p less than 0.001) in micturition pressure, residual volume and compliance measured under urethane anesthesia. Unanesthetized ligated rats in comparison to control rats voided smaller volumes of urine more frequently (0.7 vs 1.1 voids/hr., p less than 0.001). Electrophysiological examination of vesical afferent and efferent peripheral neural pathways in…

Short-term transcriptomic changes in the mouse neural tube induced by an acute alcohol exposure.

Alcohol exposure during the formation and closure of the neural tube, or neurulation (embryonic day [E] 8-10 in mice; ∼4th week of human pregnancy), perturbs development of midline brain structures and significantly disrupts gene expression in the rostroventral neural tube (RVNT). Previously, alcohol exposure during neurulation was found to alter gene pathways related to cell proliferation, p53 signaling, ribosome biogenesis, immune signaling, organogenesis, and cell migration 6 or 24 h after administration. Our current study expands upon this work by investigating short-term gene expression c

Mechanosensory feedback circuits for copulation control in Drosophila males.

Successful copulation requires the precise integration of sensory cues and motor actions. Here, we identify three classes of genital mechanosensory neurons in male Drosophila that are required for copulation. These neurons exhibit distinct tactile response properties and act through separate pathways that selectively recruit male-specific motor neurons. Functionally, these pathways differentially contribute to copulation initiation, maintenance, and termination. Together, our findings define neural pathways through which genital mechanosensory feedback is transformed into motor output, thus pr…

A systematic analysis of the joint effects of ganglion cells, lagged LGN cells, and intercortical inhibition on spatiotemporal processing and direction selectivity.

Simple cells in the visual cortex process spatial as well as temporal information of the visual stream and enable the perception of motion information. Previous work suggests different mechanisms associated with direction selectivity, such as a temporal offset in thalamocortical input stream through lagged and non-lagged cells of the lateral geniculate nucleus (LGN), or solely from intercortical inhibition, or through a baseline selectivity provided by the thalamocortical connection tuned by intercortical inhibition. While there exists a large corpus of models for spatiotemporal receptive fiel

Reciprocal Inhibitory Glomerular Circuits Contribute to Excitation-Inhibition Balance in the Mouse Olfactory Bulb.

The major inhibitory interneurons in olfactory bulb (OB) glomeruli are periglomerular cells (PGCs) and short axon cells (SACs). PGCs and SACs provide feedforward inhibition to all classes of projection neurons, but inhibition between PGCs and SACs is not well understood. We crossed Cre and GFP transgenic mice and used virally-delivered optogenetic constructs to selectively activate either SACs or GAD65cre-ChR2-positive PGCs while recording from identified GAD65cre-ChR2-positive PGCs or SACs, respectively, to investigate inhibitory interactions between these two interneuron types. We show that

Training based on mirror visual feedback influences transcallosal communication.

Mirror visual feedback (MVF) therapy has been demonstrated to be successful in neurorehabilitation, probably inducing neuroplasticity changes in the primary motor cortex (M1). However, it is not known whether MVF training influences the hemispheric balance between the M1s. This topic is of extreme relevance when MVF training is applied to stroke rehabilitation, as the competitive interaction between the two hemispheres induces abnormal interhemispheric inhibition (IHI) that weakens motor function in stroke patients. In the present study, we evaluated, in a group of healthy subjects, the effect

Model calcium sensors for network homeostasis: sensor and readout parameter analysis from a database of model neuronal networks.

In activity-dependent homeostatic regulation (ADHR) of neuronal and network properties, the intracellular Ca(2+) concentration is a good candidate for sensing activity levels because it is correlated with the electrical activity of the cell. Previous ADHR models, developed with abstract activity sensors for model pyloric neurons and networks of the crustacean stomatogastric ganglion, showed that functional activity can be maintained by a regulation mechanism that senses activity levels solely from Ca(2+). At the same time, several intracellular pathways have been discovered for Ca(2+)-dependen

Effects of electrical stimulation of the striatum on bladder activity in cats.

AIMS: Parkinson's disease (PD) affects the nigrostriatal projections leading to micturition disturbance in most cases. Overactive bladder (OAB) symptoms such as urinary urgency or urgent urinary incontinence are common amongst PD patients. Several urodynamic studies have revealed that detrusor overactivity causes OAB symptoms in PD patients. We assert that striatal dysfunction might contribute to the pathogenesis of detrusor overactivity in PD patients. However, the role of the striatum in bladder contraction remains unclear. METHODS: We generated spontaneous isovolumetric bladder contraction

Pedophilia: neuropsychological evidence encouraging a brain network perspective.

Although the vast majority of current pathogenetic theories support a neurobiological understanding of psychiatric disorders, the brain functional correlates of pedophilia are largely unknown. Based on prior behavior genetics research on human sexual orientation and phenomenology as well as the phenotypical intersection of pedophilia with other psychiatric spectrum disorders, we hypothesize the involvement of striato-thalamo-cortical processing loops in the formation of pedophilic urges and behaviors. Data from a current neuropsychological pilot study in four pedophiles encourage our brain fun

Gestural recovery and the role of forward and reversed syllabic repetitions as stuttering inhibitors in adults.

