Glucose degradation products (GDP) retard remesothelialization independently of D-glucose concentration.

BACKGROUND: Glucose degradation products (GDP) present in heat-sterilized dialysis fluids are thought to contribute to cellular dysfunction and membrane damage during peritoneal dialysis. To examine the effects of specific GDP on the remesothelialization process, the impact of conventional and low GDP peritoneal dialysis solutions, D-glucose, and individual GDP in a scratch-wounding model was assessed. METHODS: Scratch (0.5 to 0.6 mm)-wounded human peritoneal mesothelial cells (HPMC) were treated, at pH 7.4, with either (1) control medium (M199), (2) laboratory-prepared heat or filter-sterili

[Effects of ethanol on the nervous and vascular systems: the mechanisms of alcohol-induced hypertension].

Many previous experimental and epidemiological studies have shown that alcohol consumption has a positive correlation with the incidence of hypertension. The effects of ethanol on the nervous and vascular systems in relation to the mechanisms of alcohol-induced hypertension proposed so far are reviewed here. Alcohol ingestion influences many pathophysiological functions which regulate blood pressure, as follows: 1) Sympathetic nervous activity is increased after drinking. 2) Ethanol acts directly on the contractility of vascular smooth muscle. Ethanol acutely contracts some arteries and increa

ADH1 promotes Candida albicans pathogenicity by stimulating oxidative phosphorylation.

OBJECTIVE: Alcohol dehydrogenase I is encoded by ADH1 in Candida albicans, and is one of the key enzymes in fungal metabolism by which it catalyzes the conversion from acetaldehyde to ethanol. The role of the associated protein Adh1p, encoded by ADH1 in fungal pathogenicity has not been thoroughly studied despite its near ubiquity in the fungal kingdom. Using C. albicans as a model, this study proposes to determine the possible pathogenic roles for ADH1 and its possible underlying mechanisms. METHODS: The SAT1 flipper strategy was used to construct the ADH1 deletion mutant. Growth curves and

Roles of the genetic polymorphisms of alcohol-metabolizing enzymes on the immunology in high-risk drinkers.

Alcohol metabolism involves several enzymes and the individual genetic variations in the alcohol metabolism are related to the absorption, distribution, and elimination of alcohol and metabolites such as acetaldehyde. Therefore, the genetic variations of alcohol-metabolizing enzymes are responsible for the different toxicity of alcohol in several organs like liver and immunological systems. The purpose of this study was to evaluate if the life styles such as drinking and smoking and the genetic variations of alcohol-metabolizing enzymes (ADH2, ALDH2, CYP2E1, and CAT) were associated with the i

ESR and HPLC-EC analysis of ethanol oxidation to 1-hydroxyethyl radical: rapid reduction and quantification of POBN and PBN nitroxides.

Extensive ESR spin-trapping studies have shown that ethanol is oxidized to 1-hydroxyethyl radical (HER) by rat and deer mice liver microsomal systems. The ESR detection of POBN/HER nitroxide in bile, and formation of antibodies, which recognize HER adducts in alcoholics, suggest that HER is produced in vivo. In liver, where ethanol is primarily metabolized, only traces of PBN/HER nitroxide are documented. One limitation of the ESR spin-trapping technique, however, is that the nitroxides formed in the presence of cellular reductants can be metabolized to the corresponding ESR "silent" hydroxyla

Unexpected properties of NADP-dependent secondary alcohol dehydrogenase (ADH-1) in Trichomonas vaginalis and other microaerophilic parasites.

Our previous observation that NADP-dependent secondary alcohol dehydrogenase (ADH-1) is down-regulated in metronidazole-resistant Trichomonas vaginalis isolates prompted us to further characterise the enzyme. In addition to its canonical enzyme activity as a secondary alcohol dehydrogenase, a pronounced, so far unknown, background NADPH-oxidising activity in absence of any added substrate was observed when the recombinant enzyme or T. vaginalis extract were used. This activity was strongly enhanced at low oxygen concentrations. Unexpectedly, all functions of ADH-1 were efficiently inhibited by

Pesticide poisoning of livestock: a review of cases investigated.

The cases of suspected poisoning of livestock by a range of organic chemicals, mainly pesticides, investigated at the Central Veterinary Laboratory during the period 1977 to 1980 are reviewed. The incidences of poisoning by various types of compound and the species most at risk are discussed. Organophosphorus pesticides and rodenticides were the commonest causes of poisoning. Pesticides accounted for one in 25 of all fatal poisonings listed by the veterinary investigation diagnosis analysis (VIDA) of the Ministry of Agriculture, Fisheries and Food.

The liver: a pharmacologic perspective.

One of the may vital functions of the liver is the biodegradation of foreign substances. The enzyme systems responsible for this liver function are frequently the site of drug interactions, both therapeutic and detrimental. Various substances can alter these enzymes by inducing, inhibiting, or competing with them, thus affecting drug response. In most instances, the liver detoxifies and deactivates chemicals, protecting the body from their harmful effects. In some biotransformation processes, however, toxic metabolites are produced that may be injurious to liver tissue as well as other body or

The Photoionization Spectrum of the Acetyl Radical and the Kinetics of Its Unimolecular and Bimolecular Reactions at Elevated Pressures.

