Infusional cisplatin plus cyclophosphamide in advanced ovarian cancer.

Twenty-one patients with previously untreated advanced ovarian cancer received a two-drug regimen of cisplatin delivered as a systemic infusion at 20 mg/m2/d for 5 days concomitant with oral cyclophosphamide 150 mg/d in divided doses. Courses were repeated at 3- to 4-week intervals. Responses were observed in 14 of 19 evaluable patients (74%) with three complete responses (one pathologically confirmed at second-look laparotomy) and 11 partial responses. Median time to disease progression in the responders was 12 months (range, 4 to 24 months with four patients maintaining remission at 11, 13,

Wegener's granulomatosis: successful treatment with high-dose cyclophosphamide.

A 30-year-old male taxi driver developed extensive purpuric necrotic lesions all over his body. He had a consolidated shadow in the right upper lung field, proteinuria with microscopic hematuria, and granulomatous change in the nasal mucosa. The standard regimen of prednisolone alone or in combination with cyclophosphamide showed only a limited effect on the lesion. However, weekly intravenous injections of a high dose of cyclophosphamide (500 mg/week) in combination with 60 mg/day of prednisolone exhibited dramatical suppression of the disease activity.

Rituximab plus fludarabine and cyclophosphamide prolongs progression-free survival compared with fludarabine and cyclophosphamide alone in previously treated chronic lymphocytic leukemia.

PURPOSE: Rituximab, a monoclonal antibody that targets the CD20 cell surface antigen, has clinical activity in patients with non-Hodgkin's lymphoma and other B-lymphocyte disorders when administered alone or in combination with chemotherapy. Promising results have previously been reported in nonrandomized studies in patients with chronic lymphocytic leukemia (CLL). This trial was designed to compare chemoimmunotherapy with chemotherapy alone in patients with previously treated CLL. PATIENTS AND METHODS: This international, multicenter, randomized trial compared six cycles of rituximab plus fl

A prospective, multicenter, randomized trial comparing steroids and pulse cyclophosphamide versus steroids and oral cyclophosphamide in the treatment of generalized Wegener's granulomatosis.

OBJECTIVE: To investigate the effectiveness and side effects of oral versus pulse cyclophosphamide (CYC) in combination with corticosteroids (CS) in the treatment of systemic Wegener's granulomatosis (WG). METHODS: Patients with newly diagnosed systemic WG were enrolled in a prospective, randomized trial. At the time of diagnosis, prior to randomization, every patient received a daily injection of methylprednisolone for 3 days, followed by daily oral prednisone (1 mg/kg/day) and a 0.7-gm/m2 pulse of CYC. Patients were then randomly assigned to receive either prednisone plus intravenous pulse

Effects of chlordiazepoxide, diazepam and oxazepam on the antitumor activity, the lethality and the blood level of active metabolites of cyclophosphamide and cyclophosphamide oxidase activity in mice.

Effects of chlordiazepoxide, diazepam and oxazepam on the antitumor activity and acute toxicity of cyclophosphamide and the level of its active metabolites in the plasma were investigated in mice. Cyclophosphamide was administered 24 h after the final injection of chlordiazepoxide, diazepam or oxazepam (100 mg/kg/d for 3 d, i.p.). Pretreatment with these drugs increased the acute toxicity of cyclophosphamide (300 or 450 mg/kg, i.p.), whereas drugs had no effect on the antitumor activity of cyclophosphamide (100 mg/kg, i.p.) against Ehrlich solid carcinoma. A high level of active metabolites of

Protocatechuic acid and tanshinone IIA improve cyclophosphamide-induced immunosuppression.

Protocatechuic acid (PCA) and tanshinone IIA (TanIIA), the major functional constituents of Phellinus baumii (PB) and Salvia miltiorrhiza (SM), respectively, regulate innate and adaptive immune pathways and converge on nuclear factor-κB (NF-κB) signaling, providing a mechanistic basis for cooperative immune enhancement. Based on these characteristics, this study evaluated the immunomodulatory effects of a PB-SM complex and examined whether combined treatment with PCA and TanIIA could cooperatively enhance immune recovery. In a cyclophosphamide (CPA)-induced immunosuppressed mouse model, PB-SM

Regulation of cyclophosphamide induced hepatotoxicity by REV-ERBα modifiers.

INTRODUCTION: Cyclophosphamide (CPA) is a widely used broad-spectrum antitumor drug with severe hepatotoxicity. Finding an effective way to mitigate the hepatotoxicity caused by CPA is a challenge in its clinical application. METHODS: In Rev-erbα knockout and wild-type mice, hepatotoxicity was evaluated by ALT, AST, and histopathological scores 4-h post dose of CPA (i.p. 300 mg/kg). CYP2B10 expression and pharmacokinetic behavior of CPA were also detected. SR9009 (i.p. 10 mg/kg) and Berberine (BBR, i.p. 50 mg/kg) were pre-administered to mice. Then, the measurements were carried out following

Graft-versus-Host Disease Prophylaxis with Cyclophosphamide and Cyclosporin.

BACKGROUND: Allogeneic peripheral-blood stem-cell transplantation (SCT) from a matched related donor after myeloablative conditioning is the preferred curative treatment for patients with high-risk blood cancers. The combination of a calcineurin inhibitor and an antimetabolite remains standard care for graft-versus-host disease (GVHD) prophylaxis in these patients. Data from two randomized trials have suggested that post-transplantation cyclophosphamide can reduce the risk of GVHD after SCT from a matched donor when it is added to or replaces the antimetabolite. However, the effects of post-tr

Cyclophosphamide mitigates non-ICANS neurotoxicities following ciltacabtagene autoleucel treatment.

