Mitochondria and the lectin pathway of complement.

Mitochondria, the powerhouses of our cells, are remnants of a eubacterial endosymbiont. Notwithstanding the evolutionary time that has passed since the initial endosymbiotic event, mitochondria have retained many hallmarks of their eubacterial origin. Recent studies have indicated that during perturbations of normal homeostasis, such as following acute trauma leading to massive necrosis and release of mitochondria, the immune system might mistake symbiont for enemy and initiate an inappropriate immune response. The innate immune system is the first line of defense against invading microbial pa

Immune correlates of protection in human invasive aspergillosis.

Protective immunity against Aspergillus depends on a highly coordinated interaction between the innate and adaptive arms of the immune system. Fungal recognition via pattern recognition receptors, such as pentraxin 3, dectin-1, and Toll-like receptors, leads to complement activation, phagocytosis, and killing of ingested fungi. Aspergillus-specific T-helper 1 and 17 cells produce cytokines such as interferon γ and interleukin 17, which facilitate macrophage activation and neutrophil recruitment, respectively. Genetic (or drug-induced) defects in components of these networks of antifungal immun

Membrane remodelling mediates lipopeptide-induced immunity in Arabidopsis.

Bacteria-derived lipopeptides are immunogenic triggers of host defences in metazoans and plants. Root-associated rhizobacteria produce cyclic lipopeptides that activate induced systemic resistance against microbial infection in various plant species. Whether and how these molecules are perceived at the plant cell surface remains elusive. Here we reveal that immune activation in Arabidopsis thaliana by the lipopeptide elicitor surfactin is mediated via a specific interaction with membrane sphingolipids. It relies on host membrane remodelling and subsequent activation of mechanosensitive ion cha

The potential of targeting TREM-1 in IBD.

Innate immune dysfunction is a hallmark of the pathogenesis of Inflammatory Bowel Disease, both in Crohn's disease and ulcerative colitis. Despite considerable efforts in research to better understand the pathophysiology of IBD and for the development of new therapeutic modalities for IBD patients, there is no therapy specifically targeting the dysregulations of the innate immune response available today in that field. TREM-1 is exclusively expressed by innate immune cells and is an immune amplifier. Its engagement following the primary activation of Pattern Recognition Receptors, including To

Pathogenicity of and plant immunity to soft rot pectobacteria.

Soft rot pectobacteria are broad host range enterobacterial pathogens that cause disease on a variety of plant species including the major crop potato. Pectobacteria are aggressive necrotrophs that harbor a large arsenal of plant cell wall-degrading enzymes as their primary virulence determinants. These enzymes together with additional virulence factors are employed to macerate the host tissue and promote host cell death to provide nutrients for the pathogens. In contrast to (hemi)biotrophs such as Pseudomonas, type III secretion systems (T3SS) and T3 effectors do not appear central to pathoge

Immuno-functionomics reveals geographical variation and a role for TLR8 in mRNA vaccine responses.

The innate immune system plays a pivotal role in pathogen defense via pattern recognition receptor sensing, initiating responses upon infection or vaccination. Understanding its functional capacity is crucial for deciphering correlates of vaccine efficacy and understanding responses to infection. Here, we developed a holistic approach to study immune function, generating >3,100 readouts across 16 cell types, 18 pattern recognition receptors, and 11 produced cytokines using spectral flow cytometry. To explore geographical variation, we compared Europeans and urban and rural Indonesians. We obse

Roles in Innate Immunity.

Microglia are best known as the resident phagocytes of the central nervous system (CNS). As a resident brain immune cell population, microglia play key roles during the initiation, propagation, and resolution of inflammation. The discovery of resident adaptive immune cells in the CNS has unveiled a relationship between microglia and adaptive immune cells for CNS immune-surveillance during health and disease. The interaction of microglia with elements of the peripheral immune system and other CNS resident cells mediates a fine balance between neuroprotection and tissue damage. In this chapter,

From inflammasomes to fevers, crystals and hypertension: how basic research explains inflammatory diseases.

