Metformin (N,N-dimethylguanylguanidine) is one of the most prescribed drugs with pleiotropic, exerted in part by not fully elucidated mechanisms of action. We developed and validated a gas chromatography-mass spectrometry (GC-MS) method for the quantitative analysis of metformin (metformin-d0) in 10-µL aliquots of human serum and urine using N,N-[dimethylo-2H6]guanylguanidine (metformin-d6) as the internal standard. The acylation proceeds smoothly in the presence of Cu(NTf2)2 (0.1 mol%) using carboxylic anhydrides, acyl chlorides, or isopropenyl acetate as acylating agents. Direct access to benzo-fused aza-macrolides was successfully achieved through iridium-catalysed intermolecular decarboxylative coupling of vinylethylene carbonates with isatoic anhydrides under relatively mild reaction conditions. Notably, this reaction proceeded through sequential allyl-amination/macrolactonization upon extrusion of CO2. Moreover, favourable fluorescence properties could be observed in the title macrocyclic products.

The trifluoromethylation of benzoic acids with TMSCF3 was achieved through nucleophilic substitution with the use of anhydrides as an in situ activating reagent. Under the reaction conditions, a wide range of carboxylic acids including the bioactive ones worked well, thus providing a facile and efficient method for preparing aryl trifluoromethyl ketones from the readily available starting materials.

The cross-coupling of epoxides with acyl chlorides or anhydrides by a nickel/titanocene dual catalytic system is established. A variety of synthetically useful β-hydroxy ketones were obtained in good to high yields by using modified pyridine-oxazoline ligand. Twelve new N-aminosuccinimides were synthesized by the condensation of hydrazines with succinic anhydrides in glacial acetic acid. The compounds were evaluated in the maximal electroshock seizure and subcutaneous pentylenetetrazol seizure threshold tests for anticonvulsant activity and in the rotorod test for neurotoxicity in mice. The lowest dose at which several of the compounds exhibited anticonvulsant activity was 300 mg/kg.

The acetylation of tert-butanol with acetic anhydride catalyzed by 4-(dimethylamino)pyridine (DMAP) has been studied at the Becke3 LYP/6-311 + G(d,p)//Becke3 LYP/6-31G(d) level of theory.