AbstractPotentially fatal non-immune effector cell-associated neurotoxicity syndrome neurotoxicities (NINTs) after B-cell maturation antigen-targeted chimeric antigen receptor (CAR) T-cell therapy have been linked to high CAR T-cell expansion. Cyclophosphamide to ablate CAR T cells is an effective strategy to mitigate steroid-refractory NINTs.

Ameliorative effect of citicoline on cyclophosphamide-induced lung injury.

BACKGROUND: Cyclophosphamide (CP) is known to cause pulmonary injury through free radical production and proinflammatory cytokines activation. Lung damage is associated with significant mortality, primarily due to the development of severe inflammation and pulmonary edema. On the other hand, citicoline, a key intermediate in the phosphatidylcholine biosynthesis pathway, has neurovascular protective and reparative properties. Numerous studies have highlighted the potential antioxidant and antiinflammatory effects of this agent in the context of various pathological conditions. AIM: Our study a

Posttransplant cyclophosphamide: a universal graft versus host disease prophylaxis.

PURPOSE OF REVIEW: The purpose of this review is to outline current graft versus host disease (GvHD) prophylaxis, in the era of posttransplant cyclophosphamide (PTCY), in patients with malignant and nonmalignant hematologic disorders. The original combination of PTCY with a calcineurin inhibitor (CNI) and mycophenolate (MMF), reported from the Johns Hopkins University in Baltimore, was designed for patients receiving a graft from a donor mismatched at one haplotype, so called haploidentical donor (HAPLO). In the past decade, PTCY has been widely used in HAPLO transplants worldwide, confirming

Removal efficiency of antineoplastic drug cyclophosphamide by hypochlorous acid.

Hypochlorous acid (HClO), one of the major reactive oxygen species, is obtained by electrolyzing a sodium chloride solution. HClO is a safe and effective disinfectant and decomposing agent widely used as an alternative to sodium hypochlorite (NaClO). In this study, the authors aimed to evaluate the safety and efficiency of HClO generated by electrolyzing sodium chloride as a decontaminant. Cyclophosphamide (CPA), an antineoplastic drug, was selected as the model drug, and various solvents (HClO, NaClO, etc.) were compared to identify the solvents that could react with and efficiently decompose

Rhodotorula mucilaginosa ZTHY2 Attenuates Cyclophosphamide-Induced Immunosuppression in Mice.

Rhodotorula mucilaginosa (R. mucilaginosa) can enhance the immune and antioxidant function of the body. However, whether R. mucilaginosa has an immunoregulatory effect on cyclophosphamide (CTX)-induced immunosuppressed animals remains to be clarified. In this study, the R. mucilaginosa ZTHY2 that we isolated from the coastal waters of the South China Sea previously was prepared in order to investigate its immunoprotective effect on CTX-induced immunosuppression in mice, and the effects were compared to those of Lactobacillus acidophilus (LA) (a well-known probiotic). Seventy-two male SPF mice

Therapeutic Potential of Capsaicin against Cyclophosphamide-Induced Liver Damage.

Cyclophosphamide (CPM) is a classical alkylating agent used in different cancer chemotherapy regimens and is restricted due to severe adverse effects, including hepatotoxicity. Natural or plant-derived antioxidants such as capsaicin were utilized in this study to examine the hepatoprotective benefits against cyclophosphamide-induced hepatotoxicity. The rats were divided into five groups: a normal control group, a toxic group (CPM), an intraperitoneal injection of a single dose of 200 mg/kg b.w. on the fourth day, a pretreated group with two doses of CPS (10 mg and 20 mg/kg b.w.) orally for six

Severe Hyponatremia During First Cycle of Cyclophosphamide/Doxorubicin Chemotherapy.

Syndrome of inappropriate anti-diuretic hormone release (SIDAH) is a condition characterized by an unregulated release of anti-diuretic hormone (ADH) resulting in increased water retention and decreased plasma osmolarity. Without regulation, ADH release will cause a significant decrease in plasma sodium concentration and can present with cramping, nausea, vomiting, and in severe cases, seizures, and potentially falling into a comatose state. The causes of SIADH are variable and range from infections, some malignancies to some medications. We report a rare case of SIADH resulting from a single

Potential Protective Effects of Antioxidants against Cyclophosphamide-Induced Nephrotoxicity.

Cyclophosphamide is an alkylating antineoplastic agent, and it is one of the most successful drugs with wide arrays of clinical activity. It has been in use for several types of cancer treatments and as an immunosuppressive agent for the management of autoimmune and immune-mediated diseases. Nowadays, its clinical use is limited due to various toxicities, including nephrotoxicity. Even though the mechanisms are not well understood, cyclophosphamide-induced nephrotoxicity is reported to be mediated through oxidative stress. This review focuses on the potential role of natural and plant-derived

Cyclophosphamide exposure via sweat of patients receiving CHOP therapy.

INTRODUCTION: "Secondary exposure to anticancer drugs" refers to exposure to anticancer drugs after chemotherapy via the patient's urine and other excretions. The necessity of countermeasures against secondary exposure to anticancer drugs has been recently highlighted. Although anticancer drugs are also excreted through sweat, few studies have reported exposure to drug residues via this route. We investigated the amount of cyclophosphamide (CPA) excreted in the sweat of patients receiving CHOP therapy (cyclophosphamide, doxorubicin, vincristine, and prednisone). METHODS: The study population
