Structure-activity relationship of N-cyclopropylmethyl-7α-(m-methylaminophenyl)-6,14-endoethano-northebaine derivatives as G-protein-biased KOR-selective agonists.

4,5-Epoxymorphinan derivatives represent a privileged chemotype among opioid ligands, with well-established clinical utility, and continue to serve as a widely used scaffold for the development of novel opioid-like therapeutics. This work describes the design and synthesis of a series of N-cyclopropylmethyl-7α-(m-methylaminophenyl)-6,14-endoethano-northebaine derivatives and identifies compound 8a as a structurally optimized KOR agonist with high KOR affinity, pronounced MOR/KOR and DOR/KOR subtype selectivity, and marked G-protein bias. In vivo, compound 8a exhibited robust, dose-dependent an

Sinomenine alleviates cancer-induced bone pain by promoting p65 ubiquitination and modulating microglial polarization.

Cancer-induced bone pain (CIBP), a debilitating complication of metastatic bone cancer, necessitates prompt and effective therapeutic interventions. Sinomenine (SIN), an alkaloid extracted from Sinomenium acutum, demonstrates anti-inflammatory and analgesic properties; nevertheless, its efficacy and mechanisms in CIBP management is inadequately investigated. This study looked at how SIN affected CIBP in a female rat model with bone discomfort caused by tumor implantation and BV2 microglial cells that were activated by lipopolysaccharide (LPS). Behavioral assessments (von Frey filaments and spo

Sinomenine enhances recovery from corneal alkali burns by attenuating macrophage M1 polarization through modulating the METTL3/IRAKM axis.

Corneal alkali burns trigger the infiltration of inflammation-associated macrophages, which can potentially lead to vision impairment. Although Sinomenine (SIN) is recognized for its anti-inflammatory properties, its role on the polarization of macrophages and its effects on corneal alkali injury remain inadequately understood. In this study, corneal alkali damage was induced using NaOH. HE staining and immunohistochemistry were conducted to assess macrophage infiltration in corneal tissues. Macrophages were isolated via flow cytometry assay. Quantitative real-time PCR (qRT-PCR) was employed t

Sinomenine promotes flap survival by upregulating eNOS and eNOS-mediated autophagy via PI3K/AKT pathway.

Large skin defects and surgical tissue reconstructions are frequently covered utilizing random flaps. The flap has the advantage of being designed according to the size and shape of a surgical wound. However, the necrosis of the distal part of the flap restricts the clinical application of flaps. Sinomenine (SIN) is the major active component of sinomenium acutum. SIN has been demonstrated to inhibit oxidative stress and stimulate autophagy in a cell, animal, and clinical studies. The protective and proliferative effects of sinomenium on HUVECs were evaluated by scratched test, CCK-8, and EDU

Essential structure of orexin 1 receptor antagonist YNT-707: Conversion of the 16-cyclopropylmethyl group to the 16-sulfonamide group in d-nor-nalfurafine derivatives.

The five-membered D-ring nalfurafine (d-nor-nalfurafine) derivatives with a 16-sulfonamide group were synthesized. Conversion of the 16-cyclopropylmethyl group to the 16-benzenesulfonamide group in the d-nor-nalfurafine derivatives drastically improved the orexin 1 receptor (OX1R) antagonist activities. The intramolecular hydrogen bond between the 14-hydroxy and the 16-sulfonamide groups may play an important role in increasing the probability that the 6-amide group would be located at the lower side of the C-ring, leading to an active conformation for OX1R. The assay results and the conformat

Nalfurafine, a G-Protein-Biased KOR (Kappa Opioid Receptor) Agonist, Enhances the Diuretic Response and Limits Electrolyte Losses to Standard-of-Care Diuretics.

Nalfurafine is a G-protein-biased KOR (kappa opioid receptor) agonist that produces analgesia and lacks central nervous system adverse effects. Here, we examined the cardiovascular and renal responses to intravenous and oral nalfurafine alone and in combination with furosemide, hydrochlorothiazide, or amiloride. We hypothesized that nalfurafine, given its distinct mechanism of vasopressin inhibition, would increase urine output to these diuretics and limit electrolyte loss. Following catheterization, conscious Sprague-Dawley rats received an isotonic saline infusion and were then administered

Cardiopulmonary effects of butorphanol tartrate intravenously administered for placement of duodenal cannulae in isoflurane-anesthetized yearling steers.

Cardiopulmonary effects of IV administered butorphanol tartrate (BUT) were assessed in 7 yearling steers medicated with atropine and anesthetized with guaifenesin, thiamylal sodium, and isoflurane in O2 for surgical placement of duodenal cannulae. Heart rate, respiratory rate, arterial blood pressures, pHa, PaCO2, PaO2, arterial [HCO3-], esophageal temperature, and end-tidal isoflurane concentrations were measured before and after IV administration of BUT (10 mg). Mean respiratory rate increased significantly (P less than 0.05) only at 45 and 60 minutes after BUT administration. Mean respirato

Physiological response of gray wolves to butorphanol-xylazine immobilization and antagonism by naloxone and yohimbine.

