Aflatoxin, liver enzymes, and hepatitis B virus infection in Gambian children.

The relative contribution of, and possible mechanism of interaction between, aflatoxin and hepatitis B virus (HBV) in the development of primary hepatocellular carcinoma can be better investigated now that markers of individual exposure to both factors are available. In this study, blood samples were collected over a 1-month period from 117 children aged 3 to 4 years, resident in Kuntair or Kerr Cherno in the Upper Niumi District of The Gambia. Samples were analyzed for aflatoxin-albumin (AF-alb) adducts, markers of HBV infection, liver enzymes [serum alanine aminotransferase (ALT)] as markers

Effects of aflatoxin ingestion on the development of Moraxella bovis infectious bovine keratoconjunctivitis.

A study was conducted to determine whether measured doses of aflatoxin given under different schedules would influence the pathogenesis of Moraxella bovis induced infectious bovine keratoconjunctivitis (IBK). Calves were allotted to 4 groups (groups I-IV) of 9, 9, 9, and 8 calves, respectively. Group I calves were given aflatoxin for 11 consecutive days starting 5 days before their eyes were exposed to M. bovis. Group II calves were given aflatoxin for 5 consecutive days starting 7 days after their eyes were exposed to M. bovis. Group III calves were given aflatoxin for 5 consecutive days star

Regulation of aflatoxin biosynthesis: effect of glucose on activities of various glycolytic enzymes.

Catabolism of carbohydrates has been implicated in the regulation of aflatoxin synthesis. To characterize this effect further, the activities of various enzymes associated with glucose catabolism were determined in Aspergillus parasiticus organisms that were initially cultured in peptone-mineral salts medium and then transferred to glucose-mineral salts and peptone-mineral salts media. After an initial increase in activity, the levels of glucose 6-phosphate dehydrogenase, mannitol dehydrogenase, and malate dehydrogenase were lowered in the presence of glucose. Phosphofructokinase activity was

Effects of dietary aflatoxin on existing bacterial intramammary infections of dairy cows.

Aflatoxin, a naturally occurring mycotoxin with immunosuppressive properties, was given to 4 lactating dairy cows that had experimentally induced or naturally occurring mammary infections in 2 of their 4 mammary quarters. Aflatoxin was given orally in daily doses approximating 0.3 mg of B1 activity/kg of body weight for periods of 12 to 14 days. Infecting organisms were Streptococcus agalactiae, Staphylococcus aureus, and Staphylococcus hyicus. The 4 cows were monitored for clinical signs of disease and signs of mastitis for periods before, during, and after aflatoxin consumption. The cows dev

Research progress on core preparation variables, detection methods, and sensing performances of molecularly imprinted polymers (MIPs) for aflatoxins determination.

Aflatoxins (AFs) are difuranocoumarin toxins with main types including AFB1, AFB2, AFG1, AFG2, AFM1, and AFM2. These toxins are frequently detected in grain and oil products, agricultural raw materials, and Chinese medicinal herbs, posing a severe threat to human health and food safety. Molecularly imprinted technology (MIT) exhibits distinct advantages such as high specificity, broad applicability, and excellent stability. Its core materials, molecularly imprinted polymers (MIPs), enable the precise recognition and enrichment of trace AFs in complex matrices, thereby addressing the limitation…

The Efficacy of Additives for the Mitigation of Aflatoxins in Animal Feed: A Systematic Review and Network Meta-Analysis.

The contamination of animal feed with aflatoxins is an ongoing and growing serious issue, particularly for livestock farmers in tropical and subtropical regions. Exposure of animals to an aflatoxin-contaminated diet impairs feed efficiency and increases susceptibility to diseases, resulting in mortality, feed waste, and increased production costs. They can also be excreted in milk and thus pose a significant human health risk. This systematic review and network meta-analysis aim to compare and identify the most effective intervention to alleviate the negative impact of aflatoxins on the import…

Updates on the Functions and Molecular Mechanisms of the Genes Involved in Aspergillus flavus Development and Biosynthesis of Aflatoxins.

Aspergillus flavus (A. flavus) is a ubiquitous and opportunistic fungal pathogen that causes invasive and non-invasive aspergillosis in humans and animals. This fungus is also capable of infecting a large number of agriculture crops (e.g., peanuts, maze, cotton seeds, rice, etc.), causing economic losses and posing serious food-safety concerns when these crops are contaminated with aflatoxins, the most potent naturally occurring carcinogens. In particular, A. flavus and aflatoxins are intensely studied, and they continue to receive considerable attention due to their detrimental effects on hum…

Combining ultrasound-assisted extraction and vortex-assisted liquid-liquid microextraction for the sensitive assessment of aflatoxins in aquaculture fish species.

Although aflatoxins contamination in feedstuff is a well-known problem, and hence these residues are controlled in poultry products, there is scarce information regarding the presence of these toxic substances in aquaculture fish, facilities that use several feedstuff for fish breeding. A simple, rapid, and sensitive method has been therefore developed for aflatoxins (B1, B2, G1, and G2) assessment in aquaculture products by combining ultrasound probe-assisted extraction and vortex-assisted liquid-liquid microextraction as a sample pretreatment, and high-performance liquid chromatography-tande…

Risk of dietary exposure to aflatoxins and fumonisins in infants less than 6 months of age in Rombo, Northern Tanzania.