The acetyl radical (CH3CO) is a primary intermediate during the oxidation and combustion of CH3CHO and represents a prototypical example of a weakly bound acyl radical. However, it is challenging to detect and has rarely been probed at elevated pressures, where CH3CO decays by competing bimolecular and unimolecular reactions. The present study provides a combined modeling and experimental analysis of the comprehensive network of CH3CO unimolecular and bimolecular kinetics at 500 kPa and 300-366 K. CH3CO was generated by acetaldehyde (CH3CHO) + Cl reactions, where the Cl is produced by pulsed l

Physiochemical characteristics of date pollen extract-hybrid yoghurt as a functional dairy food modulates hypothyroidic symptoms in modeled rats.

Yoghurt is a widely consumed dairy product across the globe. It possesses healing properties and qualities that are significant for human health. This study aimed to evaluate the properties of date pollen ethanolic extract-hybrid yoghurt (DPEE-HY) on hypothyroidism as a functional dairy food. Standardized cow's milk was hybridized with 1 or 2% of DPEE, while the control yoghurt (CY) was not hybridized. Adult male Wistar albino rats were used to evaluate the pharmaceutical impacts of DPEE-HY on propylthiouracil (PTU)-induced hypothyroidic rats. The rats were randomly assigned to four groups as

Formation of carbonyl compounds during ozonation of lake water and wastewater: Development of a non-target screening method and quantification of target compounds.

Ozonation of natural waters is typically associated with the formation of carbonyl compounds (aldehydes, ketones and ketoacids), a main class of organic disinfection byproducts (DBPs). However, the detection of carbonyl compounds in water and wastewater is challenged by multiple difficulties inherent to their physicochemical properties. A non-target screening method involving the derivatisation of carbonyl compounds with p-toluenesulfonylhydrazine (TSH) followed by their analysis using liquid chromatography coupled to electrospray ionisation high-resolution mass spectrometry (LC-ESI-HRMS) and

Ethanol metabolism: The good, the bad, and the ugly.

Throughout the world, ethanol is both an important commercial commodity and a source of major medical and social problems. Ethanol readily passes through biological membranes and distributes throughout the body. It is oxidized, first to acetaldehyde and then to acetate, and finally by the citric acid cycle in virtually all tissues. The oxidation of ethanol is irreversible and unregulated, making the rate dependent only on local concentration and enzyme activity. This unregulated input of reducing equivalents increases reduction of both cytoplasmic and intramitochondrial NAD and, through the la

On the sources and sinks of atmospheric VOCs: an integrated analysis of recent aircraft campaigns over North America.

We apply a high-resolution chemical transport model (GEOS-Chem CTM) with updated treatment of volatile organic compounds (VOCs) and a comprehensive suite of airborne datasets over North America to (i) characterize the VOC budget and (ii) test the ability of current models to capture the distribution and reactivity of atmospheric VOCs over this region. Biogenic emissions dominate the North American VOC budget in the model, accounting for 70 % and 95 % of annually emitted VOC carbon and reactivity, respectively. Based on current inventories anthropogenic emissions have declined to the point wher

Carcinogenic Air Toxics Exposure and Their Cancer-Related Health Impacts in the United States.

Public health protection from air pollution can be achieved more effectively by shifting from a single-pollutant approach to a multi-pollutant approach. To develop such multi-pollutant approaches, identifying which air pollutants are present most frequently is essential. This study aims to determine the frequently found carcinogenic air toxics or hazardous air pollutants (HAPs) combinations across the United States as well as to analyze the health impacts of developing cancer due to exposure to these HAPs. To identify the most commonly found carcinogenic air toxics combinations, we first ident

Prenatal effects of alcohol.

Suspicion of alcohol's teratogenic potential stretches back many centuries, but it is only recently that solid support for this possibility has been produced. There is now little doubt that alcohol can produce developmental defects, but there are many questions that still remain to be answered concerning the impact of alcohol on the conceptus. One major question that remains to be resolved is why only a small percentage of alcoholic women give birth to children with Fetal Alcohol Syndrome (FAS), whereas other alcoholic women who drink the same amount do not. Another important issue concerns th

High-Resolution Mass Spectrometric Analysis of Selected DNA Adducts in Smokeless Tobacco Users' Oral Cells.

Smokeless tobacco, used by more than 360 million people worldwide, is an accepted cause of cancers of the oral cavity, esophagus, and pancreas. DNA adducts are considered as critical intermediates in the carcinogenic process, but there are no published studies that have used structure-specific methods to identify and quantify oral cell DNA adducts in people who use smokeless tobacco. In the study presented here, we used liquid chromatography-nanoelectrospray ionization-high resolution tandem mass spectrometry to identify and quantify DNA adducts in oral cells of 24 individuals who use smokeles

Identification of mitochondria-associated hub genes related to alcohol-associated liver fibrosis progression in aldehyde dehydrogenase 2 deficiency.

BACKGROUND & AIMS: The pathogenesis of alcohol-associated liver fibrosis remains incompletely understood. Aldehyde dehydrogenase 2 (ALDH2) is the primary enzyme responsible for detoxifying ethanol-derived acetaldehyde. Notably, the ALDH2 rs671 variant is carried by approximately 8% of the global population, with a prevalence of 30%-50% in East Asians. This variant reduces ALDH2 enzymatic activity. However, the contribution of impaired ALDH2 function to fibrotic susceptibility and the underlying regulatory molecules remains to be fully elucidated. METHODS: Blood samples from patients with alco
