A method is described whereby quantum dot (QD) nanoparticles can be used for correlative immunocytochemical studies of human pathology tissue using widefield fluorescence light microscopy and transmission electron microscopy (TEM). To demonstrate the protocol we have immunolabeled ultrathin epoxy sections of human somatostatinoma tumor using a primary antibody to somatostatin, followed by a biotinylated secondary antibody and visualization with streptavidin conjugated 585 nm cadmium-selenium (CdSe) quantum dots (QDs). Inherited epidermolysis bullosa (EB) includes a number of distinctive diseases that are characterized by the presence of fragile skin and the tendency to develop blisters and erosions. The current classification separates the types of EB on the basis of the ultrastructural level of the blisters. A 75 year old patient with respiratory infections since childhood, bronchiectasis and situs inversus is presented. Fiberoptic bronchoscopy was performed obtaining samples of bronchial mucosa which allowed the observation of the existence of ultrastructural lesions such as ciliary fusion, changes in the form of the cilia, microtubular changes and changes of the dynein arms by electronic microscopic study. Diagnosis of Kartagener's syndrome was established. In the case presented moderate clinical involvement was present which allowed the survival of the patient despite his advanced age.

The common procedures used for preparing some organs and tissues for electron microscopy, in which a fixative with the buffer portion adjusted to near-isotonicity to plasma is perfused in vivo, causes intolerable shrinkage of rat pineal cells. The present study was undertaken to optimize the parameters involved in the fixation of the pineal gland. The buffer and its concentration and the aldehyde or aldehydes used were among the variables investigated. The buffers tried were phosphate, cacodylate, PIPES, and HEPES. Decreasing the buffer concentration prevented shrinkage with all four buffers.

Five explanted posterior chamber lenses were examined under the scanning electron microscope to determine the changes. The intraocular time of the lenses was 1 week to 3 years. The polypropylene loops showed superficial cracks in all cases, but only in the curve and the insertion areas. The findings therefore indicated that the morphological changes are more likely caused by mechanical stress than by biological degradation alone.

The initial presentation of scleroderma (systemic sclerosis) with a manifestation involving only the breast has not been reported in the literature. Previous reports document scleroderma occurring after cosmetic mammaplasties. The presumed etiology is related to silicone, which has some adjuvant effect in development of the disease. This report documents a case of scleroderma presenting as a breast problem in a 28-year-old woman without prior exposure to silicone. We also describe the light and electron microscopy findings.
