Effect of psychotropic drug treatment on sterol metabolism.

Cholesterol metabolism is vital for brain function. Previous work in cultured cells has shown that a number of psychotropic drugs inhibit the activity of 7-dehydrocholesterol reductase (DHCR7), an enzyme that catalyzes the final steps in cholesterol biosynthesis. This leads to the accumulation of 7-dehydrocholesterol (7DHC), a molecule that gives rise to oxysterols, vitamin D, and atypical neurosteroids. We examined levels of cholesterol and the cholesterol precursors desmosterol, lanosterol, 7DHC and its isomer 8-dehydrocholesterol (8DHC), in blood samples of 123 psychiatric patients on vario

Azole-antifungal binding to a novel cytochrome P450 from Mycobacterium tuberculosis: implications for treatment of tuberculosis.

Although antibiotics against Mycobacterium tuberculosis have decreased the incidence of tuberculosis infections significantly, the emergence of drug-resistant strains of this deadly pathogen renders current treatments ineffective. Therefore, it is imperative to identify biochemical pathways in M. tuberculosis that can serve as targets for new anti-mycobacterial drugs. We recently cloned, expressed, and purified MT CYP51, a soluble protein from M. tuberculosis that is similar in sequence to CYP51 (lanosterol-14alpha-demethylase) isozymes, pharmacological targets for several anti-mycotic compoun

Fluconazole: a new antifungal agent.

The chemistry, activity, pharmacokinetics, clinical efficacy, adverse effects, drug interactions, and administration of fluconazole are reviewed. Fluconazole is a triazole antifungal agent labeled for use in the treatment of oropharyngeal and esophageal candidiasis and cryptococcal meningitis. Like the imidazoles, fluconazole inhibits the C-14 demethylation of lanosterol, but it has little or no effect on mammalian cytochrome P-450 enzymes. Fluconazole's activity in vitro does not reflect its effectiveness in vivo. Pharmacokinetic characteristics include water solubility and excellent bioavail

Mechanistic insights into the interaction of Abamectin and Ivermectin with bovine serum albumin and the protective role of ganoderic acid A: An integrated spectroscopic and in silico study.

Abamectin and Ivermectin are widely used macrocyclic lactones whose exposure may induce oxidative stress and protein structural alterations. Since serum albumin plays a key role in xenobiotic transport, understanding ligand-albumin interactions is important for evaluating their molecular effects. Ganoderic acid A (GAA), a bioactive triterpenoid from Ganoderma lucidum, has reported antioxidant properties and may reduce ligand-induced protein damage. The interactions of Abamectin and Ivermectin with bovine serum albumin (BSA) were investigated in the absence and presence of GAA using UV-Vis, flu

Ganoderic acid A ameliorated LPS-induced depression-like behaviors via suppression of acute neuroinflammation.

Research context reveals that major depressive disorder (MDD), as a prevalent chronic relapsing psychiatric condition affecting up to one-third of patients with treatment resistance, necessitates urgent development of novel therapeutic agents. Emerging evidence implicates neuroinflammatory mechanisms in MDD pathophysiology, while the antidepressant potential of Ganoderic acid A (GAA), a triterpenoid compound derived from Ganoderma lucidum, particularly through modulation of lipopolysaccharide (LPS)-induced depression-like behaviors via acute neuroinflammation suppression remains underexplored.

HSD17B7 is required for the function of sensory hair cells by regulating cholesterol synthesis.

Cholesterol homeostasis is fundamental to cellular function, and its disruption underlies a wide range of human diseases. However, the contribution of cholesterol biosynthesis to auditory physiology remains poorly understood. HSD17B7 (17β-Hydroxysteroid dehydrogenase type 7) catalyzes the conversion of zymosterone to zymosterol, a key step in the post-lanosterol cholesterol biosynthetic pathway. Here, we found that Hsd17b7 is highly enriched in sensory hair cells of zebrafish and mice. The deficiency of Hsd17b7 reduced intracellular cholesterol levels in HEI-OC1 cells and zebrafish hair cells,

Comparative Analysis of Two Candida parapsilosis Isolates Originating from the Same Patient Harbouring the Y132F and R398I Mutations in the ERG11 Gene.

