Effect of ultraviolet light on the release of neuropeptides and neuroendocrine hormones in the skin: mediators of photodermatitis and cutaneous inflammation.

Ultraviolet (UV) irradiation of the skin causes both inflammation and alterations in the skin immune system. There is increasing experimental evidence that UV-induced skin inflammation is influenced by the sensory nervous system and the neuroendocrine system in the skin. The resulting complex network of cytokines, chemokines, neuropeptides, neuropeptide-degrading enzymes, neurohormones, and other inflammatory mediators mediate photodermatitis and cutaneous inflammation. Neuropeptides such as substance P (SP) and calcitonin gene-related peptide (CGRP) are released from sensory nerves innervatin

Zbtb44-mediated transcriptional repression critically regulates colonic inflammation.

The regulatory mechanisms that maintain colonic immune homeostasis remain incompletely understood. Here we show that genetic deletion of Zbtb44, a previously uncharacterized member of the ZBTB transcription factor family, leads to spontaneous colitis and rectal prolapse. Zbtb44-/- T cells expressed increased IFN-γ, a pro-inflammatory cytokine strongly associated with human inflammatory bowel disease. Mechanistically, Zbtb44 undergoes SUMOylation, which facilitates its interaction with a conserved SUMO-interacting motif in T-bet, enabling binding to the IFN-γ promoter and enhancer to restrain c

Inflammation-Mediated Mechanisms of Arrhythmias After Acute Myocardial Infarction.

Acute myocardial infarction (AMI) remains a leading cause of cardiovascular death. Arrhythmias are the most common complication after AMI and can worsen disease progression and lead to sudden cardiac death (SCD). Therefore, prompt and accurate diagnosis, together with proactive and effective treatment, is crucial. This article reviews the mechanisms of inflammation in post-AMI arrhythmias, including inflammatory cell infiltration, cytokine release, oxidative stress, cardiac structural remodeling, and neuroimmune interactions. Additionally, this review discusses the predictive value of inflamma

MyD88 restricts dysbiosis-mediated inflammation in filaggrin deficient skin.

Atopic dermatitis (AD) is a common inflammatory skin disease associated with epidermal barrier dysfunction, immune dysregulation, and microbial dysbiosis. Loss-of-function mutations in filaggrin, a critical epidermal protein, represent the strongest genetic risk factor for AD and result in compromised skin barrier integrity and altered immune responses. MyD88 is an adaptor protein essential for TLR and IL-1 receptor signaling, with dual roles in promoting inflammation and regulating immune tolerance. However, the function of MyD88 in maintaining skin homeostasis in the context of filaggrin def

The association between Helicobacter pylori and osteoporosis: the mediating effect of inflammation.

BACKGROUND: Osteoporosis, a widespread metabolic bone disorder, leads to diminished bone density, heightened susceptibility to fractures, and overall skeletal fragility. As the world's population continues to age, this condition has emerged as a pressing public health issue. Recent research has explored the possible link between Helicobacter pylori (H. pylori) infection and various systemic diseases beyond the stomach. Yet, the connection between H. pylori and osteoporosis remains hotly debated, with the exact biological mechanisms still unclear. METHODS: The study recruited participants from

The Inflammation-Mediated Bidirectional Relationship Between Cardiovascular Disease and Cancer.

Cancer and atherosclerotic cardiovascular disease (ASCVD) represent two of the leading causes of death worldwide. Increasingly, these two are being recognized as biologically related conditions rather than entirely segregated disease states. In addition to traditional risk factors such as aging, smoking, and obesity, chronic inflammation may be a key factor connecting the two illnesses. Endothelial dysfunction, oxidative stress, plaque progression, and thrombosis are all facilitated by inflammatory signaling in ASCVD. Similar pathways are known to contribute to cancer growth and invasion. Emer

Targeting S100A9-mediated inflammation: a novel therapeutic approach for CLL.

