Inadequate antibody validation places substantial numbers of animal and human tissue samples at risk of waste.

Lack of antibody validation by researchers frequently misdirects biomedical research, yet the ethical consequences-particularly avoidable use of animal and human biological materials-remain unquantified. Using focus groups (n = 12), surveys (n = 107), and systematic analysis of 785 publications, we examined how researchers select and validate antibodies, and quantified the downstream ethical costs. Antibody selection is substantially influenced by social factors such as previous laboratory use and peer recommendations, while systematic evaluation of performance characteristics remains limited.

Parameter Optimization and Performance Validation of a Cone-Type Macadamia Nut Sheller Based on the Discrete Element Method.

This study investigates the shelling characteristics of macadamia nuts using the discrete element method (DEM), aiming to improve the shelling rate and whole kernel rate. First, the three principal dimensions (hilum, width, and horizontal directions) of macadamia nuts from Lincang, Yunnan, were measured using a caliper. A universal testing machine was used to measure the shelling force and contact characteristics of the nuts. An EDEM model with BondingV2 adhesive bonds was developed and validated through cylinder-lifting experiments, with a simulation error of less than 5%. Single-factor simul

Measurement reliability, construct validity, and transparent reporting in original and replication psychological research.

Published (replication) studies that use measures to assess psychological phenomena need to transparently report information on measurement procedures, validity, and reliability to allow verification and use in future (replication) research. However, earlier results highlighted widespread poor reporting of psychological measurement. Here, we investigated measurement reporting in a sample of 77 measures within 56 Many Labs replications and related original articles (14-17) and found that the information relevant for reusing measures was reported in full in around half the replication measures,

Diagnostic Performance and Reproducibility of the CARS Endoscopic Score in Achalasia: A Systematic Review and Meta-Analysis.

BACKGROUND AND AIM: Achalasia's insidious onset and nonspecific presentation contribute to diagnostic delays, often exceeding 20 months from symptom onset to confirmation. The four-domain CARS endoscopic score (Content, Anatomy, Resistance, Stasis) may expedite risk stratification. We performed a meta-analysis to assess diagnostic accuracy, interobserver reliability, and treatment-monitoring potential. METHODS: We searched PubMed, Embase, Web of Science, the Cochrane Library, and ClinicalTrials.gov through June 2025. Study-level 2 × 2 contingency data were then synthesized using bivariate ran

A mechanism-guided simulation framework for synthetic tax-risk data: Constraint validation and reproducible benchmarking.

Synthetic benchmarks can support tax-risk method development when administrative records are inaccessible, but a simulator can also predetermine the patterns later reported as detection findings. We present the Tax Data Generation Model (TDGM), a mechanism-guided simulator that enforces specified income-statement and balance-sheet identities, generates a three-year panel for 10,000 firms, and injects four prespecified risk strategies. This study uses no administrative audit data; its purpose is to test whether the implementation reproduces its design assumptions and to provide a reproducible b

The argument-based approach to validity: theory, assumptions, evidence, and consequences.

BACKGROUND: Concerns about measurement validation are often expressed as imperatives to use a "valid measure". However, validity is not a characteristic of a measure. Instead, validation and validity refer to score interpretation within a context of use. This is important because responses to health measures can be different in different contexts, influencing equitable consequences of measurement. In this paper, we aim to (1) outline the theory of the argument-based approach to validity, and (2) discuss assumptions and evidence in relation to equitable consequences of measurement. METHODS: Th

Measuring mindfulness: A systematic review and evaluation of self-report mindfulness scales.

The growing body of research on mindfulness has led to the development of a wide range of self-report instruments designed to assess the construct. However, conceptual and psychometric issues have arisen, partly as a result of inconsistent definitions and operationalizations, but also from challenges in scale development and validation. This systematic review synthesizes and evaluates the psychometric qualities of 21 self-report mindfulness scales intended for adult populations and tested in their English versions. The scales were evaluated in reference to nine psychometric quality domains, ea

Skinfolds Measurement Protocols and Standards: A Narrative Review.

Skinfold thickness assessment is one of the most widely used anthropometric methods for estimating subcutaneous adipose tissue and overall body adiposity in clinical, epidemiological, and sports settings. Despite its long-standing application, substantial heterogeneity persists across measurement protocols and standardization systems, potentially affecting reproducibility and comparability across studies. This narrative review aims to critically synthesize the historical, conceptual, methodological, and technical development of the main skinfold measurement protocols and standards, systematica

Toward self-validating ECG Systems: A personalized, uncertainty-aware approach for detecting and estimating lead misplacement.