Pattern-recognition receptors, such as Toll-like receptors and NOD-like receptors (NLRs), are able through the recognition of pathogen-associated molecular patterns and danger-associated molecular patterns to sense microbe-dependent and microbe-independent danger and thereby initiate innate immune responses. In some autoinflammatory conditions, abnormalities in NLR signaling pathways are involved in pathogenesis, as exemplified by NOD2 mutations associated with Crohn's disease. Some other NLRs are components of the inflammasome, a caspase-1- and prointerleukin-1beta-activating complex. Clinica

Short 5' UTRs serve as a marker for viral mRNA translation inhibition by the IFIT2-IFIT3 antiviral complex.

Recognition of "non-self" nucleic acids, including cytoplasmic dsDNA, dsRNA, or mRNAs lacking proper 5' cap structures, is critical for the innate immune response to viruses. Here, we demonstrate that short 5' untranslated regions (UTRs), a characteristic of many viral mRNAs, can also serve as a molecular pattern for innate immune recognition via the interferon-induced proteins IFIT2 and IFIT3. The IFIT2-IFIT3 heterodimer, formed through an intricate domain swap structure resolved by cryo-EM, mediates viral mRNA 5' end recognition, translation inhibition, and ultimately antiviral activity. Alpha-kinase 1 (ALPK1) agonist DF-006 demonstrates potent efficacy in mouse and primary human hepatocyte (PHH) models of hepatitis B.

BACKGROUND AND AIMS: In the treatment of chronic hepatitis B (CHB) infection, stimulation of innate immunity may lead to hepatitis B virus (HBV) cure. Alpha-kinase 1 (ALPK1) is a pattern recognition receptor (PRR) that activates the NF-κB pathway and stimulates innate immunity. Here we characterized the preclinical anti-HBV efficacy of DF-006, an orally active agonist of ALPK1 currently in clinical development for CHB. APPROACH AND RESULTS: In adeno-associated virus (AAV)-HBV mouse models and primary human hepatocytes (PHHs) infected with HBV, we evaluated the antiviral efficacy of DF-006. In

YBX1 regulates RNA polymerase III transcripts to prevent inflammation.

Tolerance of endogenous nucleic acids is crucial for cellular homeostasis and when perturbed can lead to immune dysregulation. RNA-binding proteins (RBPs) bind, modify, and compartmentalize endogenous RNAs to prevent their recognition by the innate immune system. We found that loss of a highly abundant RBP, the cold shock domain containing Y-box binding protein 1 (YBX1), results in the spontaneous induction of an IL6/STAT3 inflammatory response and the upregulation of interferon-stimulated genes. A meta-analysis of other studies reveals that the depletion of YBX1 orthologs and paralogs results

Scavenger receptor A impairs interferon response to HBV infection by limiting TRAF3 ubiquitination through recruiting OTUB1.

The battle between hepatitis B virus (HBV) infection and the host immune defense determines the outcome of the disease. Scavenger receptor A (SRA) is a phagocytic pattern recognition receptor involved in various cellular processes, including lipid metabolism, recognition, and clearance of pathogens or modified self-molecules. Emerging evidence pointed out that SRA might act as an immunomodulator that contributes to innate immune defense against invading pathogens. Herein, we examined the role of SRA in the initiation of type I interferon (IFN) response to HBV infection and the virus clearance.

The NLRP3 inflammasome at the crossroads of innate immune signaling and cell death.

The NLRP3 inflammasome is a central signaling pathway of innate immunity that orchestrates host defense and inflammatory responses through the activation of proinflammatory cytokines and the induction of pyroptotic cell death. Accumulating evidence indicates that NLRP3 functions within an extensive network of innate immune and cell death pathways. Reciprocal interactions between NLRP3 and other pattern-recognition receptor pathways, including Toll-like receptors, DNA-sensing cGAS-STING signaling, and RNA-sensing RIG-I/MDA5-MAVS signaling, shape the magnitude and duration of inflammatory respon

Hypoxia inducible factors regulate pneumovirus replication by enhancing innate immune sensing.