Infants less than 6 months of age receiving foods other than breast milk are at a high risk of exposure to mycotoxins. We surveyed food intake and estimated the risk of exposures to aflatoxin and fumonisin mycotoxins for infants less than 6 months of age in Northern Tanzania. A total of 143 infants were progressively recruited and three follow-up visits were made at 1, 3 and 5 months of age. A 24-h dietary recall technique was used to estimate flour intake of infants who had been introduced to maize foods. Aflatoxins and fumonisins in the flours were analysed using high-performance liquid chro…

Hepatocellular carcinoma and liver cirrhosis TP53 mutation analysis reflects a moderate dietary exposure to aflatoxins in Espírito Santo State, Brazil.

The close relationship between aflatoxins and 249ser TP53 gene mutation (AGG to AGT, Arg to Ser) in hepatocellular carcinoma (HCC) makes this mutation an indirect indicator of dietary contamination with this toxin. We have examined the prevalence of codon 249 TP53 mutation in 41 HCC and 74 liver cirrhosis (without HCC) cases diagnosed at the HUCAM University Hospital in Vitoria, Espírito Santo State, Brazil. DNA was extracted from paraffin sections and from plasma. The mutation was detected by DNA amplification, followed by restriction endonuclease digestion and confirmed by direct sequencing.…

Interactions between hepatitis B virus infection and exposure to aflatoxins in the development of hepatocellular carcinoma: a molecular epidemiological approach.

Aflatoxins and hepatitis B virus (HBV) are major risk factors for hepatocellular carcinoma (HCC) in high incidence areas for this cancer, namely southeast Asia and parts of Africa. There is evidence from both epidemiological studies and animal models that the two factors can act synergistically to increase the risk of HCC. The cellular and molecular mechanism of the interaction between these two factors is as yet undefined. However, one possible mechanism attested to by studies in HBV transgenic mice is that chronic liver injury alters the expression of specific carcinogen metabolising enzymes…

Protective Effect of Ethoxyquin and N-acetylcysteine on Biochemical and Pathological Changes Induced by Chronic Exposure to Aflatoxins in Laying Hens.

Aflatoxins (AFs) represent a major threat to poultry health and food safety due to their hepatotoxic, immunosuppressive, and carcinogenic effects. This study evaluated the chemoprotective potential of ethoxyquin (EQ) and N-acetylcysteine (NAC) in laying hens (80.8 and 33.3 mg/kg BW/d) exposed to chronic dietary AFs contamination (0.0-1.5 mg/kg). A total of 360 Hy-Line W36 Leghorn hens were monitored over 72 weeks using biochemical and histopathological analyses of liver and kidney tissues. NAC significantly (p < 0.01) increased hepatic and renal levels of reduced glutathione (GSH) and stimulat…

Structure Elucidation and Toxicity Analysis of the Byproducts Formed after Biodegradation of Aflatoxins B1 and B2 Using Extracts of Mentha arvensis.

The aqueous extracts of leaves and shoots of Mentha arvensis were checked for their potential to biodegrade aflatoxin B1 and B2 (AFB1; 100 µg/L and AFB2; 50 µg/L) through in vitro assays. Overall, the results showed that leaf extract degrades aflatoxins more efficiently than the shoot extract. First, the pH, temperature and incubation time were optimized for maximum degradation by observing this activity at different temperatures between 25 and 60 °C, pH between 2 and 10 and incubation time from 3 to 72 h. In general, an increase in all these parameters significantly increased the percentage o…

Aflatoxins and kwashiorkor. Epidemiology and clinical studies in Sudanese children and findings in autopsy liver samples from Nigeria and South Africa.

The aetiology and pathogenesis of kwashiorkor remains obscure. Clinical and epidemiological studies are in progress in Sudan to determine whether aflatoxins play a role in the pathogenesis of kwashiorkor. Local foods are screened for their aflatoxin content and children with kwashiorkor and age and sex matched children with marasmus and with normal nutrition are being investigated for aflatoxins in their blood and urine. Autopsy liver samples of children with kwashiorkor and other nutritional disorders in Nigeria and South Africa have been examined for their aflatoxin content. Aflatoxins are d…

TRIP13 Promotes Aflatoxin B1-Induced Hepatocellular Carcinoma Progression by Inhibiting Ferroptosis.

Aflatoxin B1 (AFB1) is a potent foodborne hepatocarcinogen that contributes substantially to the global burden of hepatocellular carcinoma (HCC), yet the molecular mechanisms underlying AFB1-driven HCC progression remain incompletely understood. In this study, we identified thyroid hormone receptor-interacting protein 13 (TRIP13) as an important contributor to AFB1-related HCC progression and investigated its involvement in ferroptosis regulation. TCGA-LIHC, GSE25844, and single-cell RNA sequencing data (GSE166635) revealed that TRIP13 was significantly upregulated in AFB1-related HCC, enriche