This work presents a comparative analysis of two clinical isolates of C. parapsilosis, isolated from haemoculture (HC) and central venous catheter (CVC). Both strains harboured Y132F and R398I mutations in the gene ERG11 associated with resistance to fluconazole (FLC). Differences between the HC and CVC isolates were addressed in terms of virulence, resistance to FLC, and lipid distribution. Expression of the ERG6 and ERG9 genes, lipid analysis, fatty acid composition, and lipase activity were assessed via qPCR, thin-layer chromatography/high-performance liquid chromatography, gas chromatograp

Antifungal effect of (R) and (S)-citronellal enantiomers and their predictive mechanism of action on Candida albicans from voriconazole-resistant onychomycoses.

Onychomycosis is the most common disease affecting the nail unit and accounts for at least 50% of all nail diseases. In addition, Candida albicans is responsible for approximately 70% of onychomycoses caused by yeasts. This study investigated the antifungal effect of (R) and (S)-citronellal enantiomers, as well as its predictive mechanism of action on C. albicans from voriconazole-resistant onychomycoses. For this purpose, in vitro broth microdilution and molecular docking techniques were applied in a predictive and complementary manner to the mechanisms of action. The main results of this stu

The lens epithelium as a major determinant in the development, maintenance, and regeneration of the crystalline lens.

The crystalline lens is a transparent and refractive biconvex structure formed by lens epithelial cells (LECs) and lens fibers. Lens opacity, also known as cataracts, is the leading cause of blindness in the world. LECs are the principal cells of lens throughout human life, exhibiting different physiological properties and functions. During the embryonic stage, LECs proliferate and differentiate into lens fibers, which form the crystalline lens. Genetics and environment are vital factors that influence normal lens development. During maturation, LECs help maintain lens homeostasis through mate

Induction of apoptosis and ganoderic acid biosynthesis by cAMP signaling in Ganoderma lucidum.

Apoptosis is an essential physiological process that controls many important biological functions. However, apoptosis signaling in relation to secondary metabolite biosynthesis in plants and fungi remains a mystery. The fungus Ganoderma lucidum is a popular herbal medicine worldwide, but the biosynthetic regulation of its active ingredients (ganoderic acids, GAs) is poorly understood. We investigated the role of 3',5'-cyclic adenosine monophosphate (cAMP) signaling in fungal apoptosis and GA biosynthesis in G. lucidum. Two phosphodiesterase inhibitors (caffeine and 3-isobutyl-1-methylxanthine,

Cryptococcosis: epidemiology, fungal resistance, and new alternatives for treatment.

Cryptococcosis is an important systemic mycosis and the third most prevalent disease in human immunodeficiency virus (HIV)-positive individuals. The incidence of cryptococcosis is high among the 25 million people with HIV/acquired immunodeficiency syndrome (AIDS), with recent estimates indicating that there are one million cases of cryptococcal meningitis globally per year in AIDS patients. In Cryptococcus neoformans, resistance to azoles may be associated with alterations in the target enzyme encoded by the gene ERG11, lanosterol 14α-demethylase. These alterations are obtained through mutatio

A comprehensive method for extraction and quantitative analysis of sterols and secosteroids from human plasma.

We describe the development of a method for the extraction and analysis of 62 sterols, oxysterols, and secosteroids from human plasma using a combination of HPLC-MS and GC-MS. Deuterated standards are added to 200 μl of human plasma. Bulk lipids are extracted with methanol:dichloromethane, the sample is hydrolyzed using a novel procedure, and sterols and secosteroids are isolated using solid-phase extraction (SPE). Compounds are resolved on C₁₈ core-shell HPLC columns and by GC. Sterols and oxysterols are measured using triple quadrupole mass spectrometers, and lathosterol is measured using GC

Mode of action of fenarimol against Leishmania spp.

