Efficacy of a single dose of mebendazole on prevalence and intensity of soil-transmitted nematodes in Zanzibar.

The efficacy of a single-dose of mebendazole to treat intestinal helminths was studied in Pemba Island, Zanzibar, with the view of incorporating it in future control programmes. A single Kato-Katz stool examination was performed on 2,269 individuals from all age groups. 1,883 individuals were treated with 500 mg of mebendazole (250 mg for children below two years) and re-examined one month and four months after therapy. A total of 466 presented themselves for all three surveys and the data has been reported in this study. The overall cure rate for ascariasis was 93.2% and reduction of egg load

Studies on the comparative efficacy of mebendazole, flubendazole and niclosamide against human tapeworm infections.

The efficacy of Mebendazole and Niclosamide was studied in two groups of 24 and 38 cases, respectively of patients suffering from taeniasis. Mebendazole with dose schedule of 200 and 300 mg twice daily for 3 consecutive days showed a cure rate of 71.42% and 92.30%, whereas Niclosamide at the dose rate of 200mg per patient was 94.76% effective. Flubendazole showed a cure rate of 66.66% only. Mebendazole and Niclosamide possess high taeniacidal activity, ability to reduce the clinical symptoms of taeniasis without any side effects. Niclosamide with high activity and excellent tolerance, is a dru

Measurement of Ascaris lumbricoides infection intensity and the dynamics of expulsion following treatment with mebendazole.

Intensity of Ascaris lumbricoides infection was measured in terms of egg counts and worm burden in children 2-10 years of age. The expulsion of A. lumbricoides with a 3-day treatment of mebendazole occurred over 8 days, beginning on the second day of treatment. Ninety-seven percent of the worms were expelled between the second and seventh days. A rapid means of estimating eggs per gram (epg) by the Kato Katz technique correlated well with the method described by Martin & Beaver (1968). In spite of apparent density dependence in egg production, A. lumbricoides egg counts correlated well with wo

[Medical therapy with mebendazole in hydatid cyst disease. A follow-up of 40 cases].

Mebendazole (50-60 mg/kg/day in 7-8 divided doses after fatty rich meals for at least 6 months) was given to 44 patients with cystic hydatid disease of the liver and other organs, about 1/3 having suffered from recurrences, even multiple, before beginning the pharmacological treatment. Fifteen patients underwent operation after a course of mebendazole, and macroscopic and histological degeneration of hydatid cysts was always documented; sixteen subjects were not operated due to their severe general and/or local conditions but ultrasound, radiologic (CT) and clinical evidence of cyst regression

Differential efficacy of mebendazole and albendazole against Necator americanus but not for Trichuris trichiura infestations.

The effect of single oral doses of albendazole 600 mg and mebendazole 1 g given to 56 and 60 men, respectively, with T. trichiura and/or N. americanus infestation has been studied. Both albendazole and mebendazole cured more than 90% of T. trichiura infestations, but only albendazole (95%) and not mebendazole (21%) had a high cure rate for N. americanus infestations. Thus, albendazole is the preferred benzimidazole derivate for the mass treatment of subjects with T. trichiura and/or N. americanus infestations.

Strongyloides ratti and S. stercoralis: the effects of thiabendazole, mebendazole, and cambendazole in infected mice.

The effects of benzimidazole anthelmintics in murine strongyloidiasis were examined. Thiabendazole 50 mg/kg daily produced a 91% reduction in the numbers of Strongyloides ratti larvae in the feces. A similar suppression was seen when thiabendazole was given during the intestinal phase, but no effect was noted when the drug was administered during the phase of larval migration. Thiabendazole had no effect on larvae in the skin or lungs, did not inhibit maturation of worms, and did not expel adult worms from the gut, but did reduce fecundity of adult worms in the intestines by 84%. Mebendazole a

Pro-oxidant effects of Mebendazole in albino rats experimentally infected with Trichinella spiralis.

Trichinellosis treated with Mebendazole often leads to complications in the course of the disease in humans and animals as a result of intoxication and hyper-sensitization of an organism due to the massive destruction of parasites. This study was conducted to research Mebendazole incidence on lipid peroxidation processes (LPP) in rats' blood in Trichinella spiralis-infected and parasite-free albino rats. The research was conducted to evaluate erythrocyte superoxide dismutase (SOD) activity and malonic dialdehyde (MDA) concentration in blood serum. The parameters of the SOD-MDA system in infect

High oral doses of mebendazole interfere with growth of larval Echinococcus multilocularis lesions.

