Immunolocalization of Zic2 expression in the developing mouse forebrain.

The Zic genes are a family of zinc finger transcription factors defined by their homology with the Drosophila gene, odd-paired (opa). Zic2 has a critical role in forebrain development, as is evidenced by the fact that, in both mice and humans, diminished expression results in the severe forebrain malformation known as holoprosencephaly. Published information indicates that Zic2 expression is most prominent in the dorsal neural tube/spinal cord and in the hindbrain; however, there is no published description of the pattern of expression of Zic2 in the developing forebrain where the main Zic2 as

Magnetic resonance microscopy of iron in the basal forebrain cholinergic structures of the aged mouse lemur.

Increased non-heme iron levels in the brain of Alzheimer's disease (AD) patients are higher than the levels observed in age matched normal subjects. Iron level in structures that are highly relevant for AD, such as the basal forebrain, can be detected post mortem with histochemistry. Because of the small size of these structures, in vivo MR detection is very difficult at conventional field magnets (1.5 and 4 T). In this study, we observed iron deposits with histochemistry and MR microscopy at 11.7 T in the brain of the mouse lemur, a strepsirhine primate which is the only known animal model of

The distribution of p75 neurotrophin receptor-immunoreactive cells in the forebrain of the common marmoset (Callithrix jacchus).

The distribution of neurones that could be stained immunohistochemically with antibody to the p75 neurotrophin protein was studied in the forebrain of the common marmoset. The p75-immunoreactive forebrain cells appear to correspond to choline acetyltransferase-immunoreactive (i.e., cholinergic) neurones. Two populations of cells could be distinguished on the basis of the intensity of p75 immunostaining. Moderately stained cells correspond to cholinergic interneurones of the caudate and putamen, while intensely stained cells correspond to the cholinergic neurones projecting to the cortex, amygd

Neuropeptide Y in the forebrain and retina of the killifish, Fundulus heteroclitus.

The organization of the neuropeptide Y (NPY)-immunoreactive system in the forebrain, pineal organ and retina of a biweekly spawning fish (Fundulus heteroclitus) was investigated. Immunoreactivity was encountered in neurons of the nucleus olfactoretinalis, in the large population of neurons in the floor of the telencephalon, and in the nucleus entopeduncularis. Isolated somata were encountered in the hypophysiotropic hypothalamic nuclei, viz., the nucleus preopticus periventricularis, nucleus preopticus, and nucleus lateralis tuberis. Immunoreactive somata were also seen in the nucleus dorsomed

Organization of afferent and efferent projections of the nucleus basalis prosencephali in a passerine, Taeniopygia guttata.

The connections of nucleus basalis (NB) of the rostral forebrain of the zebra finch were investigated electrophysiologically and with anterograde and retrograde tracing methods to determine their functional organization, the sources of their pontine afferents, and the targets of their telencephalic efferents. The nucleus was found to be partitioned into three major components, a rostral lingual part that received a hypoglossal projection via a lateral subnucleus of the principal sensory trigeminal nucleus (PrV), a middle beak part that received a trigeminal projection via a medial subnucleus o

Muscle atonia is triggered by cholinergic stimulation of the basal forebrain: implication for the pathophysiology of canine narcolepsy.

Narcolepsy is a sleep disorder characterized by excessive daytime sleepiness and rapid eye movement (REM) sleep-related symptoms, such as cataplexy. The exact pathophysiology underlying the disease is unknown but may involve central cholinergic systems. It is known that the brainstem cholinergic system is activated during REM sleep. Furthermore, REM sleep and REM sleep atonia similar to cataplexy can be triggered in normal and narcoleptic dogs by stimulating cholinergic receptors within the pontine brainstem. The pontine cholinergic system is, therefore, likely to play a role in triggering cat

Experimentally induced disorders of neuronal migration produce an increased propensity for electrographic seizures in rats.

