Homologative Radical 1,3-Dicarbofunctionalization of Unactivated Alkenes Enabled by Sequential Functional Group Migrations.

Despite significant advances in conventional radical-mediated 1,2-difunctionalization of alkenes over the past decade, homologative 1,3-difunctionalization remains largely underdeveloped. Herein, we report a novel photoinduced homologative 1,3-dicarbofunctionalization of unactivated alkenes via one-carbon chain elongation, achieved through sequential functional group migrations. By employing strategically designed tertiary alcohols and heteroarylsulfones as versatile coupling partners, this method affords structurally diverse, densely substituted aliphatic ketones and medium-sized cyclic scaff

α-Trifluoromethyl alkenes as fluorinated linchpins in photochemical C-H/C-C functionalization.

Photochemical activation of C-H and C-C bonds has emerged as a powerful strategy for accessing fluorinated molecules from simple feedstocks under mild conditions. In this context, α-trifluoromethyl alkenes have evolved into versatile fluorinated linchpins, because radical addition to these activated alkenes can be directed toward either defluorinative alkylation to furnish gem-difluoroalkenes or hydroalkylation to give trifluoromethylated alkanes. This review highlights representative photochemical studies that illustrate how radical generation through C-H and C-C activation, together with con

Cr-catalyzed difluoromethylation of α-trifluoromethyl alkenes for modular synthesis of polyfluorinated compounds.

We report a chromium-catalyzed allylic difluoromethylation of α-trifluoromethyl alkenes, allowing one-step access to polyfluorinated compounds characterised by both gem-difluoroalkene and difluoromethyl groups. The reaction employed inexpensive chromium as the catalyst and commercially available bench-stable difluoromethyl 2-pyridyl sulfone as the difluoromethylation reagent, and proceeded under mild conditions with a broad substrate scope. The mechanism studies performed indicated the involvement of radical species in this chromium-catalyzed difluoromethylation process. Gram-scale reactions a

Visible-light-induced iron-catalyzed atom transfer radical addition of chloroform to alkenes.

An efficient visible-light-driven iron-catalyzed atom transfer radical addition (ATRA) reaction of chloroform to alkenes has been developed. Under the optimized conditions, the reaction affords a range of 1,3,3-trichloroalkanes with high atom economy and tolerance of various functional groups. Preliminary mechanistic experiments support a radical-mediated pathway for this ATRA process. This work establishes a low-cost, sustainable, and green approach for dichloromethylation via the activation of abundant and inexpensive chloroform.

Nickel-Catalyzed 1,2-Arylalkenylation of Unactivated Alkenes Enabled by a Native Hydroxy Group.

Transition-metal-catalyzed dicarbofunctionalization of alkenes represents a powerful strategy for building molecular complexity in a single step. Here we report an unprecedented nickel-catalyzed 1,2-arylalkenylation of unactivated alkenyl alcohols using alkenyl boronates and aryl iodides. This method uniquely leverages the native hydroxyl group as an intrinsic directing handle, eliminating the need for pre-installed auxiliaries and streamlining the synthesis of structurally diverse alkenol derivatives. The reaction proceeds under mild conditions with broad functional group tolerance and excell

NaBr-Mediated Electrochemical Hydration of Alkenes: A Minimalist Synthesis of Anti-Markovnikov Alcohol.

Using inexpensive NaBr as both the mediator and supporting electrolyte and acetonitrile/water as the solvent, a variety of aryl alkenes smoothly proceed under the electrochemical anti-Markovnikov hydration conditions to furnish β-aryl ethanol compounds in good yields. Gram-scale experiments, detailed pathway investigation and the transfomation of the products to arylpropionic acid based nonsteroidal anti-inflammatory drugs (NSAIDs), tiaprofenic acid and suprofen, further demonstrated the applicability of this protocol.

Ni(II)-Catalyzed Site- and Regioselective α-Alkenylation of Cyclic Ketones with Alkenes.

Herein, a nickel-catalyzed α-alkenylation reaction of asymmetric cyclic ketones at the sterically hindered α-position with alkenes is disclosed. The high regioselectivity might originate from the acid-catalyzed enolization reaction, selectively generating the thermodynamically favored enol. The nickel catalyst facilitates the preferential alkenylation of polysubstituted enolates rather than monosubstituted enolates, thereby reversing the conventional regioselectivity of alkenylation at the α-position of ketones. This method encompasses a broad range of applicable substrates of alkenes.

