Globally, the number of patients suffering from a diverse spectrum of kidney diseases, including parenchymal acute kidney injury (AKI), chronic kidney diseases (CKD), which includes diabetic kidney disease (DKD), and other chronic nephropathies, along with malignant renal cell carcinoma (RCC), is rising rapidly. Although they exhibit clinical heterogeneity, these conditions share common pathomechanisms, including tubular epithelial injury, inflammation, and progressive fibrosis, yet lack unified pharmacological targets that can address this spectrum simultaneously. Among these pathways, the Hi

This review article examines the molecular pathways through which the environmental contaminant deoxynivalenol (DON) influences the regeneration and renewal of both intestinal tissues and intestinal stem cells via the Hippo Yes-associated protein (YAP)/transcriptional co-activator with PDZ-binding motif (TAZ) signaling cascade, while also exploring possible clinical intervention approaches. To begin, the review outlines the prevalent issue of DON contamination and its adverse effects on gastrointestinal health, underscoring the necessity for a comprehensive understanding of its implications. PURPOSE: To investigate whether Hippo pathway inhibition via a novel Lats kinase inhibitor (LKI) induces regeneration of retinal pigment epithelium (RPE).
METHODS: An in vitro scratch "injury" on confluent RPE monolayers was produced using the Woundmaker Tool. A 6×6 grid of laser spots was delivered to the central retinae of Dutch Belted rabbits via indirect laser. Following laser injury, eyes received 50 µL intravitreal LKI or vehicle. Fundus photography and optical coherence tomography (OCT) were performed prior to sacrifice for immunohistological analyses.
RESULTS: In vitro RPE demonstrat

Anti-epidermal growth factor receptor (EGFR) therapies are the most recommended first-line treatment for RAS/RAF wild-type unresectable metastatic colorectal cancer (CRC) according to the European Society for Medical Oncology guidelines. However, primary resistance renders this treatment ineffective for almost 40% of patients. Our previous work identified Aurora kinase A (AURKA) as a key resistance driver through non-canonical, Hippo-independent Yes-associated protein 1 (YAP1) activation. However, the role of the other main Hippo coactivator, transcriptional coactivator with PDZ-binding motif

Human heart failure, a leading cause of death worldwide, is a prominent example of a chronic disease that may result from poor cell renewal. The Hippo signaling pathway is an inhibitory kinase cascade that represses adult heart muscle cell (cardiomyocyte) proliferation and renewal after myocardial infarction in genetically modified mice. Here, we investigated an adeno-associated virus 9 (AAV9)-based gene therapy to locally knock down the Hippo pathway gene Salvador (Sav) in border zone cardiomyocytes in a pig model of ischemia/reperfusion-induced myocardial infarction. Two weeks after myocardi

In dividing cells, centrosomes are the primary microtubule-organizing centers and typically duplicate only once per cell division cycle, ensuring that there is just a single centrosome at each mitotic spindle pole. Centrosome amplification presents a paradox: although supernumerary centrosomes can induce chromosomal instability and promote invasive behavior, they may also impede cell proliferation by activating the Hippo-LATS pathway and the PIDDosome-p53 axis. Therefore, cells with supernumerary centrosomes must either tolerate the abnormal centrosome numbers or restore centrosome homeostasis

The terminal cells of the larval Drosophila tracheal system extend dozens of branched cellular processes, most of which become hollow intracellular tubes that support gas exchange with internal tissues. Previously, we undertook a forward genetic mosaic screen to uncover the pathways regulating terminal cell size, morphogenesis, and the generation and maintenance of new intracellular tubes. Our initial work identified several mutations affecting terminal cell size and branch number, and suggested that branch complexity and cell size are typically coupled but could be genetically separated. Pediatric brain tumors are the second most prevalent malignancy in children with therapeutic options remaining challenging due to vulnerability of the developing brain and the significant side effects of current treatment with targeted therapies offering a promising alternative strategy. The Hippo signaling pathway has emerged as a promising molecular approach since it effectively integrates biochemical and mechanical cues to regulate cell growth and survival, tissue integrity and stem cell behaviour through interaction with major signalling pathways such as Wnt-β-catenin, TGF-β/SMAD, PI3K/Akt

BACKGROUND: Tri-ortho-cresyl phosphate (TOCP), a widely used plasticizer, has been shown to impair ovarian function. While tanshinone IIA exhibits ovarian protective effects in aging models, its potential to counteract TOCP-induced ovarian damage and associated signaling mechanisms remains unexplored. This study investigates the therapeutic effects of tanshinone IIA on TOCP-damaged ovaries in mice, with focus on Hippo, AKT, and MAPK pathways.
RESULTS: TOCP exposure (200 mg/kg/d for 28 days) significantly reduced ovarian follicle counts (primordial, preovulatory, and mature follicles) and disr