BACKGROUND: Electrode positioning directly influences the interpretation and diagnostic quality of ECG recordings. While current solutions mainly focus on detecting lead swaps in standard full-lead configurations, the growing adoption of portable and reduced-lead devices underscores the need for effective methods to identify and quantify electrode misplacement in various settings. METHODS: We developed and evaluated an end-to-end, personalized, uncertainty-aware framework that took ECG waveforms as input and automatically detected and estimated potential electrode misplacement, using an annot

Minimum Significant Ratio for Defining Treatment-Boosted ADA Responses Using Signal-to-Noise as an Alternative to Titer.

Signal-to-noise ratio (S/N) has emerged as a practical alternative to endpoint titer for evaluating anti-drug antibody (ADA) response magnitude. When titration is not performed, a scientifically justified threshold is needed to interpret longitudinal increases in S/N, particularly for identifying treatment-boosted ADA responses in subjects with pre-existing ADA. In conventional titer-based assessments, treatment-boosted responses are typically defined by an increase of two dilution steps relative to the pre-dose titer. In the absence of titer testing, an alternative threshold is needed to dete

A Response to Mailend et al.: Refined Operational Definitions for the Apraxia of Speech Rating Scale.

PURPOSE: This letter addresses a recent paper by Mailend et al. (2025) that expressed concerns about the Apraxia of Speech Rating Scale (ASRS) and offered guidelines and definitions regarding its principles, scale labels, item definitions, and scoring procedures, including the addition of new items to the scale. The primary purpose of this letter is to clarify the uncertainty that other users of the scale may have about whether the current ASRS version (ASRS 3.5; Duffy et al., 2023) should be altered to include the numerous proposed revisions. It provides examples of proposed revisions that, i

Limited structural reliability in public educational prediction benchmarks: a four-dimension audit of seven datasets.

Across seven public educational prediction datasets, three passed all four pre-modeling reliability checks; the remaining four either failed group-aware generalization tests or lacked the provenance metadata needed to run them. One dataset was initially classified as failing but corrected after excluding group-identifier features from the holdout matrix, demonstrating that the audit can distinguish genuine cross-group confounding from feature-encoding artifacts. Each dataset was audited before model optimization using four checks: baseline gap, split instability, null separation, and metadata

Clinical translation of artificial intelligence in hypospadias: a systematic review and meta-analysis.

Artificial intelligence (AI) has emerged as a promising tool to improve the objectivity and reproducibility of hypospadias assessment. However, evidence regarding its clinical applications remains fragmented. This systematic review and meta-analysis evaluated the current role, diagnostic performance, and status of clinical translation of AI in hypospadias. A systematic search of PubMed/MEDLINE, Embase, and Scopus was performed according to PRISMA 2020 guidelines (PROSPERO: CRD420261296235). Studies evaluating AI applications in pediatric hypospadias were included. AI applications were categori

Status report on novel intraoral scanner-based registration method of axes of rotation of the mandible: Proof of potential applicability and technological glass ceiling of in-house development.

A fast, equipment- and radiation-free input methodology for digital articulation is desirable for basic dental and surgical procedures of relatively low complexity. Our team previously proposed an intraoral scanner (IOS)-based strategy for calculating the mandibular axis of rotation using dual bite registrations in open and closed positions. In this study, we aimed to develop a methodology for validating this registration system using a state-of-the-art jaw motion tracking device (Modjaw). The bite alignment accuracy of the scanner used (Medit i600) proved sufficient for the task; however, its

Assessment, External Verification and Modification of Models in Diagnosing Primary Aldosteronism Subtypes.

Diagnostic models are used to identify primary aldosteronism (PA) subtypes, including unilateral primary aldosteronism (UPA) and bilateral primary aldosteronism (BPA), only UPA could be treated through surgery. PA diagnostic models were screened from the NCBI, Embase, and Web of Science databases and assessed using the Prediction Model Risk of Bias Assessment Tool (PROBAST). External validation of these models was conducted using our Cohort. Models with high specificity were selected as the foundation for further modification. Based on the data from Cohort A and Cohort B, a new diagnostic mode