The natural development of the larval stage of Echinococcus multilocularis in man has been studied in 7 patients after presumed radical operation 2 8/12-11 2/12 years prior to detection of a relapse. The volumes of the recurring lesions were assessed by CT-scanning, and assuming linear growth a median increase of 14.8 ml/year (range 3.8-220 ml/year) was calculated. In 6 patients treated for a median duration of 4 5/12 years with high oral doses of mebendazole a median growth rate of -3.0 ml/year (range-470- + 10.2 ml/year) was found, which differed significantly from the natural growth rate (P

Anthelmintic activity of mebendazole against induced and naturally occurring helminth infections in cats.

A formulation of mebendazole was used to determine the optimal dosage level against induced and/or naturally occurring infections of Toxocara cati, Ancylostoma tubaeforme, and Taenia taeniaeformis in cats. Amounts of 11 mg/kg of body weight/day or 22 mg/kg/day were given for 2, 3, or 4 days. Feces were collected for 7 days and were examined for expelled parasites. Cats were necropsied to recover retained parasites. A regimen of 22 mg/kg/day for 3 days was determined to be optimal with efficacies of 100%, 99%, and 50% against T cati, A tubaeforme, and Taenia taeniaeformis, respectively.

Efficacy and safety of albendazole plus ivermectin, albendazole plus mebendazole, albendazole plus oxantel pamoate, and mebendazole alone against Trichuris trichiura and concomitant soil-transmitted helminth infections: a four-arm, randomised controlled trial.

BACKGROUND: Existing anthelmintic drugs (eg, albendazole and mebendazole) have low efficacy against the intestinal nematode species Trichuris trichiura and the drug pipeline is exhausted. We aimed to investigate the strategy of combination chemotherapy with existing drugs to establish whether their efficacy could be enhanced and broadened. METHODS: In this randomised controlled trial, we compared three drug combinations and one standard drug alone in children aged 6-14 years in two schools on Pemba Island, Tanzania infected with T trichiura and concomitant intestinal nematodes. We assigned ch

Therapeutic efficacy of mebendazole and artemisinin in different phases of trichinellosis: a comparative experimental study.

The present work aimed at studying the efficacy of mebendazole (MBZ) compared to artemisinin (ART) for the treatment of trichinellosis at various phases of infection. Seventy Swiss albino mice were orally infected by 300 Trichinella spiralis (T. spiralis) larvae. Mice were divided into infected untreated control group and infected groups treated with 50 mg kg-1 MBZ and 300 mg kg-1 ART for three and five consecutive days, respectively, at the enteral phase [2-4 days post infection (PI)], invasive phase (10-12 days PI) and encapsulated phase (28-30 days PI). All mice were sacrificed 35-42 days P

Effective treatment of diverse medulloblastoma models with mebendazole and its impact on tumor angiogenesis.

BACKGROUND: Medulloblastoma is the most common malignant brain tumor in children. Current standard treatments cure 40%-60% of patients, while the majority of survivors suffer long-term neurological sequelae. The identification of 4 molecular groups of medulloblastoma improved the clinical management with the development of targeted therapies; however, the tumor acquires resistance quickly. Mebendazole (MBZ) has a long safety record as antiparasitic in children and has been recently implicated in inhibition of various tyrosine kinases in vitro. Here, we investigated the efficacy of MBZ in vario

Enhanced bioavailability and anthelmintic efficacy of mebendazole in redispersible microparticles with low-substituted hydroxypropylcellulose.

BACKGROUND: Mebendazole (MBZ) is an extremely insoluble and therefore poorly absorbed drug and the variable clinical results may correlate with blood concentrations. The necessity of a prolonged high dose treatment of this drug increases the risk of adverse effects. METHODS: In the present study we prepared redispersible microparticles (RDM) containing MBZ, an oral, poorly water-soluble drug, in different proportions of low-substituted hydroxypropylcellulose (L-HPC). We investigated the microparticulate structures that emerge spontaneously upon dispersion of an RDM in aqueous medium and eluci