Chronic lymphocytic leukemia (CLL) presents challenges in treatment despite advancements in targeted therapies, often facing resistance or relapse. Chronic inflammation plays a significant role in CLL biology, with heightened inflammatory responses and immune dysfunction. Elevated levels of inflammatory cytokines support this notion. Activating signaling pathways such as NF-κB, Phosphoinositide 3-kinase delta (PI3Kδ), and MAPK via B-cell receptors and CD40 confers advantages to leukemic lymphocytes. Our research focuses on the proinflammatory protein S100A9 in CLL progression. We previously de

Lipid-mediated resolution of inflammation and survival in amyotrophic lateral sclerosis.

Neuroinflammation impacts on the progression of amyotrophic lateral sclerosis (ALS), a fatal neurodegenerative disorder. Specialized pro-resolving mediators trigger the resolution of inflammation. We investigate the specialized pro-resolving mediator blood profile and their receptors' expression in peripheral blood mononuclear cells in relation to survival in ALS. People living with ALS (pwALS) were stratified based on bulbar versus limb onset and on key progression metrics using a latent class model, to separate faster progressing from slower progressing ALS. Specialized pro-resolving mediato

JAG2 AS A KEY MEDIATOR IN PORPHYROMONAS GINGIVALIS-INDUCED PERIODONTAL INFLAMMATION.

Porphyromonas gingivalis (P. gingivalis) is a key pathogen in chronic periodontitis, but its exact pathogenic mechanisms remain unclear. This study aimed to investigate the transcriptomic changes induced by P. gingivalis and identify potential key pathways involved in periodontal pathogenesis. We performed high-throughput sequencing and bioinformatic analyses on oral epithelial cell exposed to P. gingivalis. Differential gene expression analysis revealed 106 significantly altered genes (p<0.05, |log2FC|>1). KEGG pathway enrichment analysis highlighted the involvement of TNF signaling, Notch si

TLR-induced STK25 activation promotes IRF5-mediated inflammation.

The transcription factor interferon regulatory factor 5 (IRF5) functions as an important mediator of the inflammatory response downstream of MyD88-dependent TLRs. Whereas dysregulation of IRF5 activity has been implicated in the development of numerous autoimmune diseases including systemic lupus erythematosus, the factors that modulate TLR-induced IRF5 post-translational modifications are poorly understood. The focus of this study was to identify novel kinases in TLR-MyD88-IRF5 signaling. We performed a kinome-wide siRNA screen in human THP-1 monocytic cells and identified serine/threonine pr

Piezo1 at the crossroads: Mediating inflammation and mechanical stress in joint disorders.

Piezo1 is a mechanosensitive ion channel, which plays a pivotal role in translating mechanical stress into cellular signaling, thereby influencing inflammatory responses and tissue remodeling in joint disorders. Current evidences showed that Piezo1 functions as a mechanotransducer and amplifier of inflammation across joint disorders including osteoarthritis (OA), rheumatoid arthritis (RA), ankylosing spondylitis (AS), and intervertebral disc degeneration (IVDD). In OA, Piezo1 mediates chondrocyte apoptosis and matrix degradation via calcium influx, NF-κB/MAPK activation, and ferroptosis, formi

CXCL16 mediates nociception and inflammation in murine post-traumatic osteoarthritis.

UNLABELLED: This study investigates the role of the chemokine CXCL16 and its receptor, CXCR6, in post-traumatic osteoarthritis (PTOA) and joint nociception, highlighting the potential of targeting the CXCL16-CXCR6 axis for therapeutically managing joint inflammation and pain. Following joint injury in mice, the CXCL16-CXCR6 signaling axis is activated in synovium, driven by synovial fibroblasts and macrophages. Human OA synovium also exhibited increased CXCL16 and CXCR6 gene expression. CXCL16 stimulated a pro-inflammatory response in fibroblasts and macrophages, contrasting with an anti-infla

INTEGRIN FUNCTION IN LEUKOCYTE-MEDIATED INFLAMMATION-ACTINOPATHIES IN IMMUNE DISEASES.

