Hemodynamic, hormonal, and renal effects of intracerebroventricular adrenomedullin in conscious sheep.

Adrenomedullin, the recently described vasodilator that exhibits potent hypotensive actions when administered systemically, is also found in the central nervous system, suggesting a role for adrenomedullin as a neurohormone. However, only a limited number of studies have examined the central effects of adrenomedullin. Therefore, we have examined the integrative hemodynamic, renal, and hormonal effects of intracerebroventricular (I.C.V.) adrenomedullin in conscious sheep. Eight surgically prepared sheep received I.C.V. infusions of adrenomedullin at two doses (2 ng/kg x min followed immediately…

Regulation of uterine immune function by progesterone--lessons from the sheep.

Survival of the fetal allograft results from orchestrated adjustments in activity of maternal lymphoid cells as well as in trophoblast gene expression. One molecule that regulates uterine immune responsiveness is progesterone. In fact, uterine skin graft survival and susceptibility to bacterial infections are increased by progesterone. This review focuses on the role of progesterone in regulation of uterine immune function in the sheep. While the importance of progesterone as a regulator of immune function likely varies between species, concepts derived from the sheep model may prove pertinent…

In vitro effect of glucocorticoids on phagocytic function of sheep alveolar macrophages.

The potentiation of infection is a recognized adverse effect of steroids. This study examined the in vitro effect of glucocorticoids on sheep alveolar macrophage phagocytic function, an essential means of cellular defence in the lungs, by evaluating the phagocytosis of zymosan particles after 30 min of contact between alveolar macrophages and steroids. Hydrocortisone, dexamethasone phosphate and methylprednisolone significantly reduced the phagocytic capability of sheep alveolar macrophages, but the effects of dexamethasone isonicotinate, prednisolone and flumethasone were less marked.

Behavior of mitral allografts in the tricuspid position in the growing sheep model.

BACKGROUND: On the basis of a previous experience in a chronic sheep model in which partial mitral allografts remained viable and properly functioning 12 months after operation, we assessed the results obtained by replacing the tricuspid valve with fresh antibiotic-preserved mitral allografts. METHODS: Twenty 3-month-old sheep with a mean weight of 23.7 +/- 2.3 kg underwent cardiopulmonary bypass and had a fresh antibiotic-preserved mitral allograft implanted in the tricuspid position with the heart beating under normothermic conditions. The tricuspid valve apparatus was not excised. After a …

Tick-borne diseases of sheep and goats and their related vectors in Iran.

Haemoparasitic diseases have long been considered as a major problem to efficient sheep and goats production in Iran. Theileriosis due to Theileria hirci and babesiosis due to Babesia ovis and B. motasi are the most pathogenic protozoa. B. crassa, Anaplasma ovis and Eperythrozoon ovis are usually non-pathogenic and do not cause any apparent problem. The major tick genera found on sheep and goats are Hyalomma, Rhipicephalus, Haemaphysalis, Ixodes and rarely Dermacentor distributed in all part of Iran. Our studies indicate that only Hyalomma ticks transmit Theileria species but the others transm…

Conventional vaccines for tick-borne haemoparasitic diseases of sheep and goats.

Of the many tick-borne haemoparasites of sheep and goats, three may be controlled by vaccination. A live virulent blood vaccine for Cowdria ruminantium infection (heartwater) is used widely in southern Africa in an infection and treatment procedure. A live vaccine for Babesia ovis attenuated by passage in splenectomised sheep has been used extensively in Bulgaria with good results. A live vaccine for Theileria hirci, consisting of schizonts propagated in a lymphoid cell culture and attenuated by passage, has been used successfully in the Middle East.

The effect of experimental fascioliasis on the pharmacokinetics of antipyrine and sulphadimidine in desert sheep.

Healthy adult male desert sheep were experimentally infected with Fasciola gigantica, to investigate the influence of experimental fasciolasis on the pharmacokinetics of antipyrine and sulphadimidine. Each animal received 500 metacercariae orally. The experimental infection was confirmed histologically, by detection of Fasciola eggs in faeces and by measuring the activities of the enzymes sorbitol dehydrogenase (SD), glutamate dehydrogenase (GD) and aspartate aminotransferase (AST) in plasma during the course of the disease. Changes in the pharmacokinetics of antipyrine and sulphadimidine were…

Effect of chronic cocaine exposure on the hemodynamic response to vasopressors in sheep.

BACKGROUND: The purpose of this study was to determine how chronic cocaine exposure affects the hemodynamic response to epinephrine, dopamine, phenylephrine, and ephedrine in awake sheep. METHODS: The hemodynamic response to dopamine (10 microg/kg), phenylephrine (1.5 microg/kg), and ephedrine (0.15 mg/kg) boluses was determined at baseline before low-dosage cocaine exposure and again after 15 and 18 days of cocaine exposure. The hemodynamic response to epinephrine (0.15 microg/kg), phenylephrine (1.5 microg/kg), and ephedrine (0.15 mg/kg) was determined at baseline before high-dosage cocaine…

The effects of allopurinol on the ultrastructure of ischaemic and reperfused large intestine of sheep.