Photoinduced Copper-Catalyzed Enantioselective Alkylalkynylation of Alkenes via Polarity-Matched Hydrogen Atom Transfer.

Achieving precise chemo-, regio-, and stereoselective functionalization of C(sp3)-H bonds in inexpensive and readily accessible chemical feedstocks continues to pose a significant challenge in synthetic chemistry. However, while significant efforts have centered on substrates bearing hydridic C(sp3)-H bonds, enantioselective radical transformations of abundant feedstocks containing protic C(sp3)-H bonds remain underdeveloped. In this work, we report a photoinduced copper-catalyzed enantioselective three-component alkylalkynylation of alkenes with a wide range of chemical feedstocks, including

Cooperative NHC and Photoredox-Catalyzed Radical Azidoacylation of Alkenes to β-Amino Ketones.

The construction of azido-functionalized molecules is a common step in synthesizing nitrogen-based scaffolds of natural products and pharmaceutical compounds. Herein, we report a three-component radical azidoacylation of alkenes through cooperative N-heterocyclic carbene (NHC) and photoredox catalysis. This approach involves single-electron transfer (SET) oxidation of trimethylsilyl azide (TMSN3), followed by radical addition and subsequent coupling between a ketyl radical and a C-centered radical. Manipulation of the azido group yields a series of β-amino ketones. Mechanistic studies support

Trifluoromethylations of (Hetero)arenes and Polarized Alkenes Using Trifluoroacetic Anhydride under Photoredox Catalysis.

A novel trifluoromethylation of (hetero)arenes and polarized alkenes using simple trifluoroacetic anhydride (TFAA) as the low price CF3 source under photoredox catalysis was developed without using additives such as bases, hyperstoichiometric oxidant, or auxiliaries. The reaction showed excellent tolerance, including to some important natural products and prodrugs, even on the gram scale and extended to ketones. This simple protocol provides a practical utilization of TFAA. Several perfluoroalkylations and trifluoromethylation/cyclizations were successfully achieved under identical conditions.

Diphosphine Ligand-Enabled Nickel-Catalyzed Chelate-Assisted Inner-Selective Migratory Hydroarylation of Alkenes.

The precise control of the regioselectivity in the transition metal-catalyzed migratory hydrofunctionalization of alkenes remains a big challenge. With a transient ketimine directing group, the nickel-catalyzed migratory β-selective hydroarylation and hydroalkenylation of alkenyl ketones has been realized with aryl boronic acids using alkyl halide as the mild hydride source for the first time. The key to this success is the use of a diphosphine ligand, which is capable of the generation of a Ni(II)-H species in the presence of alkyl bromide, and enabling the efficient migratory insertion of al

Enantioselective Copper-Catalyzed Intermolecular Cyanobenzoyldifluoromethylation of Alkenes: Access to Chiral β-Difluoroacyl Nitriles.

A novel asymmetric copper-catalyzed intermolecular cyanobenzoyldifluoromethylation of alkenes with iododifluoromethyl ketones and TMSCN has been reported, which provides a particularly valuable route to access chiral β-difluoroacyl nitriles with excellent enantioselectivities. The method permits the efficient cyanation of varied β-difluoroacyl-benzylic radicals in mild conditions with high functional group tolerance. The reaction proceeds through a radical pathway. In order to get insight into the stereochemical outcome, computational mechanistic studies were conducted.

Hydroalkylation of Aryl Alkenes with Organohalides Catalyzed by Molybdenum Oxido Based Lewis Pairs.

Three molybdenum(VI) dioxido complexes [MoO2(L)2] bearing Schiff base ligands were reacted with B(C6F5)3 to afford the corresponding adducts [MoO{OB(C6F5)3}(L)2], which were fully characterized. They exhibit Frustrated Lewis-Pairs reactivity when reacting with silanes. Especially, the [MoO{OB(C6F5)3}(L)2] complex with L=2,4-dimethyl-6-((phenylimino)methyl)phenol proved to be active as catalyst for the hydroalkylation of aryl alkenes with organohalides and for the Atom-Transfer Radical Addition (ATRA) of organohalides to aliphatic alkenes. Atom-Transfer Radical Addition to Unactivated Alkenes by using Heterogeneous Visible-Light Photocatalysis.

