LPO and ethanol biotransformation systems in the liver as markers of predisposition to ethanol hepatotoxicity.

An original experimental model for detecting organ-specific markers of predisposition to ethanol hepatotoxicity is proposed. A relationship between congenital activity of LPO processes in rat liver (before ethanol intoxication) and the type and severity of ethanol-induced damage to the liver was demonstrated using methods of mathematical modeling. It was proven that intact rats with genetically high MDA levels in the liver and more active systems of MDA generation in ascorbate- and NADPH-dependent reactions are prone to ethanol-induced damage to the liver.

Effect of trichloroethylene (TCE) inhalation on biotransformation enzymes of rat lung and liver.

Trichloroethylene (TCE) is widely used as an industrial solvent and cleaning fluid. In the present study the toxic effects of TCE inhalation on pulmonary and hepatic biotransformation enzymes in rats have been investigated by assay of aniline hydroxylase (AH), aminopyrine-N-demethylase (APD), benzo-a-pyrene hydroxylase (BH) and glutathione-s-transferase (GST) activities and glutathione (GSH) contents in liver as well as lungs of exposed animals. In both organs phase I and phase II drug metabolizing enzymes have been found to be increased along with decrease in GSH contents following TCE inhala

Influence of the halogen-substituent pattern of fluoronitrobenzenes on their biotransformation and capacity to induce methemoglobinemia.

In the present study both the biotransformation patterns and the capacity to induce methemoglobinemia of a series of fluoronitrobenzenes were investigated. This was done to investigate to what extent variation in the number and position of the halogen substituents influence the metabolic fate of the fluoronitrobenzenes, thereby influencing their capacity to induce methemoglobinemia. The results obtained were compared to the effect of the fluorine substituent patterns on the calculated electronic characteristics and, thus, on the chemical reactivity of the fluoronitrobenzenes. Analysis of the i

In vitro biotransformation of finasteride in rat hepatic microsomes. Isolation and characterization of metabolites.

Metabolism of finasteride ([N-(1,1-dimethylethyl)-3-oxo-4-aza-5 alpha-androst-1-ene-17 beta- carboxamide]; MK-906), a new type of specific inhibitor of testosterone 5 alpha-reductase, was investigated using rat hepatic microsomes. The metabolism of finasteride by rat hepatic microsomes was oxygen- and NADPH-dependent, and addition of metyrapone, 7,8-benzoflavone, and cytochrome c to the incubation mixture inhibited the metabolism of finasteride. It is suggested that the metabolic reaction of finasteride was mediated by a mixed function oxidase involving P-450. Four major metabolites were detec

Effects of valproate on xenobiotic biotransformation in rat liver. In vivo and in vitro experiments.

Male Wistar rats were in vivo exposed for 2 weeks to 100 micrograms/ml sodium valproate by subcutaneous implantation of osmotic pumps and hepatocytes were isolated. As an in vitro model co-cultures of rat hepatocytes with epithelial cells were daily treated with valproate (25, 50, 100, 200 micrograms/ml) for 2 weeks. In both models the cytochrome P-450 content and the enzymatic activities of 7-ethoxycoumarin O-deethylase, aldrin epoxidase and glutathione S-transferase were determined in valproate-treated hepatocytes, in controls and in phenobarbital-induced cells. It appeared that in both syst

[Biotransformation and toxicity of xenobiotics in various routes of administration of vitamin A into the rat body].

Deficiency of vitamin A in rats caused a decrease in metabolic rate of acetanilide as well as in elimination of voltaren from blood plasma, while excess of the vitamin (3,000 IU/kg) stimulated voltaren elimination and increased excretion with urine of aminophenol conjugates of acetanilide with sulfuric and glucuronic acids. Under conditions of vitamin A deficiency formation of acetanilide toxic metabolites was increased as a result of which excretion of mercapturic acids with urine was elevated but excess of vitamin A and its hyperdose inhibited these reactions. At the same time, deficiency of

Biotransformation of the antiestrogen clomiphene to chemically reactive metabolites in the immature female rat.