OBJECTIVE: To test the possible inhibitory effect of allopurinol on reperfusion injury, caused by oxygen-derived free radicals, of sheep large intestine. DESIGN: An ultrastructural study on caecal tissues from control and treated groups. ANIMALS: Fifty sheep in four ischaemic and reperfused (treatment) groups and one control group. Three of the treatment groups were subdivided for half to be injected with allopurinol and the other half with its solvent, potassium hydroxide (KOH). PROCEDURE: Ischaemia of the caecum was induced in the four treatment groups for 60 minutes by clamping the apex.…

Contagious caprine pleuropneumonia and other pulmonary mycoplasmoses of sheep and goats.

Contagious caprine pleuropneumonia (CCPP) is now a well-defined disease that is caused by Mycoplasma capricolum subsp. capripneumoniae. CCPP is infectious, contagious and fulfils the classic Koch postulates that characterise such types of disease. The distribution of the disease is not exactly known, but reports of mycoplasma isolation and official declarations to the Office International des Epizooties (OIE) enable a probable distribution map to be obtained. There are many other mycoplasmas that can infect goat and sheep lungs and induce pleuropneumonia. However, pleuropneumonia is often rest…

Localization of chronic Chlamydia psittaci infection in the reproductive tract of sheep.

Twelve sheep experimentally infected with Chlamydia psittaci during pregnancy either aborted or gave birth to weak, low-birth-weight lambs as a result of uteroplacental infection. Subsequently, these ewes excreted chlamydial antigen from their reproductive tracts during estrus. About 1 year after pregnancy failure, immunohistochemistry and polymerase chain reaction were used to examine sections of vagina, uterus, and oviduct for evidence of C. psittaci. Four noninfected control ewes were similarly examined. C. psittaci antigen or DNA was detected in vagina, uterus, and oviduct samples from chr…

Physico-chemical fate of chromium compounds in the sheep lung model.

Transmission electron microscopy (TEM) equipped with energy-dispersive x-ray analysis (EDX), electron spectroscopy for chemical analysis (ESCA), secondary ion mass spectrometry (SIMS), and laser microprobe for mass analysis (LAMMA) were used to follow the fate of chromium compounds deposited in the sheep tracheal lobe. Four chromium compounds were used: two chromium(VI) compounds (lead chromate and chromium trioxide) and two chromium(III) compounds (chronic oxide and chromium sulfate). Chromium trioxide is very soluble and the other three are slightly soluble. The compositions, concentrations,…

Ineffectiveness of calcitonin on a local-disuse osteoporosis in the sheep: a histomorphometric study.

Local immobilization is a good model for studying disuse-induced bone loss and to appreciate the effects of drugs, especially preventive action of antiresorptive therapy. In fact, increased osteoclastic activity is the main point of such a bone loss. The effect of salmon calcitonin was investigated on immobilization-induced osteoporosis in the sheep. Twenty-four nonovariectomized, adult, female, Welsh mountain sheep were submitted, by an external fixator procedure, to hock joint immobilization from the tibia to the the metatarsus for 12 weeks. The sheep were randomized into two groups receivin…

The clinical differentiation of nervous and muscular locomotor disorders of sheep in Australia.

Many of the nervous and muscular locomotor disorders that affect sheep throughout Australia are commonly referred to as "staggers" syndromes. The range of clinical signs displayed by sheep suffering these disorders is sufficiently diverse to enable each syndrome to be graded into one of 5 progressive clinical groups. The first group, the limb paresis syndromes, includes the primary myopathies associated with the ingestion of Ixiolaena brevicompta, Malva parviflora, and Trachymene ochracea, as well as selenium and Vitamin E disorders, Paroo virus staggers, congenital progressive muscular dystro…

Reduction of feed intake in sheep by enterostatin, the procolipase activation peptide.

Enterostatin, a peptide formed during the activation of pancreatic procolipase in the duodenum, is likely a mediator of satiety in the rat. The objective of this research was to determine whether administration of enterostatin into the lateral cerebral ventricle of sheep unfed for 2 h resulted in reduced feed intake. Bolus doses of 2 micrograms did reduce (P < .03) feed intake during the hour after injection by one- to two-thirds of control. Doses 20 micrograms or greater were not effective, and .2 microgram also did not have any statistically significant effect. This range of effective doses …

Changes in the blood-gas status of sheep with experimentally induced heartwater.

The blood-gas status of seven sheep with experimentally induced heartwater during the acute and terminal stages was investigated. Changes in blood gas included a decline in arterial oxygen tension (pO2) combined with a respiratory alkalosis. Although the sheep became hypoxaemic, blood-gas changes associated with respiratory failure were not observed.

The myocardial effects of fenoterol, isoprenaline and salbutamol in normoxic and hypoxic sheep.

Three groups, each of four sheep, were randomly allocated under blinded conditions to receive the beta-adrenoceptor agonists, salbutamol, fenoterol or isoprenaline, in doses of 0.5, 2, 8, 32 and 128 micrograms/kg intravenously at 15-minute intervals. A separate group of four animals received equal volumes of saline. Heart rate was recorded immediately prior to each drug administration and serum potassium was measured. Three to 4 days later the experiment was repeated during induced systemic hypoxia and the animals then necropsied. All the agonists produced significant increases in heart rate. …
