On modeling the neuronal activity in movement disorder patients by using the Ornstein Uhlenbeck process.

Mathematical models of the neuronal activity in the affected brain regions of Essential Tremor (ET) and Parkinson's Disease (PD) patients could shed light into the underlying pathophysiology of these diseases, which in turn could help develop personalized treatments including adaptive Deep Brain Stimulation (DBS). In this paper, we use an Ornstein Uhlenbeck Process (OUP) to model the neuronal spiking activity recorded from the brain of ET and PD patients during DBS stereotactic surgery. The parameters of the OUP are estimated based on Inter Spike Interval (ISI) measurements, i.e., the time int

Dynamic pseudo-observations: a robust approach to dynamic prediction in competing risks.

In this article, we propose a new approach to the problem of dynamic prediction of survival data in the presence of competing risks as an extension of the landmark model for ordinary survival data. The key feature of our method is the introduction of dynamic pseudo-observations constructed from the prediction probabilities at different landmark prediction times. They specifically address the issue of estimating covariate effects directly on the cumulative incidence scale in competing risks. A flexible generalized linear model based on these dynamic pseudo-observations and a generalized estimat

A smoothing expectation and substitution algorithm for the semiparametric accelerated failure time frailty model.

This paper proposes an estimation procedure for the semiparametric accelerated failure time frailty model that combines smoothing with an Expectation and Maximization-like algorithm for estimating equations. The resulting algorithm permits simultaneous estimation of the regression parameter, the baseline cumulative hazard, and the parameter indexing a general frailty distribution. We develop novel moment-based estimators for the frailty parameter, including a generalized method of moments estimator. Standard error estimates for all parameters are easily obtained using a randomly weighted boots

Localization of the lumbar discs using machine learning and exact probabilistic inference.

We propose a novel fully automatic approach to localize the lumbar intervertebral discs in MR images with PHOG based SVM and a probabilistic graphical model. At the local level, our method assigns a score to each pixel in target image that indicates whether it is a disc center or not. At the global level, we define a chain-like graphical model that represents the lumbar intervertebral discs and we use an exact inference algorithm to localize the discs. Our main contributions are the employment of the SVM with the PHOG based descriptor which is robust against variations of the discs and a graph

Analysis of multiple-cycle data from couples undergoing in vitro fertilization: methodologic issues and statistical approaches.

The number of in vitro fertilization (IVF) cycles in the United States increased from fewer than 46,000 in 1995 to more than 120,000 in 2005. IVF and other assisted reproductive technology (ART) data are routinely collected and used to identify outcome predictors. However, researchers do not always make full use of the data due to their complexity. Design approaches have included restriction to first-cycle attempts only, which reduces power and identifies effects only of those factors associated with initial success. Many statistical techniques have been used or proposed for analysis of IVF da

Correlation between the dioptric power, astigmatism and surface shape of the anterior and posterior corneal surfaces.

A knowledge of the shape of the cornea is of major importance for the planning and monitoring of surgery, and for the correct diagnosis of corneal diseases. Many authors have studied the geometry of the second corneal surface in the central region and it has been stated that there is a high correlation between the central radii of curvature and asphericities of the two corneal surfaces. In this work we extend this study to a larger, central, 6 mm diameter of the cornea. Surface height data, obtained with an Oculus Pentacam from 42 eyes of 21 subjects, were analysed to yield surface power vecto

SEnD NMR: sensitivity enhanced n-dimensional NMR.

Sparse sampling offers tremendous potential for overcoming the time limitations imposed by traditional Cartesian sampling of indirectly detected dimensions of multidimensional NMR data. However, in many instances sensitivity rather than time remains of foremost importance when collecting data on protein samples. Here we explore how to optimize the collection of radial sampled multidimensional NMR data to achieve maximal signal-to-noise. A method is presented that exploits a rigorous definition of the minimal set of radial sampling angles required to resolve all peaks of interest in combination

Extending the skill test for disease diagnosis.

For diagnostic tests, we present an extension to the skill plot introduced by Briggs and Zaretski (Biometrics 2008; 64:250-261). The method is motivated by diagnostic measures for osteopetrosis in a study summarized by Hans et al. (The Lancet 1996; 348:511-514). Diagnostic test accuracy is typically defined using the area (or partial area) under the receiver operator characteristic (ROC) curve. If partial area is used, the resulting statistic can be highly subjective because the focus region of the ROC curve corresponds to a set of low false-positive rates that are chosen by the experimenter.

The "coherent majority average" for juries' evaluation processes.

I propose a method to synthesize the performance scores for artistic sports such as rhythmic gymnastics, figure skating, synchronized swimming, and diving by taking into account inter-judge variability, while maintaining all the reliable scores. This procedure is based on the assumption that the majority of the scores in each event are reliable and they relate well to those scores that are closest to them. The method consists of putting scores in order and considering clusters of m consecutive scores, where m is the number of judges making up the simple majority. For each cluster, the differen

Exact analysis of dose response for multiple correlated binary outcomes.

The neurotoxicity of a substance is often tested using animal bioassays. In the functional observational battery, animals are exposed to a test agent and multiple outcomes are recorded to assess toxicity, using approximately 40 animals measured on up to 30 different items. This design gives rise to a challenging statistical problem: a large number of outcomes for a small sample of subjects. We propose an exact test for multiple binary outcomes, under the assumption that the correlation among these items is equal. This test is based upon an exponential model described by Molenberghs and Ryan (1

[Multifactor analysis of the combined use of an antibiotic and a low molecular weight immunomodulator of microbial origin in experimental plague infection].