Novel metabolites of clomiphene (CLO), an antiestrogen effective in experimental breast cancer models, were characterized in studies using immature female rats. After i.p. administration, CLO was eliminated unchanged in feces and as two components which were chromatographically identical to synthetic CLO analogues bearing a m-methoxy-p-hydroxyphenyl (guaiacol) moiety in place of one or the other of its phenyl rings. These components were also found in liver tissue. In the presence of liver microsomal homogenate, CLO underwent p-hydroxylation of either of its phenyl rings, affording 4-hydroxy-C

Environmental fate and risk of diverse oxygenated isomeric prothioconazole biotransformation products in soil.

Isomeric biotransformation products (TPs) of organic pesticides substantially expand structural diversity and may exhibit distinct environmental behaviors compared with their parent compounds. However, the formation mechanisms and ecological risks of such isomers (regioisomers) in soil remain poorly understood. In this study, the triazole fungicide prothioconazole (PTC) was incubated in agricultural soil, and 21 isomeric TPs were identified and classified into six structural groups using high-resolution mass spectrometry (HR-MS) based on diagnostic fragmentation patterns. These secondary oxyge

Biotransformation-electrochemistry coupling for multiplexed on-site detection of hydrophobic polycyclic aromatic pollutants.

Rapid on-site detection of polycyclic aromatic compounds (PACs), such as biphenyl and carbazole, remains challenging because of their poor aqueous solubility, weak electroactivity, and matrix interference. Here, we developed a biotransformation-enabled electrochemical decoding strategy that couples a biphenyl 2,3-dioxygenase (BphA)-expressing whole-cell module with a miniaturized reduced graphene oxide/colloidal gold (rGO/CG)-modified screen-printed electrode for biphenyl and carbazole quantification. BphA from Pseudomonas putida B6-2 was heterologously expressed in Escherichia coli to convert

Laccase-mediated biotransformation potential for fluorinated compounds by geographically diverse human gut microbiota.

The growing prevalence of synthetic organofluorine substances in agrochemicals, food packaging, and consumer products has led to increasing gastrointestinal exposure, with potential consequences for human health. Despite the extreme stability of fluorinated compounds, several microbial pathways for their transformation are known, including those involving laccases, a type of multicopper oxidase. However, the functionality of laccases in the gut microbiome, a natural contact point between food-associated chemicals and microbial biotransformation pathways, is poorly defined. Through a multi-stud

Production of biliverdin by biotransformation of exogenous heme using recombinant Pichia pastoris cells.

Biliverdin, a bile pigment hydrolyzed from heme by heme oxygenase (HO), serves multiple functions in the human body, including antioxidant, anti-inflammatory, and immune response inhibitory activities. Biliverdin has great potential as a clinical drug; however, no economic and efficient production method is available currently. Therefore, the production of biliverdin by the biotransformation of exogenous heme using recombinant HO-expressing yeast cells was studied in this research. First, the heme oxygenase-1 gene (HO1) encoding the inducible plastidic isozyme from Arabidopsis thaliana, with t

Biosorption and biotransformation behaviours of veterinary antibiotics under aerobic livestock wastewater treatment processes.

To study the fate of veterinary antibiotics released from swine wastewater treatment plants (SWTP), 10 antibiotics were investigated in each unit of a local SWTP periodically. Over a 14-month period of field investigation into target antibiotics, it was confirmed that tetracycline, chlortetracycline, sulfathiazole, and lincomycin were used in this SWTP, with their presence observed in raw manure. Most of these antibiotics could be effectively treated by aerobic activated sludge, except for lincomycin, which was still detected in the effluent, with a maximum concentration of 1506 μg/L. Benzimidazole fungicide biotransformation by comammox Nitrospira bacteria: Transformation pathways and associated proteomic responses.

Benzimidazole fungicides are frequently detected in aquatic environments and pose a serious health risk. Here, we investigated the metabolic capacity of the recently discovered complete ammonia-oxidizing (comammox) Nitrospira inopinata and kreftii to transform a representative set of benzimidazole fungicides (i.e., benzimidazole, albendazole, carbendazim, fuberidazole, and thiabendazole). Ammonia-oxidizing bacteria and archaea, as well as the canonical nitrite-oxidizing Nitrospira exhibited no or minor biotransformation activity towards all the five benzimidazole fungicides. In contrast, the i

Response mechanism of microbial community during anaerobic biotransformation of marine toxin domoic acid.

