The expression of prophenoloxidase (proPO) transcript in the tissues of the Pacific white shrimp Litopenaeus vannamei was studied by Northern blot, RT-PCR and in situ hybridisation. Northern blot analysis reveals that proPO was constitutive mainly in shrimp haemocytes. Its transcripts, when detected with RT-PCR, were widely distributed in haemocytes, gill, heart, lymphoid organ, stomach, midgut, anterior midgut caecum and ganglion. OBJECTIVE: To identify the mutations of the tyrosinase gene (TYR) and P gene in patients with oculocutaneous albinism (OCA).
METHODS: Polymerase chain reaction (PCR) and denaturing high performance liquid chromatography (DHPLC) were applied to detect the mutations in all exons of TYR gene and P gene. Then DNA sequencing and restriction endonuclease analysis were used to confirm the mutations detected by DHPLC. Novel mutations were screened in 100 unrelated persons with normal phenotypes to exclude the possibility of polymorphism.

One conclusion reached from a study of a case of tyrosinase negative albinism was that the structure of melanizing apparatus was normal. Its regulation and ability to be stimulated was functionally normal also. The characteristic defect in albinism must be somewhere in the availability or inhibition of tyrosinase based on a genetically induced structural defect of the protein pattern.

Visual evoked potential testing was performed on 15 Negro carriers of the gene for tyrosinase positive oculocutaneous albinism in order to detect whether they have the same visual pathway decussation anomalies as do homozygotes. No subject showed 01-02 asymmetry on monocular testing, indicating that decussation follows the normal pattern. It is concluded that visual evoked potential testing is probably not useful in the detection of Negroes heterozygous for the gene for tyrosinase positive oculocutaneous albinism.