Heterogeneous visible-light photocatalysis represents an important route toward the development of sustainable organic synthesis. In this study visible light-induced, heavy metal-free atom-transfer radical addition to unactivated terminal olefins is carried out by using the combination of heterogeneous titanium dioxide as photocatalyst and a hypervalent iodine(III) reagent as co-initiator. The reaction can be applied to a range of substrates with good functional-group tolerance under very mild conditions. In addition to a number of commonly used atom-transfer reagents, the relatively challengi

Chemo- and Stereoselective Semireduction of Alkynes to E-Alkenes Catalyzed by Nonheme Iron Enzymes.

The semireduction of alkynes represents a powerful yet challenging transformation that requires simultaneous control of stereoselectivity and chemoselectivity. Currently, synthetic methods for the trans-reduction of alkynes to E-alkenes remain limited, particularly in the realm of biocatalysis. Herein, we report the repurposing of a nonheme iron dioxygenase for the trans-reduction of alkynes. Through screening and directed evolution, we obtained variants of gentisate 1,2-dioxygenase that enable the preparation of structurally diverse E-alkenes in high yields (up to 99% yield) with excellent st

Cobaloxime-catalysed regiodivergent hydrogen atom transfer for alkenyl and allylic carbamoylation with branched alkenes.

Hydrogen atom transfer (HAT) represents a fundamental reaction in synthetic chemistry, with ongoing innovations aimed at improving its selectivity and efficiency. Leveraging the unique reactivity of cobaloxime, we developed a regioselective HAT process that enables divergent carbamoylation of branched alkenes. Through modulation of the electronic and steric properties of the ligands, this cobalt complex achieves precise discrimination between hydrogen atoms exhibiting minor differences in bond dissociation energies (BDEs), affording vinyl and allylic amides with predictable selectivity. Photocatalytic Aminocarboxylation of Unactivated Alkenes and Heteroarenes with CO2 toward β-Amino Acid Derivatives.

β-Amino acids represent vital structural motifs in bioactive molecules, finding broad utility in organic synthesis, pharmaceuticals, and materials science. Although CO2 has been widely exploited as a C1 carboxyl source for the synthesis of carboxylic acids, the direct aminocarboxylation of unactivated alkenes with CO2 to afford β-amino acids remains largely unexplored and faces formidable challenges. Herein, we report the first photocatalytic aminocarboxylation of unactivated alkenes and heteroarenes using CO2 as the ideal one-carbon feedstock, enabling the efficient synthesis of diverse valua

Photoreductive Coupling of Unactivated Ketones and Alkenes via Kinetic Suppression of Ketyl Radical Overreduction.

A direct photoinduced electron transfer strategy that employs phenolate anions as photocatalysts for the efficient generation of ketyl radicals was developed. The photoexcited phenolate catalyst serves as the sole potent reductant in the reaction system, and its inherent transient nature kinetically suppresses overreduction of the transiently formed ketyl radical intermediates. This precise kinetic control preserves the high reactivity of ketyl radicals, enabling their efficient coupling with unactivated aliphatic alkenes and alkynes under strongly reducing conditions.

Photoredox-Catalyzed 1,4-Dichloromethyldimerization of Alkenes with Chloroform: Access to Polychlorinated Vicinal Diaryl Alkanes.

A visible-light-mediated strategy is reported for the direct synthesis of polychlorinated vicinal diaryl alkanes from aryl alkenes and chloroform. In this approach, two haloalkyl radicals generated from chloroform via halogen atom transfer (XAT) and direct single electron transfer (SET) within the same photoredox catalysis cycle enable the 1,4-dichloromethyldimerization of alkenes. Besides chloroform, this strategy is applicable to carbon tetrachloride, bromotrichloromethane, and α-bromo carboxylic esters, yielding corresponding 1,4-disubstituted vicinal diaryl alkanes. Diverse polychlorinated
