Degradation of the tricyclic antipsychotic drug chlorpromazine under environmental conditions, identification of its main aquatic biotic and abiotic transformation products by LC-MSn and their effects on environmental bacteria.

The search for environmental transformation products of organic pollutants (like drugs) is a difficult task and usually only few compounds are detected. This might be due to effective degradation but could also be a result of analytical deficits dealing with complex matrices. Especially transformation products of very low concentrations in sludge were difficult to identify so far. Additionally, the use of standard separation techniques might lead to the loss of isomeric compounds, which possess identical spectroscopic and spectrometric properties. To date no complete study investigating the en

Visualization of multidrug-resistant bacterial infection trends in the intensive care units.

BACKGROUND: The Korean National Healthcare-associated Infections Surveillance System (KONIS) monitors multidrug-resistant (MDR) bacterial infections in intensive care units (ICUs). However, simultaneously monitoring hundreds of ICUs remains challenging. Our study aimed to visualize the trends of MDR gram-negative bacterial infections in ICUs monitored by KONIS. METHODS: We evaluated KONIS data from 137 ICUs (2006-2011) and from 368 ICUs (2017-2022). Pneumonia, urinary tract infection, and bloodstream infection caused by Klebsiella pneumoniae, Pseudomonas aeruginosa, and Acinetobacter baumanni

Simultaneous abiotic reduction-biotic oxidation in a microbial-MnO2-catalyzed Fenton-like system.

The possibility of simultaneous activity of superoxide-mediated transformations and heterotrophic aerobic bacterial metabolism was investigated in catalyzed H(2)O(2) propagations (CHP; i.e., modified Fenton's reagent) systems containing Escherichia coli. Two probe compounds were used: glucose for the detection of heterotrophic metabolism of E. coli, and tetrachloromethane (CCl(4)) for the detection of superoxide generated in a MnO(2)-catalyzed CHP system. In the MnO(2)-catalyzed CHP system without bacteria, only CCl(4) loss was observed; in contrast, only glucose degradation occurred E. coli s

Effect of toll-like receptor agonists on the formation of macrophage/foam cells upon acute peritonitis in mice.

We studied effects of zymosan, double-stranded RNA, LPS of E. coli and bacterial CpG DNA, agonists of toll-like receptor TLR2, TLR3, TLR4 and TLR9, respectively, on the formation of macrophage/foam cells 24 h after induction of acute peritonitis. Administration of agonists led to transformation of peritoneal macrophages into foam cells and significant activation of cell biosynthesis and increased the content of triglycerides and cholesterol esters in the absence of LDL and irrespective of the capacity of TLR agonists to stimulate neutrophil infiltration and TNF-α production in the peritoneal c

On-Demand Changeable Theranostic Hydrogels and Visual Imaging-Guided Antibacterial Photodynamic Therapy to Promote Wound Healing.

Antibacterial wound dressings are confronted with the challenges in real-time imaging of infected wounds and effective removal of bacterial debris after sterilization to promote the healing process. Herein, injectable theranostic hydrogels were constructed from antimicrobial peptide ε-polylysine (ePL) and polydopamine (PDA) nanoparticles for real-time diagnosis of infected wounds, imaging-guided antibacterial photodynamic therapy (PDT), and on-demand removal of bacterial debris. Ureido-pyrimidinone was conjugated on ePL to produce PLU hydrogels through quadruple hydrogen bonding, and the inocu

Quantification of filamentation by uropathogenic Escherichia coli during experimental bladder cell infection by using semi-automated image analysis.

Several rod-shaped pathogens including Escherichia coli, Salmonella spp. and Klebsiella pneumonia are capable of adopting highly filamentous cell shapes under certain circumstances. This phenomenon occurs as a result of continued cell elongation during growth without the usual septation into single rod-shaped cells. Evidence has emerged over the past decade suggesting that this morphological transformation is controlled and reversible and provides selective advantages under certain growth conditions, such as during infection in humans. In order to identify the factors which induce filamentatio

Surviving the storm: exploring the role of natural transformation in nutrition and DNA repair of stressed Deinococcus radiodurans.

Deinococcus radiodurans, a natural transformation (NT)-enabled bacterium renowned for its exceptional radiation resistance, employs unique DNA repair and oxidative stress mitigation mechanisms as a strategic response to DNA damage. This study excavates into the intricate roles of NT machinery in the stressed D. radiodurans, focusing on the genes comEA, comEC, endA, pilT, and dprA, which are instrumental in the uptake and processing of extracellular DNA (eDNA). Our data reveal that NT not only supports the nutritional needs of D. radiodurans under stress but also has roles in DNA repair. The st

Paenibacillus polymyxa L62 synergistically regulates Pb2+ speciation and reconstructs soil microecological networks via surface complexation and PbS mineralization.

The adsorption activity of Paenibacillus polymyxa L62 toward Pb2+ was evaluated in aqueous systems. Strain L62 achieved a Pb2+ removal efficiency of 95.97%. Its cell surface was enriched with functional groups such as -OH and -COO⁻, which promoted Pb2+ adsorption via complexation and ion exchange. In aqueous systems, XRD, FTIR, and EDS analyses suggested the possible formation of stable PbS crystalline precipitates on the cell surface. In soil incubation experiments, strain L62 was inoculated into Pb-contaminated soil at 1% (L1) and 3% (L3). Both treatments significantly promoted Pb transforma

Functional comparison of anti-restriction and anti-methylation activities of ArdA, KlcA, and KlcAHS from Klebsiella pneumoniae.