Hepatocellular carcinoma (HCC) is one of the most fatal and fastest growing malignancies. Recently, nonalcoholic steatohepatitis (NASH), characterized by liver steatosis, inflammation, cell injury (hepatocyte ballooning), and different stages of fibrosis, has emerged as a major catalyst for HCC. Because the STE20-type kinases, MST3 and MST4, have been described as critical molecular regulators of NASH pathophysiology, we here focused on determining the relevance of these proteins in human HCC. By analyzing public datasets and in-house cohorts, we found that hepatic MST3 and MST4 expression was

One of the most widely used types of assisted reproduction technology is the in vitro fertilization (IVF), in which women undergo controlled ovarian stimulation through the administration of the appropriate hormones to produce as many mature follicles, as possible. The most common hormone combination is the co-administration of gonadotropin-releasing hormone (GnRH) analogues with recombinant or urinary-derived follicle-stimulating hormone (FSH). In the last few years, scientists have begun to explore the effect that different gonadotropin preparations have on granulosa cells' maturation and ap

The discovery that the 5'AMP-activated protein kinase (AMPK) serves to link the tumour suppressors LKB1 and the tuberous sclerosis complex and functions to slow macromolecular synthesis through attenuation of the mechanistic target of rapamycin complex 1 revealed a role for AMPK in tumour suppression. On the other hand, the well-recognized role of AMPK in maintaining ATP homeostasis, through suppression of anabolism and promotion of catabolism, as well as the role of AMPK in neutralizing reactive oxygen species, via maintenance of NADPH-dependent reductive capacity, point to tumour-protective

The understanding of osteoarthritis (OA) has gradually evolved from the traditional perspective of it being a mere "wear-and-tear" joint disease to a more comprehensive view that characterizes it as a degenerative joint disease marked by chronic low-grade inflammation and immune imbalance. The progression of OA is closely linked to abnormal mechanical loading and the activation of the innate immune system. This narrative review examines the role of mechanosensory-immune signaling networks in OA, with a specific focus on ion channels as receptors for upstream mechanical and chemical stimuli. Th

Hepatocellular carcinoma (HCC) is the most common primary liver malignancy and a leading cause of cancer-related mortality worldwide. Despite advances in diagnosis and therapy, the prognosis for advanced HCC remains poor due to late-stage diagnosis, high recurrence rates, therapeutic resistance, and pronounced molecular heterogeneity. HCC development is driven by complex somatic gene alterations, epigenetic reprogramming, dysregulated signaling pathways, metabolic changes, and an immunosuppressive tumor microenvironment. Molecular profiling studies have identified key oncogenic pathways involv

BACKGROUND: Serine/threonine kinase 3 (STK3) is recognized as a key regulator in Hippo pathway and a tumor-suppressing gene in various cancer types. However, its non-canonical role has been gradually revealed in cancer development.
METHODS: Our objective is to elucidate the upregulation pattern and molecular mechanisms of STK3 in advancing gastric cancer (GC) progression. The regulation of YAP1 on STK3 was assessed through a combination of bulk and single-cell RNA-sequencing, Western blot, ChIP-qPCR, gene knockout mouse models, and functional rescue assays. The oncogenic roles of STK3 were co

Mammalian sterile 20-like kinase 1 (Mst1), a ubiquitously expressed 60 kDa serine/threonine kinase, plays crucial roles in various cellular processes. Mst1 is activated by autophosphorylation at Thr183 and caspase 3-mediated cleavage, producing a catalytically active 36/37 kDa (or, in some studies, a 34 kDa) fragment under apoptotic conditions. However, whether the 34 and 36/37 kDa fragments observed in different studies are identical remains unclear, given their similar sizes and the specific detection of a 34 kDa fragment in normal mouse liver and intestinal epithelial cells. This study aime

Spinal cord injury is a severe clinical problem, and research searching activity molecular that can promote spinal cord injury repairing is very prevalent. Mst3b can promote repair of damaged peripheral nerves and the optic nerve, but has been rarely reported in spinal cord injury research. Through detecting its expression in different periods of injured spinal cord, we found that the expression of Mst3b was significantly upregulated in injured spinal cord neurons. Increasing Mst3b expression using adenovirus in vivo and in vitro promoted axonal regeneration of spinal cord neurons, which led t

Central nervous system (CNS) tumors are the most common solid malignancy in the pediatric population. These lesions are the result of the aberrant cell signaling step proteins, which normally regulate cell proliferation. Mitogen-activated protein kinase (MAPK) pathways and tyrosine kinase receptors are involved in tumorigenesis of low-grade gliomas. High-grade gliomas may carry similar mutations, but loss of epigenetic control is the dominant molecular event; it can occur either due to histone mutations or inappropriate binding or unbinding of DNA on histones. Therefore, despite the absence of