Captive gray wolves (Canis lupus) were immobilized (loss of consciousness) with 2.0 mg/kg xylazine hydrochloride (XYL) and 0.4 mg/kg butorphanol tartrate (BUT) administered intramuscularly. Induction time was 11.8 +/- 0.8 min (mean +/- SE). Immobilization resulted in bradycardia, respiratory depression, and normotension. Fifteen min after induction, six wolves were given either 0.05 mg/kg naloxone hydrochloride (NAL) and 0.125 or 0.250 mg/kg yohimbine hydrochloride (YOH), or an equal volume of saline (control) intravenously. Antagonism resulted in shortened recovery times compared to control a

Cardiovascular effects of etorphine in rats.

1. Cardiovascular effects of intravenously administered etorphine were investigated in mechanically ventilated normotensive rats under pentobarbitone anaesthesia. 2. Etorphine (0.1-2 micrograms kg-1) induced a dose-related bradycardia and hypotension which was prevented by pretreatment with naloxone (0.1 mg kg-1). 3. After bilateral vagotomy etorphine (1 microgram kg-1) produced a pressor effect which was prevented by prazosin (0.5 mg kg-1), but unaltered by adrenalectomy. 4. The bradycardia due to etorphine was abolished by bilateral vagotomy, but only partially reduced by atropine (1 mg kg-1

Nalbuphine: a supplement to isoflurane and enflurane anaesthesia.

A retrospective study was carried out to review the intra-operative use of nalbuphine at the average dose of 1.5 mg/kg as a supplement to isoflurane and enflurane in balanced anaesthesia in 108 surgical patients. Intra-operative cardiovascular stability and the quality of emergence were examined. The amount of halogenated anaesthetic used was compared to the theoretical amount that would have been needed in the absence of nalbuphine. In 90% to 95% of patients, blood pressures remained within 20% of baseline for the duration of anaesthesia. At emergence, 80% of patients had no pain. Nalbuphine

Clinical actions of fentanyl and buprenorphine. The significance of receptor binding.

Receptor binding assays were undertaken in an attempt to elucidate the opioid binding characteristics of fentanyl and buprenorphine, and to investigate some of the differences between them. Buprenorphine showed slow receptor association (30 min), but with high affinity to multiple sites from which dissociation was very slow (T 1/2 = 166 min) and incomplete (50% binding after 1 h). This contrasted with the receptor binding of fentanyl, which achieved rapid equilibrium (within 10 min) and dissociated equally rapidly (T 1/2 = 6.8 min) and completely (100% by 1 h). Competitive displacement showed

Prevention of late fentanyl-induced respiratory depression after the injection of opiate antagonists naltrexone and S-20682: comparison with naloxone.

Compared with naloxone, two opiate antagonists (naltrexone and S-20682) were tested in the intact dog in order to reverse respiratory depression, induced by fentanyl 50 micrograms kg-1 i.v. Respiratory rate and arterial blood-gases were measured at 5, 30, 60, 120, 180 and 240 min after the additional i.v. injection of the antagonist (1 microgram kg-1). Only S-20682, but not naloxone or naltrexone, prevented late (240 min) fentanyl-induced depression of respiratory drive. A late respiratory depression coincided with an increase in amplitudes and a reduction of frequency in electrical cortical a

Buprenorphine: differential interaction with opiate receptor subtypes in vivo.

The mixed agonist-antagonist buprenorphine previously was shown to display a bell-shaped dose-response curve with peak agonistic (antinociceptive) opiate effects at approximately 0.5 mg/kg s.c., 60 min after the dose and a gradual decline of the effects in the dosage range between 0.5 to 10 mg/kg (electrically induced vocalization test). In vitro, buprenorphine as well as the potent pure agonist etorphine possessed very high affinity for all of the opiate receptor subtypes that are labeled by the universal tracer [3H]diprenorphine. However, in vivo, [3H]buprenorphine and [3H]etorphine appeared

Immobilization of game animals in trypanosomiasis research.

The proximity of human populations to communities of large animals in Africa creates suitable conditions for the transmission of zoonotic diseases. Since the indiscriminate destruction of these communities to control zoonoses is highly undesirable, the epidemiological role of the animals must be properly assessed in order that alternative methods of control can be developed. For this purpose, techniques should be available to permit large animals to be examined alive. Immobilization techniques using various drugs were tried but only limited success was achieved with the species most likely to

Sinomenine ameliorates experimental autoimmune neuritis by suppressing pro-inflammatory immune components and restoring immune balance.

Sinomenine (SIN), a bioactive alkaloid derived from Sinomenium acutum, exhibits well-documented anti-inflammatory and analgesic properties with a favorable safety profile. Here, we pharmacologically characterized the preventive and therapeutic efficacy of SIN in a rat model of experimental autoimmune neuritis (EAN), an established model of immune-mediated peripheral neuropathy. Although SIN has been evaluated in several neuroinflammatory disorders, its mode of action in EAN has not been systematically addressed. Disease progression, body-weight changes, and neurological scores were monitored f

Sinomenine ameliorates adjuvant-induced arthritis by inhibiting the autophagy/NETosis/inflammation axis.

Studies have found that neutrophil extracellular traps (NETs) which are the specific dying form of neutrophil upon activation have fundamental role in the rheumatoid arthritis onset and progression. The purpose of this study was to explore the therapeutic effect of Sinomenine on adjuvant-induced arthritis in mice, and the neutrophil activities regulated by Sinomenine. The rheumatoid arthritis model was established by local injection of adjuvant and the Sinomenine treatment was administered orally for 30 days, during which, arthritic scores were evaluated and the joint diameter was measured to