Anti-restriction proteins are typically encoded by plasmids, conjugative transposons, or phages to improve their chances of entering a new bacterial host with a type I DNA restriction and modification (RM) system. The invading DNA is normally destroyed by the RM system. The anti-restriction proteins ArdA, KlcA, and their homologues are usually encoded on plasmid of carbapenemase-resistant Klebsiella pneumoniae. We found that the plasmid sequence and restriction proteins affected horizontal gene transfer, and confirmed the anti-restriction and anti-methylation activities of ArdA and KlcA during

Specific DNA recognition mediated by a type IV pilin.

Natural transformation is a dominant force in bacterial evolution by promoting horizontal gene transfer. This process may have devastating consequences, such as the spread of antibiotic resistance or the emergence of highly virulent clones. However, uptake and recombination of foreign DNA are most often deleterious to competent species. Therefore, model naturally transformable gram-negative bacteria, including the human pathogen Neisseria meningitidis, have evolved means to preferentially take up homotypic DNA containing short and genus-specific sequence motifs. Despite decades of intense inve

Ursodeoxycholic acid, 7-ketolithocholic acid, and chenodeoxycholic acid are primary bile acids of the nutria (Myocastor coypus).

Because ursodeoxycholic and chenodeoxycholic acids are interconverted in humans via 7-ketolithocholic acid, bile acid metabolism was studied in the nutria (Myocastor coypus), the bile of which is known to contain these three bile acids. Relative concentrations of ursodeoxycholic (37% +/- 20%), 7-ketolithocholic (33% +/- 17%), and chenodeoxycholic (17% +/- 9%) acids in gallbladder bile were unchanged by 5-20 h of complete biliary diversion (n = 7). Ruminal bacterial communities differ in early-lactation dairy cows with differing risk of ruminal acidosis.

INTRODUCTION: Early-lactation Holstein cows (n= 261) from 32 herds in three regions (Australia, California, and Canada) were previously categorized using a discriminant analysis model as being at a high (26.1% of cows), medium (26.8% of cows), or low risk (47.1% of cows) of ruminal acidosis. We aimed to investigate if (1) risk of acidosis would be associated with ruminal bacterial taxa and dietary nutrient components, (2) there would be individual or combinations of bacterial taxa associated with acidosis-risk groups, and (3) the abundance of bacterial taxa would be associated with the intake

Recombinant Protein Production and Purification of Rift Valley Fever Virus Nucleoprotein from Escherichia coli Expression Systems.

The recombinant expression and purification of viral proteins are a key component in the study of the immune response of viruses, as well as the creation of diagnostic techniques for the detection of viruses. For structurally simple proteins, one commonly used technique is the production of recombinant proteins in bacterial expression systems, which enable the large-scale synthesis and purification of recombinant viral proteins. In this technique, the cDNA encoding for a viral protein is cloned into a bacterial expression vector (with an appropriate purification tag), produced in a modified ba

Multiple dual C-Cl isotope patterns associated with reductive dechlorination of tetrachloroethene.

Dual isotope slopes are increasingly used to identify transformation pathways of contaminants. We investigated if reductive dechlorination of tetrachloroethene (PCE) by consortia containing bacteria with different reductive dehalogenases (rdhA) genes can lead to variable dual C-Cl isotope slopes and if different slopes also occur in the field. Two bacterial enrichments harboring Sulfurospirillum spp. but different rdhA genes yielded two distinct δ(13)C to δ(37)Cl slopes of 2.7 ± 0.3 and 0.7 ± 0.2 despite a high similarity in gene sequences. This suggests that PCE reductive dechlorination could

Bacterial pyridine hydroxylation is ubiquitous in environment.

Ten phenol-degrading bacterial strains were isolated from three geographically distant environments. Five of them, identified as Diaphorobacter, Acidovorax, Acinetobacter (two strains), and Corynebacterium, could additionally transform pyridine, through the transcription of phenol hydroxylase genes induced both by phenol and pyridine. HPLC-UV and LC-MS analyses indicated that one metabolite (m/e = 96.07) with the same molecular weight as monohydroxylated pyridine was produced from the five phenol-degrading strains, when pyridine was the sole carbon source. Phenol (50 mg l(-1)) could initially

Integrating natural gradients and controlled assays to reveal bacterial responses to cadmium in Theobroma cacao L., soils.

Cadmium (Cd), a toxic heavy metal found in agricultural landscapes worldwide, has been pointed out in cropped soils with Theobroma cacao L., as one of the main contaminants that translocate into plant tissues. Among the factors linked to cadmium translocation into plants, the role of soil bacterial communities in chemical transformation in soils has been poorly investigated. Overall, soil bacterial communities are shaped by diverse environmental and anthropogenic factors that influence crop yield and health. Cadmium alters soil microbial communities and increases the risk to human health throu