Instead of being the core stuttering 'problem', syllabic repetitions may be a biological mechanism, or 'solution', to the central involuntary stuttering block. Simply put, stuttering is an endogenous transitory state of 'shadowed speech', a choral speech derivative that allows for a neural release of the central block. To investigate this possibility, 14 adults who stutter read while listening to forward fluent speech, reversed fluent speech, forward stuttered speech, and reversed stuttered speech. All conditions induced significant degrees of stuttering inhibition when compared to a control c

Circadian rhythm of melatonin release in pineal gland culture: arg-vasopressin inhibits melatonin release.

The mammalian pineal gland is known to receive a noradrenergic sympathetic efferent signal from the suprachiasmatic nucleus (SCN) via the superior cervical ganglion. Arg-vasopressin (AVP) containing neurons in the SCN is one of the output paths of circadian information to the other brain areas. AVP release from the SCN is suppressed by melatonin. In turn, we determined the direct effect of AVP on melatonin release using pineal gland explant culture. AVP (1 microM) suppressed melatonin release. Noradrenaline stimulated melatonin release was attenuated by AVP. In turn, the expression of the mela

Dorsal root lesions block the expression of morphine withdrawal elicited from the rat spinal cord.

Evidence has been accumulating to indicate that the spinal cord plays an important role in the expression of several narcotic abstinence signs in the intact animal. One characteristic and reproducible withdrawal sign which can be elicited in both intact and spinal-transected dependent rats is the naloxone-induced increase in arterial blood pressure. Employing this model, this autonomic component of narcotic withdrawal was quantitated in dependent spinal (C1)-transected rats with intact dorsal roots and in those with surgical lesions of the dorsal roots from T3 to L4. The withdrawal-associated

Clinical features of status epilepticus in patients with HIV infection.

Loss of insulin-producing pancreatic islet β-cells is a hallmark of type 1 diabetes. Several experimental paradigms demonstrate that these cells can, in principle, be regenerated from multiple endogenous sources using signaling pathways that are also used during pancreas development. A thorough understanding of these pathways will provide improved opportunities for therapeutic intervention. It is now appreciated that signaling pathways should not be seen as "on" or "off" but that the degree of activity may result in wildly different cellular outcomes. In addition to the degree of operation of

Neural mechanisms underlying synchronization of movement to musical cues in Parkinson disease and aging.

INTRODUCTION: External and internal musical cues provide therapeutic techniques for gait rehabilitation in aging and neurological disorders. For people with Parkinson disease (PwPD), mental singing is a type of internal cue that can regularize gait timing. No studies to date have directly measured brain activity during external and internal musical cues as used in gait rehabilitation. Evidence suggests the neural mechanisms of external vs. internal cued movement differ. External cues are thought to drive movement via recruitment of cerebello-thalamo-cortical (CTC) pathways, while internal cues

Chemogenetic inhibition of lateral habenula projections to the dorsal raphe nucleus reduces passive coping and perseverative reward seeking in rats.

The lateral habenula (LHb) processes information about aversive experiences that contributes to the symptoms of stress disorders. Previously, we found that chemogenetic inhibition of rat LHb neurons reduced immobility in the forced swim test, but the downstream target of these neurons was not known. Using an intersectional viral vector strategy, we selectively transduced three different output pathways from the LHb by injecting AAV8-DIO-hM4Di into the LHb and CAV2-CRE (a retrograde viral vector) into one of the three target areas as follows: dorsal raphe nucleus (DRN), ventral tegmental area (

[Baroreflex sensitivity and chronic pain : Pathogenetic significance and clinical implications].

The interaction of cardiovascular dynamics and pain perception is an important component of intrinsic pain regulation. In healthy subjects acute pain stimuli cause increased sympathetic arousal and increased mean arterial pressure. Arterial baroreceptors sense phasic blood pressure changes and relay the information to the lower brainstem via the dorsomedial nucleus tractus solitarius (dmNTS). Projections in the brainstem and also higher cortical areas result in elevation of blood pressure as part of the autonomic nervous system as well as modulation of sleep, anxiety and pain. In healthy subje

Visualization of disconnection syndromes in humans.

Knowledge of the relationship between structure and function is essential to the exploration of the architecture of cognition. Cognitive processes require the coordinated activity of large-scale brain networks consisting of distant cortical regions, connected by long-range white matter tracts. Despite decades of connectional tracing studies in monkeys, the backwardness of human anatomy makes it difficult to draw conclusions from lesion studies and functional neuroimaging when brain connectivity is at issue. We propose an approach to clinico-anatomical correlation, based on a standardized atlas