Domoic acid (DA) is a potent neurotoxin produced by toxigenic Pseudo-nitzschia blooms and quickly transfers to the benthic anaerobic environment by marine snow particles. DA anaerobic biotransformation is driven by microbial interactions, in which trace amounts of DA can cause physiological stress in marine microorganisms. However, the underlying response mechanisms of microbial community to DA stress remain unclear. In this study, we utilized an anaerobic marine DA-degrading consortium GLY (using glycine as co-substrate) to systematically investigate the global response mechanisms of microbia

Sertraline as a new potential anthelmintic against Haemonchus contortus: toxicity, efficacy, and biotransformation.

Haemonchus contortus is a parasitic nematode of ruminants which causes significant losses to many farmers worldwide. Since the drugs currently in use for the treatment of haemonchosis are losing their effectiveness due to the drug-resistance of this nematode, a new or repurposed drug is highly needed. As the antipsychotic drug sertraline (SRT) has been shown to be effective against the parasitic nematodes Trichuris muris, Ancylostoma caninum and Schistosoma mansoni, the aim of the present study was to evaluate the possible effect of SRT on H. contortus. The potential hepatotoxicity of SRT was

Genetic variation in biotransformation enzymes, air pollution exposures, and risk of spina bifida.

Spina bifida is a birth defect characterized by incomplete closure of the embryonic neural tube. Genetic factors as well as environmental factors have been observed to influence risks for spina bifida. Few studies have investigated possible gene-environment interactions that could contribute to spina bifida risk. The aim of this study is to examine the interaction between gene variants in biotransformation enzyme pathways and ambient air pollution exposures and risk of spina bifida. We evaluated the role of air pollution exposure during pregnancy and gene variants of biotransformation enzymes

Biotransformation of hexavalent chromium into extracellular chromium(III) oxide nanoparticles using Schwanniomyces occidentalis.

OBJECTIVES: To demonstrate biotransformation of toxic Cr(VI) ions into Cr2O3 nanoparticles by the yeast Schwanniomyces occidentalis. RESULTS: Reaction mixtures containing S. occidentalis NCIM 3459 and Cr(VI) ions that were initially yellow turned green after 48 h incubation. The coloration was due to the synthesis of chromium (III) oxide nanoparticles (Cr2O3NPs). UV-Visible spectra of the reaction mixtures showed peaks at 445 and 600 nm indicating (4)A2g → (4)T1g and (4)A2g → (4)T2g transitions in Cr2O3, respectively. FTIR profiles suggested the involvement of carboxyl and amide groups in nan

Enhancement of anaerobic carbon tetrachloride biotransformation in methanogenic sludge with redox active vitamins.

Carbon tetrachloride (CT) is an important groundwater pollutant which is only subject to biotransformation in the absence of oxygen. The anaerobic biotransformation of CT is influenced by electron shuttling compounds. The purpose of this study was to evaluate the impact of redox active vitamins on CT (100 microM) metabolism in a methanogenic sludge consortium (0.5 g VSS l(-1)) supplied with volatile fatty acids as electron donor (0.2 g COD l(-1)). The redox active vitamins, tested at concentrations ranging from 0.5 to 20 microM, were riboflavin (RF) and two forms of vitamin B12, cyanocobalamin

Fusel oil as precursor for aroma generation by biotransformation using lipase: scientific note.

The feasibility of using mixtures of C4 and C5 chain-length aliphatic alcohols from fusel oil as the bulk organic media for lipase-mediated synthesis of laurate esters was assessed. Reaction mixtures consisted of lauric acid, lipase, solvent (if added), and appropriate amount of fusel oil (previously dehydrated with inorganic salts and molecular sieves). The influence of the reaction conditions such as substrate concentrations and temperature were investigated. Increased molar ratio of acyl donor to acyl acceptor allowed the esterification to proceed with no need for solvent addition.