Disorders of neuronal migration in humans are associated with intractable epilepsy and some evidence suggests a causal relationship. This study evaluated electroencephalograms (EEG) of rats with experimentally induced disorders of neuronal migration. Fetal Sprague-Dawley rats were exposed to 196 cGy external irradiation on days 16 and 17 of gestation. This produced adult offspring with diffuse cortical dysplasias, agenesis of the corpus callosum, periventricular heterotopias, and dispersion of the pyramidal cell layer of the hippocampus. Epidural electrodes were implanted in four experimental

Luteinizing hormone-releasing hormone (LHRH) and neural cell adhesion molecule (NCAM)-immunoreactivity in development of the forebrain and reproductive system.

The origin and migration of LHRH neurons (detected by immunocytochemical procedures) is preceded by a migration of NCAM-immunoreactive cells from the olfactory epithelium, and the formation of an NCAM-immunoreactive cellular aggregate between the olfactory epithelium and the developing forebrain. The central processes of the olfactory nerves grow into the lateral parts of this aggregate and the terminal and vomeronasal nerves grow into the medial parts. No nerve fibers of the main or accessory olfactory systems grow directly into the forebrain. The LHRH neurons, following the course of the ter

Tremor-bursts induced by a combined application of chlorpromazine and pentobarbital in guinea pigs are asymmetrically influenced by right- and left-brain lesions.

The effect of right- and left-brain lesions on pentobarbitone-chlorpromazine induced tremor-bursts was studied in guinea pigs. The left-brain transections did not affect these tremor bursts except post-thalamic sections, which caused a complete disappearance of extensor jerks with coinciding tremors at four legs. These tremor bursts simultaneously occurring at four legs completely disappeared after a right-forebrain transection at the level of the rostral striatum. Extensor jerks without tremors continued to occur, however. It is concluded that the postural system is asymmetrically controlled

The influence of the olfactory placode on the development of the telencephalon in Xenopus laevis.

Removal of the sensory plate in Xenopus laevis embryos was performed to study the influence of the olfactory anlage on the development of the forebrain. Embryos, which at stage 22-23 underwent removal of the olfactory anlagen, were killed from stage 47 to 60. In 79% of the animals, two olfactory organs reformed and gave origin to two olfactory nerves which contacted the forebrain. In this instance, the telencephalic hemispheres developed normally. In 14% of the animals, one olfactory organ reformed which contacted the brain by means of one olfactory nerve. This resulted in the development of a

Amnesia following a discrete basal forebrain lesion.

Destructive lesions of the basal forebrain are associated with memory impairment in both humans and experimental animals. The basal forebrain is thought to contribute to memory function by providing cholinergic innervation to critical memory structures such as the hippocampus and amygdala. In previously reported clinical cases of basal forebrain amnesia, multiple neuroanatomical regions have been damaged, preventing identification of the minimal critical lesion necessary to produce an amnestic syndrome. We describe a patient who developed persistent, global anterograde and retrograde amnesia f

Forebrain Lineage Factor DMRTA2 Is Not Required for Maintenance of H3G34-Mutant Paediatric High-Grade Gliomas.

Developmental transcriptional programs are increasingly recognized as key drivers of paediatric gliomagenesis. Here, we unexpectedly find that the forebrain-specific doublesex- and mab-3-related transcription factor a2 (DMRTA2), despite being highly expressed in H3G34-mutant diffuse hemispheric glioma (DHG-H3G34) and implicated in tumour initiation, is not required for the maintenance of established patient-derived tumour cells. These findings support a stage-specific framework in which transcriptional dependencies are dynamically rewired during tumour progression in paediatric brain tumours.

Ank3 loss in adult forebrain excitatory neurons disrupts behavior, neuronal activity, membrane proteome, and myelination.