The immune mechanisms responsible for protection and pathogenesis in pneumoviral infection are not well defined. We demonstrated that pharmacological activation of the hypoxic inducible factor (HIF) signaling axis using daprodustat limited viral replication through enhanced immune signaling. Transcriptomic analysis revealed HIF augmented activation of innate immune response genes, including interferon-stimulated gene 15 (Isg15), in the lung and spleen of mice infected with pneumonia virus of mice (PVM). In human respiratory syncytial virus (hRSV)-infected airway epithelial cells, daprodustat i

A bacterial fatty acid enzyme hijacks nucleic acid-sensing Toll-like receptor signaling.

Innate immune recognition shapes infection outcomes by linking microbial detection to host defense. Although pattern recognition receptors are classified by the ligands they detect (lipids, peptidoglycans, or nucleic acids) cross-talk between these pathways is increasingly recognized. Staphylococcus aureus, a major cause of skin and soft tissue infections, can persist intracellularly, evading immunity and antibiotics. Here, we describe a lipid-based immune evasion strategy in which the S. aureus enzyme oleate hydratase (OhyA) converts host fatty acids into hydroxylated lipids that antagonize T

Engineering polymeric RNA scaffolds as programmable combinatorial innate immune agonists.

Sentinel cells in the innate immune system decode combinations of pathogen-associated molecular patterns to shape immune responses, presenting an exploitable feature for vaccine adjuvants and cancer immunotherapies. Delivering innate immune agonists can direct responses toward patient-specific tumour antigens and enhance checkpoint blockade therapies. However, few adjuvants have been clinically approved, largely due to limited potencies or high toxicities. Key barriers remain in adjuvant design and translation: (i) reliance on single agonists, which poorly recapitulate complex pathogen sensing

Endogenous bacterial flora in pregnant women and the influence of maternal genetic variation.

Preterm labour, premature prelabour rupture of membranes and low birth weight have all been associated with either specific maternal genital tract infections or an altered vaginal microflora during pregnancy. Factors that influence the variation in microbial-host interaction play an important role in individual susceptibility to adverse pregnancy outcomes. The innate immune responses at mucosal surfaces play a crucial role against microbial invasion. Multiple genes are responsible for the regulation of the innate immune system. Genetic polymorphisms that disrupt innate immune recognition or th

Epithelial-Dermal Immune Memory: Tracking Staphylococcus aureus-Induced Trained Immunity in the Progression of Chronic Skin Inflammation.

The clinical paradigm distinguishing chronic atopic dermatitis (AD) from acute bacterial infections is established by the emerging evidence which focuses on persistent innate immune memory as a central pathobiological factor mediating inflammatory skin diseases. While AD is classically defined by genetic susceptibility and epidermal barrier failure, progression of deep dermal Staphylococcus aureus invasion, which is characteristic of bacterial infections like cellulitis, implies a significant acute infectious condition resulting in significant tissue damage, which drives extensive innate immun

Protective role of antibodies in enteric virus infections: Lessons from primary and secondary immune deficiencies.

Enteric viruses are the main cause of acute gastroenteritis worldwide with a significant morbidity and mortality, especially among children and aged adults. Some enteric viruses also cause disseminated infections and severe neurological manifestations such as poliomyelitis. Protective immunity against these viruses is not well understood in humans, with most knowledge coming from animal models, although the development of poliovirus and rotavirus vaccines has extended our knowledge. In a classical view, innate immunity involves the recognition of foreign DNA or RNA by pathogen recognition rece

Multiple regulatory roles of UNC93B1 in innate immunity.

Uncoordinated 93 homolog B1 (UNC93B1) is a key innate immune regulator that affects the function of pattern recognition receptors. UNC93B1 is responsible for governing the intracellular trafficking and activation of nucleic acid-sensing TLRs. Recently, UNC93B1 was shown to regulate the cGAS/STING signaling pathway. As a result, UNC93B1 is a promising therapeutic target for various human diseases. In this review, we provide an update on new findings establishing the regulatory role of UNC93B1 in innate immunity, including the antibacterial activity of neutrophils, the PRR-mediated signaling pat
