INTRODUCTION AND OBJECTIVES: Many of the symptoms of patients with lactose intolerance are due to fermentation of undigested lactose in the colonic lumen, which may also lead to inflammatory cell changes in the colonic mucosa. The objective of our project was to understand the histopathological changes involving infiltration of eosinophils and mast cells in the colonic mucosa of children with lactase deficiency (LD).
METHODS: In this retrospective study we studied colonic mucosa of children and adolescents with LD to determine if any pathological changes or inflammatory cell changes were pres…

The physiological decline of lactase production in adulthood, in some individuals, is responsible for the so-called "Lactose Intolerance." This clinical syndrome presents with gastrointestinal and non-gastrointestinal symptoms following the consumption of dairy containing food. Lactose intolerance can be evaluated by means of the Lactose Breath Test (phenotype) and/or genetic evaluation of lactase-gene polymorphism (genotype). A comparison of the two tests was carried out in a large number of symptomatic adult subjects, which are selected and not representative of the general population. Congr…

BACKGROUND: Lactase activity declines during childhood in the majority of human populations leading to adult-type hypolactasia (AtH). C/T-13910 and G/A-22018 single nucleotide polymorphisms (SNPs) have been suggested to be associated with AtH in different human populations. Coeliac disease (CD) is an autoimmune condition characterized by damage to intestinal cells leading to ultimate deterioration.
AIM: This study investigated the association between coeliac disease (CD) and SNPs leading to AtH in children from North India.
SUBJECTS AND METHODS: Intestinal biopsies and saliva samples were ob…

Lactase preparations were purified from cell-free extracts of Kluyveromyces lactis to homogeneity, as determined by disc SDS polyacrylamide gel electrophoresis. A combination of techniques including ammonium sulfate or acetone precipitation and hydroxyapatite chromatography was used for the purification of the enzyme. The enzyme has a pH optimum of 6.5-7.0 with Kms' of 1.25 and 28mM for the substrates O-nitrophenylgalactopyranoside and lactose respectively. Activation energies for denaturation of enzyme and conversion of substrate to product were determined to be 15.1 and 10.0 Kcal/mol, respec…

In adulthood, the ability to digest lactose, the main sugar present in milk of mammals, is a phenotype (lactase persistence) observed in historically herder populations, mainly Northern Europeans, Eastern Africans, and Middle Eastern nomads. As the -13910∗T allele in the MCM6 gene is the most well-characterized allele responsible for the lactase persistence phenotype, the -13910C > T (rs4988235) polymorphism is commonly evaluated in lactase persistence studies. Lactase non-persistent adults may develop symptoms of lactose intolerance when consuming dairy products. In the Americas, there is no …

BACKGROUND AND AIMS: There have been few reports on lactase deficiency (LD) and lactose intolerance (LI) in Malaysia, which has a peculiar mix of three distinct major Asian races-Malay, Chinese, and Indian. The aim of this study was to determine the prevalence of LD and LI in a young multiethnic Malaysian population.
METHODS: Lactase activity was measured with a 13CO2 lactose breath test using an infrared spectrometer. Each subject took 25 g of lactose naturally enriched in 13CO2 together with 250 mL of water after an overnight fast. Breath samples were collected at baseline and at 15-min int…

Lactase persistence (LP), the dominant Mendelian trait conferring the ability to digest the milk sugar lactose in adults, has risen to high frequency in central and northern Europeans in the last 20,000 years. This trait is likely to have conferred a selective advantage in individuals who consume appreciable amounts of unfermented milk. Some have argued for the "culture-historical hypothesis," whereby LP alleles were rare until the advent of dairying early in the Neolithic but then rose rapidly in frequency under natural selection. Others favor the "reverse cause hypothesis," whereby dairying …

The article presents the authors' and literature data on the prevalence of the lactase deficiency among the nations of the Finn-Ugric language group (Finns, Karelians, Vepses, Izhors, Estonians), Eastern Slavs (Russians, Ukrainians, Belorussians) living in the northwestern region of Russia. The lactase deficiency has been established: among Russians--16%, Belorussians--15%, Ukrainians--13%, Finns--22%, Karelians--20%, Vepses--20%, Izhors--20%, Estonians--23%. Confirmations of the cultural and historical hypothesis of the lactase deficiency prevalence have been found. It has been shown that und…

The development of immune-mediated diabetes in BB rats may involve a defect of the gastrointestinal tract (GI), as suggested by increased gut permeability. This study aimed at measuring invertase, maltase, lactase, and peroxidase activities in the duodenum of diabetesprone BioBreeding (BBdp) rats and control BioBreeding rats (BBc) given free access to NIH-07 diet up to the time of killing at 60 66 d of age. After washing the entire small intestine, the duodenal mucosa was scraped off in the first 5-cm segment from the pylorus and frozen in distilled water. Invertase, maltase, and lactase activ…

Carbohydrates are hydrolyzed in the intestinal lumen by specific enzymes to monosaccharides before transport across the brush border membran … t is associated with an absence or drastically reduced levels of an intestinal enzyme, in this case lactase-phlorizin hydrolase (LPH). The pattern of reduction of activity has been termed late onset of lactase deficiency or adult type hypolactasia. It was thought that the regulation of LPH was post-translational and was associated with altered structural …

The objective of this study was to evaluate the effects of lactase on lactose malabsorption and its intolerance symptoms, as well as the available way to improve lactose absorption. Healthy adults with a history of lactose intolerance were screened by 25 g lactose tolerance test. The individuals with higher H2 expired and/or lactose intolerance symptoms were selected as the subjects. Subjects were challenged twice with "400 ml low fat milk" and "400 ml low fat milk + 9000Fcc lactase" separately in 3 days interval. The breath H2 concentration and intolerance symptoms were tested in 4 hours afte…

In 28 patients the existence of deficiency in lactase activity was examined indirectly with lactose tolerance test (LTT). Patients were divided in three groups. The first group consisted of 12 patients with confirmed diagnosis of "Irritable-colon syndrome". The second was composed of 6 patients suffering from ulcerative colitis. The third examined group was control group made of healthy persons without any gastrointestinal or metabolic disorders. To exclude the possible of monosaccharide malabsorption, all patients underwent glucose-tolerance test (GTT). Milk tolerance questionnaire was perfor…

Lactase-phlorizin hydrolase, which hydrolyzes lactose, the major carbohydrate in milk, plays a critical role in the nutrition of the mammalian neonate. Lactose intolerance in adult humans is common, usually due to low levels of small intestinal lactase. Low lactase levels result from either intestinal injury or (in the majority of the world's adult population) alterations in the genetic expression of lactase. Although the mechanism of decreased lactase levels has been the subject of intensive investigation, no consensus has yet emerged. Recent studies have begun to define the cellular and mole…
