In order to make high-performance liquid chromatography (HPLC) more widely available at home and in small-scale settings, we have simplified two of its most costly modules, namely the pump and the detector. This should make the setup affordable for home or small laboratory use. A manual HPLC pump was constructed so as to fit into a caulk gun from a local hardware store enabling the generation of 100-150 bar of pressure. A solvent mixture of 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) and dichloroacetic acid (DCAA) is used to dissolve difficultly soluble poly(ethylene 2,6-naphthalate) (PEN). Solutions can be diluted and analyzed in a common size-exclusion chromatography (SEC) eluent, HFIP. The HFIP/DCAA mixture is better at dissolving PEN than either solvent individually and it is easier and safer to work with than phenolic and strongly acidic eluents. The dual-flame thermionic detector for microcolumn liquid chromatography has been improved and optimized for nitrogen sensitivity. The total column effluent is concentrically nebulized and aspirated directly into an air-hydrogen diffusion flame, while detection limits of 1.4 X 10(-11) g nitrogen/sec at the maximum of a Gaussian peak are achieved. Detection linearity spans three orders of magnitude. An example of the analysis of underivatized barbiturate standards is provided.

Size-exclusion, ion-exchange, and reversed-phase chromatography were investigated for use in purification and characterization of Staphylococcal enterotoxin B (SEB), a 28,000 Mr protein associated with Staphylococcal food-borne intoxication. In all approaches chromatography of crude or purified SEB yielded one or more components which displayed toxin activity and contained the 28,000 Mr SEB protein plus other lower-molecular-weight protein species. Reversed-phase chromatography of detergent-treated SEB permitted resolution of the 28,000 Mr protein from lower-molecular-weight components.

The first gas chromatographic analysis of the lower atmosphere of Venus is reported. Three atmospheric samples were analyzed. The third of these samples showed carbon dioxide (96.4 percent), molecular nitrogen (3.41 percent), water vapor (0.135 percent), molecular oxygen [69.3 parts per million (ppm)], argon (18.6 ppm), neon (4.31 ppm), and sulfuir dioxide (186 ppm).