This paper evaluates the effects of certain herbicides on Leishmania spp., their mechanism of action, and the evolutionary origin of the relevant susceptible leishmanial targets. We demonstrated that a relatively nontoxic herbicide, fenarimol, successfully interferes with a leishmanial target, which is probably a relic of an ancient ancestor. Fenarimol impairs the function of leishmanial 14alpha-sterol demethylase, a key enzyme in the sterol biosynthetic pathway. Therefore, fenarimol or its derivatives may be candidates for development of anti-leishmanial drugs. Of the herbicides that have the

Identification of various medically important Candida species in clinical specimens by PCR-restriction enzyme analysis.

A single primer pair amplifying a cytochrome P-450 lanosterol-14 alpha-demethylase (L1A1) gene fragment that encodes a highly conserved region was used to detect yeast DNA in clinical specimens. Positive PCR products were obtained from genomic DNAs of Candida albicans, C. parapsilosis, C. tropicalis, C. guilliermondii, C. krusei, C. (Torulopsis) glabrata, and C. kefyr. No human, bacterial, or parasitic DNA was amplified. The sensitivity was evaluated for C. albicans genomic DNA by using various DNA concentrations (200 pg to 2 fg). The amplified DNAs of Candida species with unknown P-450 L1A1 g

Antifungal Activity of Hedychium spicatum Essential Oil Against Candidemia-Causing Candida tropicalis and Candida parapsilosis: In Vitro Evaluation and Computational Identification of Cis-γ-Bisabolene as a Putative CYP51-Binding Constituent.

Candidiasis is caused by Candida parapsilosis and Candida tropicalis emerges due to antifungal resistance and limited therapies. We aim to assess the antifungal activity of Hedychium spicatum essential oil (HSEO), followed by computational analyses to identify potential lead phytochemicals, targeting lanosterol 14α-demethylase (CYP51), a key enzyme in ergosterol biosynthesis. GC-MS identified 29 phytoconstituents, with τ-cadinol (17.94%) as the major component. Disc diffusion demonstrated concentration-dependent antifungal activity against C. parapsilosis and C. tropicalis, with inhibition zon

New anti-fungal compounds 1PB1 and 45R from Streptomyces chrestomyceticus ADP4 target exo-β-1,3-glucanase and ergosterol biosynthesis in Candida albicans.

The present study was aimed at understanding the mode of action of two newly reported anti-Candida compounds from Streptomyces chrestomyceticus strain ADP4: Phenyl 2'α,2'β,6'β-trimethyl cyclohexyl ketone (Chrestosyl cyclohexyl ketone; 1PB1) and trans-1-oxo-2,4-diacetylaminodecalin (Chrestosyl amino decalin; 45R). The docking studies revealed significant binding scores of CYP51 with 1PB1 (-7.4 kcal/mol) and with 45R (-7.6 kcal/mol). GC-MS sterol profiling of treated cells showed accumulation of lanosterol, confirming CYP51 inhibition. In vitro assays confirmed a consequent decrease in ergostero

Azole derivatives with naphthalene showing potent antifungal effects against planktonic and biofilm forms of Candida spp.: an in vitro and in silico study.

Candida infections pose a serious public health threat due to increasing drug resistance. Azoles are first-line antifungal drugs for fungal infections. In this study, we tested an in-house azole collection incorporating naphthalene ring to find hits against planktonic and biofilm forms of resistant Candida spp. In the collection, potent derivatives were identified against the susceptible strains of Candida with minimum inhibitory concentration (MIC) values lower than those of the reference drug, fluconazole. MIC values of 0.125 μg/ml against C. albicans, 0.0625 μg/ml against C. parapsilosis, a

Mutations in the gene encoding 3 beta-hydroxysteroid-delta 8, delta 7-isomerase cause X-linked dominant Conradi-Hünermann syndrome.

X-linked dominant Conradi-Hünermann syndrome (CDPX2; MIM 302960) is one of a group of disorders with aberrant punctate calcification in cartilage, or chondrodysplasia punctata (CDP). This is most prominent around the vertebral column, pelvis and long bones in CPDX2. Additionally, CDPX2 patients may have asymmetric rhizomesomelia, sectorial cataracts, patchy alopecia, ichthyosis and atrophoderma. The phenotype in CDPX2 females ranges from stillborn to mildly affected individuals identified in adulthood. CDPX2 is presumed lethal in males, although a few affected males have been reported.
