MST1 mediates neuronal loss and cognitive deficits: A novel therapeutic target for Alzheimer's disease.

Alzheimer's disease (AD) is the most prevalent form of dementia in the old adult and characterized by progressive cognitive decline and neuronal damage. The mammalian Ste20-like kinase1/2 (MST1/2) is a core component in Hippo signaling, which regulates neural stem cell proliferation, neuronal death and neuroinflammation. However, whether MST1/2 is involved in the occurrence and development of AD remains unknown. In this study we reported that the activity of MST1 was increased with Aβ accumulation in the hippocampus of 5xFAD mice. Overexpression of MST1 induced AD-like phenotype in normal mice

Repositioning of Somatic Golgi Apparatus Is Essential for the Dendritic Establishment of Adult-Born Hippocampal Neurons.

New dentate granule cells (DGCs) are continuously generated, and integrate into the preexisting hippocampal network in the adult brain. How an adult-born neuron with initially simple spindle-like morphology develops into a DGC, consisting of a single apical dendrite with further branches, remains largely unknown. Here, using retroviruses to birth date and manipulate newborn neurons, we examined initial dendritic formation and possible underlying mechanisms. We found that GFP-expressing newborn cells began to establish a DGC-like morphology at ∼7 d after birth, with a primary dendrite pointing

Targeting the Hippo pathway in cancer.

The Hippo pathway is a highly conserved signalling network that controls tissue growth and cell fate, responding to physical properties of the tissue microenvironment and cell biological features such as adhesion and polarity. Hippo signalling perturbation is associated with several human diseases, particularly various solid cancers. Hippo pathway-targeted therapies are beginning to emerge for the treatment of cancer, most of which are focused on disrupting the ability of the YAP and TAZ transcription co-activator proteins to promote transcription of genes with their cognate TEAD1-4 DNA bindin

Human cytomegalovirus inhibits the proliferation and invasion of extravillous cytotrophoblasts via Hippo-YAP pathway.

BACKGROUND: Human cytomegalovirus (HCMV) infection in utero is very common during pregnancy, which can lead to adverse outcomes in both pregnancy and progeny, but its pathogenesis has not been fully clarified. The decrease of extravillous cytotrophoblasts (EVT) invasion is an essential pathophysiological process of some pregnancy complications. Hippo-YAP signaling pathway plays an important role in regulating cell proliferation and apoptosis. However, whether YAP is involved in HCMV uterine infection remains to be studied. METHODS: The primary EVT was cultured and infected by the HCMV strain

Beyond the Cardiomyocyte: Consideration of HIPPO Pathway Cell-Type Specificity.

Increasing interest in the HIPPO signaling pathway has stemmed largely from its ability to modulate cardioprotection and heart regeneration following injury, responses originating from manipulation within the cardiomyocyte. This viewpoint discusses the need for definitive understanding of HIPPO pathway function in cardiac non-myocytes, and highlights the necessity of considering cell type specificity for effective therapeutic targeting of the HIPPO pathway in cardiovascular disease.

Exploration of MST1-Mediated Secondary Brain Injury Induced by Intracerebral Hemorrhage in Rats via Hippo Signaling Pathway.

Intracerebral hemorrhage (ICH) is a serious public health problem which causes high rates of disability and mortality in adults. Cell apoptosis is a sign of secondary brain injury (SBI) following ICH. Mammalian sterile 20-like kinase-1 (MST1), an apoptosis-promoting kinase, is a part of the Hippo signaling pathway and involved in cell death, oxidative stress, and inflammation. However, the role and underlying mechanism of MST1 in SBI induced by ICH have not yet been fully explained. The main purpose of present research was to explore the role of MST1 and its potential mechanism in SBI after IC

The hippo pathway effector YAP regulates motility, invasion, and castration-resistant growth of prostate cancer cells.

Yes-associated protein (YAP) is an effector of the Hippo tumor suppressor pathway. The functional significance of YAP in prostate cancer has remained elusive. In this study, we first show that enhanced expression of YAP is able to transform immortalized prostate epithelial cells and promote migration and invasion in both immortalized and cancerous prostate cells. We found that YAP mRNA was upregulated in androgen-insensitive prostate cancer cells (LNCaP-C81 and LNCaP-C4-2 cells) compared to the level in androgen-sensitive LNCaP cells. Importantly, ectopic expression of YAP activated androgen r

Structure-Based Virtual Screening and De Novo Design Guided by 3D-QSAR Triage for the Identification of Nanomolar STK3 Inhibitors.

