Study of line tension and its impact on protein organization in model domain-forming membranes.

Membrane domains, commonly referred to as lipid rafts, play a crucial role in numerous cellular processes; however, the factors governing their stability and associated protein organization remain debated. Here, we employ coarse-grained molecular dynamics simulations to systematically investigate the molecular determinants of interfacial line tension and its role in membrane organization. We first demonstrate that capillary wave theory provides reliable estimates of line tension, provided that boundary fluctuations are adequately sampled. Our results reveal that domain stability is not primari

Cell crowding initiates tumor invasion by triggering a nanoscale topography transition of plasma membranes.

During the progression from epithelial neoplasms to invasive carcinoma, uncontrolled cellular growth within a confined space generates pronounced cell crowding. Despite its prevalence, how cancer cells sense and respond to crowding during the early stages of tumor invasion remains poorly understood. Here, using a spontaneous crowding model that mimics the progressive compressive stress produced by proliferating cells, we show that cell crowding induces an invasive phenotype in cancer cells. This phenotypic switch is accompanied by a nanoscale smooth-to-corrugated topography transition (nSCTT)

Effects of Ethanol and Opioid Receptor Antagonists Naltrexone and LY2444296 on the Organization of Cholesterol- and Sphingomyelin-Enriched Plasma Membrane Domains.

Excessive consumption of alcohol (ethanol (EtOH)) can lead to alcohol use disorder (AUD). While AUD can be managed through behavioral interventions, there is a great need for pharmacological remedies for this condition. To date, only three pharmacotherapeutic treatments for AUD have been approved by both the U.S. Food and Drug Administration and the European Medicines Agency. This is partly due to an incomplete understanding of the intricate molecular mechanisms through which EtOH affects cellular functions. Here, we focus on EtOH effects on nanoscale organization and the lateral dynamics of m

Proteomic analysis of Rta2p-dependent raft-association of detergent-resistant membranes in Candida albicans.

In Candida albicans, lipid rafts (also called detergent-resistant membranes, DRMs) are involved in many cellular processes and contain many important proteins. In our previous study, we demonstrated that Rta2p was required for calcineurin-mediated azole resistance and sphingoid long-chain base release in C. albicans. Here, we found that Rta2p was co-localized with raft-constituted ergosterol on the plasma membrane of C. albicans. Furthermore, this membrane expression pattern was totally disturbed by inhibitors of either ergosterol or sphingolipid synthesis. Biochemical fractionation of DRMs to

Using monomolecular films to characterize lipid lateral interactions.

Membrane lipids are structurally diverse in ways that far exceed the role envisioned by Singer and Nicholson of simply providing a fluid bilayer matrix in which proteins reside. Current models of lipid organization in membranes postulate that lipid structural diversity enables nonrandom lipid mixing in each leaflet of the bilayer, resulting in regions with special physical and functional properties, i.e., microdomains. Central to understanding the tendencies of membrane lipids to mix nonrandomly in biomembranes is the identification and evaluation of structural features that control membrane l

First synthesis of free cholesterol-BODIPY conjugates.

Analogues of cholesterol (compounds 1 and 2) and coprostanol (compound 3) containing the BODIPY fluorophore in the aliphatic tail of the free sterol have been synthesized starting with bisnorcholenic acid, cholenic acid 3beta-acetate, and lithocholic acid, respectively. An ester linkage joining the fluorophore to the sterol nucleus interfered with the ability of the fluorescent sterol to pack with phospholipids in monolayers. However, an analogue in which the linker was devoid of polar atoms exhibited a substantially similar physical behavior to cholesterol in model membranes with respect to l

CD81-receptor associations--impact for hepatitis C virus entry and antiviral therapies.

Tetraspanins are integral transmembrane proteins organized in microdomains displaying specific and direct interactions with other tetraspanins and molecular partners. Among them, CD81 has been implicated in a variety of physiological and pathological processes. CD81 also plays a crucial role in pathogen entry into host cells, including hepatitis C virus (HCV) entry into hepatocytes. HCV is a major cause of liver cirrhosis and hepatocellular carcinoma. HCV entry into hepatocytes is a complex process that requires the coordinated interaction of viral and host factors for the initiation of infect

Effect of the neonicotinoid acetamiprid on molecular events during in vitro boar spermatozoa capacitation.

Acetamiprid (Ace) is a widely used neonicotinoid insecticide whose intensive application in agriculture has raised concerns about its potential effects on the reproductive health of animals and humans. Successful fertilization depends on the integrity of the plasma membrane (PM) of the spermatozoa and the proper progression of the series of events involved in capacitation. The aim of this study was to evaluate the effects of Ace on the capacitation of boar spermatozoa and several associated functional parameters: membrane carbohydrate redistribution, membrane microdomains (MDM), cholesterol ef…

Lipid rafts are primary mediators of amyloid oxidative attack on plasma membrane.

Increasing evidence indicates that cell surfaces are early interaction sites for Abeta-derived diffusible ligands (ADDLs) and neurons in Alzheimer's disease (AD) pathogenesis. Our previous data showed significant oxidative damage at the plasma membrane in fibroblasts from familial AD patients with enhanced Abeta production. Here, we report that lipid rafts, ordered membrane microdomains, are chronic mediators of Abeta-induced lipid peroxidation in SH-SY5Y human neuroblastoma cells overexpressing amyloid precursor protein (APPwt) and APPV717G genes and in fibroblasts bearing the APPV717I gene m…

Analysis of the contribution of saturated and polyunsaturated phospholipid monolayers to the binding of proteins.