ANK3, encoding the scaffolding protein ankyrin-G, is a major risk gene for bipolar disorder and schizophrenia, but its cellular and circuit-level mechanisms remain poorly defined. Here, we demonstrate that deletion of Ank3 in forebrain excitatory neurons-either prenatally (Ank3-/-:Emx1-Cre) or in adolescence (Ank3-/-:CaMKIIα-Cre) leads to convergent behavioral phenotypes in adulthood, including hyperactivity, reduced anxiety-like behavior, and decreased depression-like responses. Calcium imaging in cultured neurons and acute brain slices revealed that ankyrin-G loss reduces both spontaneous an

Immunohistochemical relationships of huntingtin-associated protein 1 with choline acetyltransferase in the forebrain cholinergic nuclei of adult mice.

Huntingtin-associated protein 1 (HAP1) is a core component of the stigmoid body (STB) and a neuroprotective interactor with the causative agents of several neurodegenerative diseases (NDs). The cholinergic system is often affected by NDs. Our previous studies suggest that cholinergic brainstem/spinal cord motoneurons are more vulnerable to neurodegeneration due to a lack of STB/HAP1 protectivity. The forebrain cholinergic nuclei are also major neurodegenerative/psychotic targets; however, the relationships of HAP1 with choline acetyltransferase (ChAT) have yet to be determined there. This stud

Celsr3 Inactivation in the Brainstem Impairs Rubrospinal Tract Development and Mouse Behaviors in Motor Coordination and Mechanic-Induced Response.

Inactivation of Celsr3 in the forebrain results in defects of longitudinal axonal tracts such as the corticospinal tract. In this study, we inactivated Celsr3 in the brainstem using En1-Cre mice (Celsr3 cKO) and analyzed axonal and behavioral phenotypes. Celsr3 cKO animals showed an 83% reduction of rubrospinal axons and 30% decrease of corticospinal axons in spinal segments, associated with increased branching of dopaminergic fibers in the ventral horn. Decreases of spinal motoneurons, neuromuscular junctions, and electromyographic signal amplitude of the biceps were also found in mutant anim

Extensive divergence of projections to the forebrain from neurons in the paraventricular nucleus of the thalamus.

Neurons in the paraventricular nucleus of the thalamus (PVT) respond to emotionally salient events and project densely to subcortical regions known to mediate adaptive behavioral responses. The areas of the forebrain most densely innervated by the PVT include striatal-like subcortical regions that consist of the shell of the nucleus accumbens (NAcSh), the dorsolateral region of the bed nucleus of the stria terminalis (BSTDL) and the lateral-capsular division of the central nucleus of the amygdala (CeL). A recent tracing experiment demonstrated that the PVT is composed of two intermixed populat

Localizing and lateralizing values of postictal behavioral impairments in epileptic rats.

Transient postictal behavioral impairments in patients with epilepsy provide clues to seizure localization, but no attempt has been made previously to study the localization/lateralization value of postseizure motor disturbances in experimental models of epilepsy. The present study investigated relation of postictal motor deficit to seizure localization in the rat model of sound-induced reflex epilepsy. Sound-induced motor seizures started with a focal brainstem seizure (running) and progressed to a secondarily generalized seizure. Depending on innate or acquired seizure susceptibility of rats

Tyrosine hydroxylase in the olfactory system, forebrain and pituitary of the Indian major carp, Cirrhinus cirrhosus: organisation and interaction with neuropeptide Y in the preoptic area.

Dopamine (DA) inhibits, whereas gonadotrophin-releasing hormone (GnRH) stimulates, luteinisiing (LH) cells in the pituitary of some but not all teleosts. A reduction in the hypophysiotropic dopaminergic tone is necessary for the stimulatory effect of GnRH on LH cells. Neuropeptide Y (NPY) has emerged as one of the potent, endogenous agent that modulates LH secretion directly or indirectly via GnRH. Involvement of NPY in the regulation of hypophysiotropic DA neurones, however, is not known, but there is good evidence suggesting an interaction in the mammalian hypothalamus. DA neurones, identifi
