We studied the steric course of the reaction catalyzed by the N-ethylmaleimide (NEM) reductase of Yarrowia (Candida) lipolytica (Y. lipolytica), using 4R-[4-2H1]NADPH and 4S-[4-2H1]NADPH as cofactors and N-ethylcitraconimide as substrate. Active substrates and inhibitors of NEM reductase and its subcellular distribution were also investigated to clarify the biochemical properties of this enzyme. Three women with a photoallergic dermatitis precipitated by benzydamine are reported. In one patient the cause of the photosensitization was a cream containing benzydamine (Tantum cream), whereas in the other two the reaction occurred after oral administration (Tantum drops). In all three cases the photosensitization was confirmed by a photopatch test that was positive for the active ingredient benzydamine.

Flurazepam, a clinically proven and widely accepted sedative hypnotic, has been shown by a number of investigators to produce convulsions at toxic doses. In the present study, CGS-8216, a benzodiazepine receptor antagonist, reduced the dose of flurazepam required to produce convulsions. This suggests that the convulsant action of FLZ is exerted at a site other than the benzodiazepine receptor.

XC386 prolonged the tail bleeding time in the conscious mice. This effect was dose-dependent and persisted for at least six hours after the oral administration. XC386 was effective in preventing ADP-induced acute pulmonary thromboembolic death in mice at dose of 100 mg/kg. Aspirin and indomethacin had no effect on this model. XC386 also reduced the mortality rate in collagen- induced thromboembolic death at the same dose as aspirin and indomethacin (200 mg/kg). Eight female patients with drug-induced agranulocytosis (five patients with definite seropositive rheumatoid arthritis (RA), three patients with upper respiratory infections) were studied for the presence of HLA-B27 and antinuclear antibodies (ANA). INTRODUCTION: Selpercatinib is a selective inhibitor of rearranged during transfection (RET) kinase and is indicated for patients with RET fusion-positive non-small cell lung cancer, advanced RET fusion-positive solid tumors, and thyroid cancer. This report describes a patient who developed renal tubular damage resulting in renal loss of electrolytes and polyuria four weeks after the start of selpercatinib. To our knowledge, tubular damage is a rare selpercatinib-induced toxicity. This report contributes to the growing literature about selpercatinib-related adverse events and their management.