The binding of peripheral proteins to membranes results in different biological effects. The large diversity of membrane lipids is thought to modulate the activity of these proteins. However, information on the selective binding of peripheral proteins to membrane lipids is still largely lacking. Lipid monolayers at the air/water interface are useful model membrane systems for studying the parameters responsible for peripheral protein membrane binding. We have thus measured the maximum insertion pressure (MIP) of two proteins from the photoreceptors, Retinitis pigmentosa 2 (RP2) and recoverin,

Cyclodextrins in chronic pain: Membrane cholesterol remodeling and neuroimmune signaling.

BACKGROUND: Chronic pain remains challenging to treat, with current therapies offering incomplete relief and significant side effects. This narrative review examines cyclodextrins (CDs) beyond their conventional excipient role, focusing on their emerging function as membrane-active modulators of cholesterol-dependent signaling. METHOD/SCOPE: We integrate preclinical evidence linking CD-associated cholesterol remodeling to altered lipid raft organization, nociceptive ion channel regulation (e.g., TRPV1, TRPA1), and neuroimmune pathways implicated in selected chronic pain conditions. Direct sup

Analysis of lipid raft molecules in the living brain slices.

Neuronal plasma membrane has been thought to retain a lot of lipid raft components which play important roles in the neural function. Although the biochemical analyses of lipid raft using brain tissues have been extensively carried out in the past 20 years, many of their experimental conditions do not coincide with those of standard neuroscience researches such as neurophysiology and neuropharmacology. Hence, the physiological methods for lipid raft analysis that can be compatible with general neuroscience have been required. Herein, we developed a system to physiologically analyze ganglioside

Role of lipids in virus replication.

Viruses intricately interact with and modulate cellular membranes at several stages of their replication, but much less is known about the role of viral lipids compared to proteins and nucleic acids. All animal viruses have to cross membranes for cell entry and exit, which occurs by membrane fusion (in enveloped viruses), by transient local disruption of membrane integrity, or by cell lysis. Furthermore, many viruses interact with cellular membrane compartments during their replication and often induce cytoplasmic membrane structures, in which genome replication and assembly occurs. Recent stu

Disrupting Lipid Raft Microdomains to Block Polyploid Giant Cancer Cell Budding and Enhance Radiotherapy Response.

Radiotherapy failure often arises from tumor repopulation by treatment-resistant cancer cells. Disrupting these lipid raft structures-via statins or anti-PCSK9 antibodies-impairs budding, suppresses PGCC-derived tumor repopulation, and enhances radiosensitivity in vitro and i …

Role of Gangliosides in Peripheral Pain Mechanisms.

Gangliosides are abundantly occurring sialylated glycosphingolipids serving diverse functions in the nervous system. Membrane-localized gangliosides are important components of lipid microdomains (rafts) which determine the distribution of and the interaction among specific membrane proteins. Different classes of gangliosides are expressed in nociceptive primary sensory neurons involved in the transmission of nerve impulses evoked by noxious mechanical, thermal, and chemical stimuli. Gangliosides, in particular GM1, have been shown to participate in the regulation of the function of ion channe

A mutation within the transmembrane domain of melanosomal protein Silver (Pmel17) changes lumenal fragment interactions.

Melanocytes synthesize and store melanin within tissue-specific organelles, the melanosomes. Melanin deposition takes place along fibrils fo … d interactions and disulfide bond-mediated associations of lumenal domains. Thus, partitioning into membrane microdomains and effects on conformation explain how the transmembrane region may contribute to the structural integrity of Silver/Pmel17 oligomers or influence toxic, amyloidogenic properties.

Translational diffusion of individual class II MHC membrane proteins in cells.

Single-molecule epifluorescence microscopy was used to observe the translational motion of GPI-linked and native I-E(k) class II MHC membrane proteins in the plasma membrane of CHO cells. The purpose of the study was to look for deviations from Brownian diffusion that might arise from barriers to this motion. Detergent extraction had suggested that these proteins may be confined to lipid microdomains in the plasma membrane. The individual I-E(k) proteins were visualized with a Cy5-labeled peptide that binds to a specific extracytoplasmic site common to both proteins. Single-molecule trajectori

The way we view cellular (glyco)sphingolipids.

Mammalian cells synthesize ceramide in the endoplasmic reticulum (ER) and convert this to sphingomyelin and complex glycosphingolipids on the inner, non-cytosolic surface of Golgi cisternae. From there, these lipids travel towards the outer, non-cytosolic surface of the plasma membrane and all membranes of the endocytic system, where they are eventually degraded. At the basis of the selective, anterograde traffic out of the Golgi lies the propensity of the sphingolipids to self-aggregate with cholesterol into microdomains termed 'lipid rafts'. At the plasma membrane surface these rafts are tho

Entry of the lymphogranuloma venereum strain of Chlamydia trachomatis into host cells involves cholesterol-rich membrane domains.

Chlamydiae are bacterial pathogens which develop strictly inside the epithelial cells of their hosts. The mechanism used by chlamydiae to enter cells is not well characterized; however, it is thought to consist of a receptor-mediated process. In addition, the formation of clathrin-coated pits appears to be dispensable for chlamydiae to be internalized by host cells. Clathrin-independent endocytosis has recently been shown to occur through cholesterol-rich lipid microdomains, which are characterized by detergent insolubility. In the present study, we investigated whether these lipid domains pla