Integrins play a critical role in leukocyte recruitment and activation within inflamed tissues. These heterodimeric cell-surface receptors recognize ligands on vascular endothelium or extracellular matrix to initiate intracellular signals leading to leukocyte adhesion, migration, and activation. The best-described role for integrins is in the leukocyte adhesion cascade, which is the process by which leukocytes exit the blood vasculature and enter the tissues in response to infection or injury. During the adhesion cascade, integrin signaling is required for changes in leukocyte cytoskeletal str

SLAM-family receptors promote resolution of ILC2-mediated inflammation.

Type 2 innate lymphoid cells (ILC2) initiate early allergic inflammation in the lung, but the factors that promote subsequent resolution of type 2 inflammation and prevent prolonged ILC2 activation are not fully known. Here we show that SLAM-family receptors (SFR) play essential roles in this process. We demonstrate dynamic expression of several SFRs on ILC2s during papain-induced type 2 immunity in mice. SFR deficiency exacerbates ILC2-driven eosinophil infiltration in the lung, and results in a significant increase in IL-13 production by ILC2s exclusively in mediastinal lymph nodes (MLN), le

Cisplatin Toxicity Causes Neutrophil-Mediated Inflammation in Zebrafish Larvae.

Cisplatin is an antineoplastic agent used to treat various tumors. In mammals, it can cause nephrotoxicity, tissue damage, and inflammation. The release of inflammatory mediators leads to the recruitment and infiltration of immune cells, particularly neutrophils, at the site of inflammation. Cisplatin is often used as an inducer of acute kidney injury (AKI) in experimental models, including zebrafish (Danio rerio), due to its accumulation in kidney cells. Current protocols in larval zebrafish focus on studying its effect as an AKI inducer but ignore other systematic outcomes. In this study, ci

PPARG-mediated autophagy activation alleviates inflammation in rheumatoid arthritis.

INTRODUCTION: Rheumatoid arthritis (RA) is a systemic inflammatory autoimmune disease characterized by joint inflammation and bone damage, that not only restricts patient activity but also tends to be accompanied by a series of complications, seriously affecting patient prognosis. Peroxisome proliferator-activated receptor gamma (PPARG), a receptor that controls cellular metabolism, regulates the function of immune cells and stromal cells. Previous studies have shown that PPARG is closely related to the regulation of inflammation. However, the role of PPARG in regulating the pathological proce

Brucella mediates autophagy, inflammation, and apoptosis to escape host killing.

Brucellosis is a serious zoonosis caused by Brucella spp. infection, which not only seriously jeopardizes the health of humans and mammals, but also causes huge economic losses to the livestock industry. Brucella is a Gram-negative intracellular bacterium that relies primarily on its virulence factors and a variety of evolved survival strategies to replicate and proliferate within cells. Currently, the mechanisms of autophagy, inflammation, and apoptosis in Brucella-infected hosts are not fully understood and require further research and discussion. This review focuses on the relationship betw

Gasdermin D-mediated neutrophil pyroptosis drives inflammation in psoriasis.

Psoriasis is a multifactorial immune-mediated inflammatory disease. Its pathogenesis involves abnormal accumulation of neutrophils and T-cell-related abnormalities. Pyroptosis is a type of regulated cell death associated with innate immunity, but its role in psoriasis is unclear. In this study, we found that gasdermin D (GSDMD) is higher in human psoriatic skin than that in normal skin, and in imiquimod-induced psoriasis-like mouse skin, the expression of Gsdmd was most significantly altered in neutrophils and Il1b was also mainly expressed in neutrophils. Immunohistochemical staining of seria

Gastrointestinal distress as a potential mediator between stress and periodontal inflammation.

OBJECTIVES: Periodontal disease is multifactorial in its aetiology, which encompasses biopsychosocial contributors, including psychological stress. Gastrointestinal distress and dysbiosis have been associated with several chronic inflammatory diseases yet have rarely been investigated with respect to oral inflammation. Given the implications of gastrointestinal distress on extraintestinal inflammation, this study aimed to evaluate the potential role of such distress as a mediator between psychological stress and periodontal disease. METHODS: Utilizing a cross-sectional, nationwide sample of 8
