Streamlined F2-isoprostane analysis in plasma and urine with high-performance liquid chromatography and gas chromatography/mass spectroscopy.

F2-Isoprostanes in plasma and urine are generally determined by labor-intensive methods requiring sample purification by solid-phase extraction and thin-layer chromatography (TLC). A streamlined and more sensitive method for the measurement of esterified plasma F2-isoprostanes was developed by replacing these steps with high-performance liquid chromatography (HPLC) using an amino column with a hexane/2-propanol gradient. Pentafluorobenzyl esters of F2-isoprostanes were prepared and purified by HPLC, silylated, and then analyzed by gas chromatography (GC) with negative chemical ionization mass

Activated carbon-coated particles as adsorbent for packed columns in gas chromatography.

This study reports the first development of gas chromatographic columns packed with activated carbon (AC)-coated adsorbent particles. The AC-coated adsorbent particles were prepared by coating an AC layer on solid support particles using a vacuum carbon coater. Glass beads, diatomite, a porous polymer, and activated alumina particles were investigated as the support materials. The AC layer was uniformly coated on the surface of each support material, achieving a thickness of approximately 300-400 nm after 500 coating cycles. The AC-coated particles were packed in a stainless steel tube with an

Profiling of Volatile Metabolites of Escherichia coli Using Gas Chromatography-Mass Spectrometry.

Current diagnostic methods for bacterial infections in critically ill patients, including ventilator-associated pneumonia (VAP), are time-consuming, while empirical antibiotic therapy contributes to rising resistance. Bacteria-derived volatile organic compounds (VOCs) are being explored as specific biomarkers for pathogen identification and treatment monitoring. This study expands knowledge of Escherichia coli metabolism by identifying VOCs produced by both multidrug-resistant and susceptible strains, characterizing their temporal profiles during growth, and assessing VOC profile changes after

[Determination of multiple residual solvents in ibandronate sodium using headspace-gas chromatography].

Ibandronate sodium, a third-generation diphosphate drug used worldwide to treat osteoporosis, has the advantages of convenient use, low toxicity, and significant therapeutic effects. However, the residual organic solvents in the synthesis process of sodium ibandronate not only have a negative impact on the efficacy of the drug, but also lead to a decrease in drug stability. Moreover, if the residual amounts of these solvents exceed safety standards, they may pose serious threats to human health. This study successfully established a convenient and efficient method based on headspace-gas chroma

Toxicological analysis of azide and cyanide for azide intoxications using gas chromatography.

Azide is a highly toxic chemical agent to human being. Accidental, but also intentional exposure to azide occurs. To be able to confirm azide ingestion, we developed a method to identify and quantify azide in biological matrices. Cyanide was included in the method to evaluate suggested in vivo production of cyanide after azide ingestion. Azide in biological matrices was first derivatized by propionic anhydride to form propionyl azide. Simultaneously, cyanide was converted into hydrogen cyanide. After thermal rearrangement of propionyl azide, ethyl isocyanate was formed, separated together with

[Determination of 29 adjuvants in pesticide formulations by gas chromatography-mass spectrometry].

A method based on gas chromatography-mass spectrometry (GC-MS) was developed for the simultaneous determination of 29 adjuvants in pesticide formulations. The pesticide formulation was diluted with methanol, and the supernatant was filtered through a 0.22-μm membrane. After separation on a VF-1701MS column, the analytes were detected by mass spectrometry in the selected ion monitoring (SIM) mode and quantified using the external standard method. Good linearities were obtained for the 29 pesticide adjuvants in the linear range of 6.2-400.0 mg/L, with correlation coefficients (R2) larger than 0.

[Determination of three ester compounds in gasoline by two-dimensional gas chromatography].

A simple and efficient method based on the technique of packed column switching-back flushing was established for the analysis of the ester compounds (including ethyl acetate, sec-butyl acetate, dimethyl carbonate) in gasoline. With the use of a non-polar pre-column, we successfully separated the components in gasoline by back flushing out components heavier than n-octane, while the lighter components and the ester compounds were flushed into a polar analytical column. In this method, external standard method was applied for quantification. As a result, good linear relationships existed among

Applications of planar microfluidic devices and gas chromatography for complex problem solving.

The application of planar microfluidic devices in GC for the separation of components of interest otherwise difficult to separate in a single analysis is presented. A variety of configurations were used for parallel chromatography, column effluent splitting, back flushing, selectivity tuning, valve less switching and column isolation, heart cutting, and comprehensive multidimensional chromatography. The synergies of recently commercialized planar microfluidic devices combined with the resolving power of fused-silica capillary columns are demonstrated. Difficult separations were accomplished in

Isotope dilution-mass spectrometry determination of blood cyanide by headspace gas chromatography.

A direct and sensitive method for the determination of blood cyanide by isotope dilution was developed. The blood is placed in a headspace vial, and K13C15N is added as internal standard. Addition of phosphoric acid liberates the cyanide as HCN. The detection is accomplished by mass spectrometry after a fine mass calibration tuning. The detection limit obtained is 0.3 micromol/L. The within- and inter-run coefficients of variation are 4.4% (for a concentration of 2.5 micromol/L) and 3.9% (for a concentration of 4.7 micromol/L), respectively. The observed recovery is 98%. A round-robin exercise

Methyl methacrylate in poly(methyl methacrylate)--validation of direct injection gas chromatography.

Gas chromatography (GC) was investigated for the determination of residual methyl methacrylate (MMA) in heat-processed poly(methyl methacrylate) (PMMA) denture base material emphasizing recovery and validation. Standard solutions of MMA and emulsion-polymerized PMMA in dichloromethane were analysed, before and after distillation by a room-temperature air stream into a liquid nitrogen trap, and in the presence of PMMA by direct injection. Quantitative NMR analysis using dimethyl sulphoxide as internal calibration standard in deuterated chloroform solutions provided validation. Good concordance

Pesticide residue analysis in fresh produce by gas chromatography-tandem mass spectrometry.

GC-MS-MS was used to analyze for multiple pesticide residues in fresh fruits and vegetables. Nineteen pesticides, including fungicides, herbicides, organophosphorus insecticides and chlorinated insecticides were spiked into seven different fruit and vegetable matrices and analyzed. The specificity of GC-MS-MS provided for low detection limits (ranging from 1-5 ppb) and unambiguous spectral confirmation for the target compounds in these complex matrices. Good precision was obtained (RSD values ranged from 1 to 9%), even at the 10 ppb level, without extensive sample clean-up steps.

Determination of N-acetylcysteine in human plasma by gas chromatography-mass spectrometry.

An automatic mass spectrometric method for the quantitation of N-acetylcysteine (NAC) in human plasma has been developed. NAC was extracted from plasma with ethyl acetate and derivatized in two steps with 2-propanol and pentafluoropropionic anhydride. The volatile derivative obtained was ideal for gas chromatographic-mass spectrometric analysis. Data obtained by analysing the plasma of healthy volunteers to whom 600 mg of NAC had been orally given are reported.

Determination of trichloroethylene metabolites in rat liver homogenate using headspace gas chromatography.

An headspace gas chromatographic (HSGC) method for determination of trichloroethylene metabolites in rat liver homogenates is described. These metabolites are chloral hydrate (CH), trichloroethanol (TCE), trichloroacetic acid (TCA) and the glucuronic acid conjugate of trichloroethanol (TCE-beta-glucuronide). The method is based on selective thermal conversion of CH and TCA into chloroform, which is determined together with trichloroethanol by HSGC using electron-capture detection. TCE-beta-glucuronide was determined as the difference between free TCE and total TCE after enzymatic hydrolysis wi