Multifactorial analysis of the combined effect of rifampicin and a low molecular immunomodulator of microbial origin in experimental plague infection was performed. Synergism of the antibiotic used in the subtherapeutic doses and the immunomodulator was shown. By the results of the study polynomial statistic models of the second order describing the survival rate and average life-span of the experimental animals were developed and nomographs (equal level curves) were plotted for rapid estimating the therapy quantitative parameters. Optimization of the combined use of rifampicin and the immunom

Improving the prediction of Phragmites australis marsh distribution by adding edaphic properties to climatic variables.

Soil properties have been widely shown to influence wetland vegetation distribution. However, the degree to which adding edaphic variables can improve climate-only models remains unclear. Here, we examine whether incorporating soil properties into climate-only Phragmites australis marsh distribution models (SDMs) can improve statistical performance and predictive power. The results show that including soil predictors significantly improved SDMs' performance compared to climate-only models (TSS: + 21.07%, Student's t-tests: t = 24.364, P < 0.01; AUC: + 8.51%, t = 27.209, P < 0.01). Dynamic risk assessment by landmark modeling of the restricted residual mean survival time.

Dynamic risk assessment is an important tool for healthcare providers to inform treatment selection and optimize patient outcomes. Frequent evaluation of clinical status provides invaluable information on adverse event risk and/or treatment urgency, based on the changes in predictive biomarkers. Landmark analysis is a useful approach for dynamic risk assessment that obviates the complexity of jointly modeling longitudinal covariates and time-to-event outcome. Traditional landmark methods often rely on the proportional hazards assumption and assume independent censoring. We propose novel method

Accounting for contact tracing in epidemiological birth-death models.

Phylodynamics bridges the gap between classical epidemiology and pathogen genome sequence data by estimating epidemiological parameters from time-scaled pathogen phylogenetic trees. The models used in phylodynamics typically assume that the sampling procedure is independent between infected individuals. However, this assumption does not hold for many epidemics, in particular for such sexually transmitted infections as HIV-1, for which contact tracing schemes are included in health policies of many countries. We extended phylodynamic multi-type birth-death (MTBD) models with contact tracing (CT

Additive hazard causal model with a binary instrumental variable.

The causal effect of a treatment on a censored outcome is often of fundamental interest and instrumental variable (IV) is a useful tool to tame bias caused by unmeasured confounding. The two-stage least squares commonly used for IV analysis in linear regression have been developed for regression analysis in a survival context under an additive hazards model. In this work, we study a distinctive binary IV framework with censored data where the causal treatment effect is quantified through an additive hazard model for compliers. Employing the special characteristics of the binary IV and adapting

Categorization of continuous covariates and complex regression models-friends or foes in intersectionality research.

OBJECTIVES: To reduce health inequities, it is important to identify intersections in characteristics of individuals subject to privilege or disadvantage. Different proposals for that have recently been published. One approach (1) considers models specified with first- and all second-order effects and another (2) the stratification based on multiple covariates; both categorize continuous covariates. A simulation study was conducted in order to review both methods with regard to identification of intersections showing true differences, rate of false-positive results, and generalizability to ind

Predicting lymphatic filariasis elimination in data-limited settings: A reconstructive computational framework for combining data generation and model discovery.

Although there is increasing importance placed on the use of mathematical models for the effective design and management of long-term parasite elimination, it is becoming clear that transmission models are most useful when they reflect the processes pertaining to local infection dynamics as opposed to generalized dynamics. Such localized models must also be developed even when the data required for characterizing local transmission processes are limited or incomplete, as is often the case for neglected tropical diseases, including the disease system studied in this work, viz. lymphatic filaria

On the evolutionary ecology of multidrug resistance in bacteria.

Resistance against different antibiotics appears on the same bacterial strains more often than expected by chance, leading to high frequencies of multidrug resistance. There are multiple explanations for this observation, but these tend to be specific to subsets of antibiotics and/or bacterial species, whereas the trend is pervasive. Here, we consider the question in terms of strain ecology: explaining why resistance to different antibiotics is often seen on the same strain requires an understanding of the competition between strains with different resistance profiles. This work builds on mode

A flexible parametric approach for estimating continuous-time inverse probability of treatment and censoring weights.

Marginal structural Cox models are used for quantifying marginal treatment effects on outcome event hazard function. Such models are estimated using inverse probability of treatment and censoring (IPTC) weighting, which properly accounts for the impact of time-dependent confounders, avoiding conditioning on factors on the causal pathway. To estimate the IPTC weights, the treatment assignment mechanism is conventionally modeled in discrete time. While this is natural in situations where treatment information is recorded at scheduled follow-up visits, in other contexts, the events specifying the

[Calculation of Standardised Unit Costs for the Economic Evaluation of Mental Disorders].

Introduction: Assessing resource utilisation and costs for treating mental disorders in Germany is difficult. This pertains especially to the numerous non-medical services that are financed by a considerable number of payers. To standardise and simplify cost calculation, we created a database of unit costs for the most important services for mental disorders. Method: We performed a literature and internet search to identify services provided to persons with mental disorders. Unit costs were calculated either by means of public statistics or data directly obtained from payers. If data could not