Serine/threonine kinase 3 (STK3) is a core kinase in the Hippo signaling pathway that regulates cell proliferation, differentiation, and apoptosis. Dysregulation of this pathway is linked to various cancers and immune disorders, emphasizing STK3 as a promising therapeutic target for diseases that involve abnormal cell growth. Despite their therapeutic relevance, only a limited number of potent STK3 inhibitors have been reported, likely due to the difficulty in designing molecules that can bind tightly to the ATP-binding site of STK3. Using the 2-(2-amino-5-phenylpyrimidin-4-yl)-5-methoxyphenol

MST1 bridges LYN and SHP-1 to suppress FcεRI-mediated mast cell activation and allergic responses.

Mammalian sterile-20-like kinase 1 (MST1) is a core component of the Hippo signaling pathway. A previous study of 24 patients with MST1 deficiency revealed that more than half of the patients presented symptoms of airway hyperresponsiveness and atopic dermatitis. We also found significantly reduced MST1 expression in patients with allergies and in mouse models of allergic asthma, suggesting that aberrant MST1 expression may be broadly relevant to allergic diseases. However, the specific mechanism by which MST1 may be related to allergic disorders has remained unclear. In our study, Mst1-/- mic

Acquired resistance to vemurafenib restrains thyroid cancer stem cell self-renewal by suppressing STAT3 activation.

Vemurafenib (PLX4032) is a B-Raf kinase-specific inhibitor that has been approved for treating BRAF-mutated melanoma but is ineffective in treating thyroid cancer. Cancer stem cells (CSCs) play an important role in drug resistance (DR). Our present study aims to determine the status of CSCs in thyroid cancer cells that have acquired adaptive drug resistance to PLX4032. We first established a DR anaplastic thyroid cancer (ATC) cell line by culturing SW1736 cells in the media containing gradually increasing concentrations of PLX4032 (0.1-4 μM) for 6 months. We found that HER3, a member of the Er

MST4 Regulates Microglia Neuroinflammation via Targeting PKM2 Nuclear Translocation in Epilepsy: An In Vivo and In Vitro Study.

Microglia-mediated neuroinflammation is closely associated with the pathogenesis of epilepsy. Mammalian sterile-20-like kinase 4 (MST4) has been suggested a regulator of inflammation. However, the effect of MST4 on microglia neuroinflammation in epilepsy remains unclear. A pilocarpine-induced rat epilepsy model was constructed and a lipopolysaccharide (LPS)-stimulated microglia cell model was applied in the current research. Knockdown or overexpression of MST4 was established using lentivirus transfection. Electroencephalograph (EEG) was employed to measure brain activities of rats. The protei

Zinc deficiency in bovine mural granulosa cells results in hippo pathway-mediated apoptosis†.

Zinc is essential for the proper functioning of a variety of cells and tissues. Due to the limited understanding of the role of zinc in the bovine ovary, our objective was to investigate the role of zinc in bovine mural granulosa cell (mGC) viability and identify cell signaling pathways regulated by zinc in this cell type. Key indicators of cell behavior were measured after cultured bovine mGCs were incubated with increasing levels of a zinc chelator, TPEN. Cell viability decreased in a dose dependent manner, with a 91% loss of cell viability seen at 10 μM TPEN (p = 1.02 × 10-9), which corresp

Targeting the Hippo Pathway for Cardiac Regeneration.

Ischemic heart disease, which affects more than 200 million people worldwide, is caused by reduced blood flow to the heart and leads to widespread cardiomyocyte death. Due to the limited regenerative potential of cardiomyocytes, the lost tissue is replaced by a fibrotic scar, resulting in reduced cardiac function and progression to heart failure. Current therapeutic interventions aim to improve blood flow but cannot address the inability of cardiomyocytes to renew after injury. However, multiple studies have shown that modulating the Hippo signaling pathway to activate Yes-associated protein (

The oncogenic potential of YAP requires Y357 phosphorylation in cholangiocytes but not in hepatocytes.

PURPOSE: The transcriptional cofactor yes-associated protein (YAP) is activated in primary liver cancers (PLCs). YAP is canonically inhibited through serine 127 (S127) phosphorylation via the Hippo pathway but activated by phosphorylation at tyrosine 357 (Y357) by SRC family kinases. Due to liver plasticity, PLCs can originate from different cell types. However, it is unknown whether YAP activation favors tumorigenesis from specific cells of origin or promotes lineage commitment during tumorigenesis. This study investigates how YAP Y357 phosphorylation influences tumor phenotype and cell of or
