Clinical and molecular mechanistic insights into the WDR72 mutation.

Distal renal tubular acidosis (dRTA) is a rare kidney disorder with an incidence of less than 1 in 100 000 people, characterised by metabolic acidosis and hypokalaemia. While mutations in genes such as ATP6V1B1, ATP6V0A4 and SLC4A1 are commonly associated with dRTA, this study reports a rare variant of the WDR72 gene in siblings from the Punjabi population in India. The individuals exhibited amelogenesis imperfecta (AI), hypokalaemic periodic paralysis (HPP) and acidaemia due to impaired ion transport in the distal convoluted tubule. Genetic testing revealed novel variants in the WDR72 gene: c…

Dental abnormalities associated with campomelic syndrome: case report.

Campomelic syndrome is a rare autosomal recessive disease. It is characterized by short stature with angulation and bowing of the lower limbs, hypoplasia of the facial bones, and various other skeletal anomalies. The facies are unusually flat with micrognathia, frequent cleft palate, hypertelorism, and micro-ophthalmia. Most infants with this syndrome die from severe respiratory distress within the first hours or days of life. Those who survive infancy are mentally retarded. This case report concerns the dental treatment of a 13-year-old female with campomelic syndrome.

Minimally invasive treatments for developmental defects of enamel.

Developmental defects of enamel (DDEs) are clinical conditions that affect the mineral structure and integrity of dental enamel, leading to adverse effects such as dentinal hypersensitivity and significant esthetic impairment. Amelogenesis imperfecta, fluorosis, molar-incisor hypomineralization, and hypoplasia are all considered DDEs. In addition to having various etiologies that may influence the choice of treatment, DDEs also require personalized treatment plans based on the depth and color of the lesion. Studies on DDEs have explored minimally invasive dentistry techniques that preserve as …

Clinical Insights Into Linear Enamel Hypoplasia: A Case Report.

Linear enamel hypoplasia (LEH) is a failure or aberration of enamel to form correctly, resulting in bands of decreased enamel on the tooth surface. Many reasons, including genetics, malnutrition, and childhood diseases, can cause LEH, which solely affects the neonatal line. LEH is a kind of environmental hypoplasia with many symmetrical and ring-like abnormalities. Amelogenesis is a one-time procedure, and any abnormality might result in permanent enamel abnormalities. Enamel, unlike other human tissues, cannot be restored. The consequences of enamel hypoplasia vary based on illness severity, …

A Recurrent FAM83H Mutation in an Extended Colombian Family and Variable Craniofacial Phenotypes.

Amelogenesis imperfecta (AI) is a collection of rare genetic disorders affecting the quantity and/or quality of the tooth enamel. AI can be classified into three major types according to the clinical phenotype: hypoplastic, hypocalcified, and hypomatured. Among them, the hypocalcified type shows the weakest physical properties, leaving rough and discolored enamel surfaces after tooth eruption. To date, mutations in the FAM83H gene are responsible for the autosomal-dominant hypocalcified AI. In this study, we recruited a four-generation Colombian family with hypocalcified AI and identified a re…

Comparison of rates of enamel synthesis in impeded and unimpeded rat incisors.

Periodic intubations of rats with solutions of fluoride (F) lead to the appearance of bands of disrupted pigmentation in continuously erupting incisors. Distances between fluorotic bands reflect time intervals between intubations. In this experiment, the periodicity of fluorotic banding was used for estimation of the rate of enamel synthesis in impeded and unimpeded rat incisors. Rats kept on a low-F diet and distilled water were intubated two or four times per week with 2 mg NaF/150 g body weight. In a group of rats, one of the mandibular incisors was cut at the gingival margin after two week

Changes of secretory ameloblasts in mini-pig fetuses exposed to ethanol in vivo.

After addition of ethanol to the ordinary fodder of pregnant mini-pigs, ultrastructural changes were found in secretory ameloblasts from tooth germs of their mid-term fetuses. Compared with controls, many mitochondria showed abnormal shape and size, and some exhibited deposition of paracrystalline material in the matrix. An abnormal deposition of stippled material intercellularly was also observed. These changes were interpreted as signs of an abnormal secretory function.

Initial aspects of mineralization at the dentino-enamel junction in embryonic mouse incisor in vivo and in vitro: a tem comparative study.

The frontier between the enamel organ and the dental papilla, the future dentino-enamel junction, undergoes coordinated modifications. The mineralization of the extracellular matrix starts within the predentine, which is a prerequisite for the formation of the first enamel crystallites in vivo. We investigated the dentino-enamel junction using the embryonic mouse incisor as a model. Our data showed that the notion of the dentino-enamel junction should not be restricted to the thin interface classically described. A temporo-spatial survey from the epithelio-mesenchymal junction to the dentino-e

Dental mineralization.

Extracellular matrix components and cell-derived microstructures are implicated in mineralization processes which occur in dental tissues. The respective role(s) of collagenic and non-collagenic matrix components are reviewed: phosphorylated and non-phosphorylated proteins, proteoglycans and phosphpholipids. Space-filling amphiphilic molecules seem to play an important role in the preorganization and oriented deposition of calcium phosphate on structures serving more or less as passive support in dentine as well as in enamel.

The effects of induced prenatal hypothyroidism on lamb mandibular third primary molars.

Intrauterine thyroidectomies were performed on nine lambs on or about the ninety-sixth postconception day. Seven other control and shamoperated lambs, and the cretin lambs were sacrificed immediately after birth. The mandibles were removed and sectioned at the midline. The right side molars were removed by dissection and caliper measured. The distal cusps of the third primary molars were sectioned, dehydrated, and embedded in Bioplastic. A slow speed diamond saw was used to section the plastic blocks and the embedded teeth. Subsequent grinding and polishing produced high quality 75 micrometer

Upper central incisor and canine tooth crown size in 47,XXY males.

Enamel and dentin thicknesses were measured in permanent tooth crowns of 47,XXY (Klinefelter syndrome) males. In 47,XXY males, enamel thickness in maxillary central incisors was significantly greater than that in control males or females, and dentin thickness in incisors and canines was significantly greater than that in control females, but smaller than that in normal males. These findings confirm the concept that human dental growth is affected by sex chromosomes. Amelogenesis is promoted by both X and Y chromosome genes, and sexual dimorphism in average tooth size is evidently determined by…

Long term results of chemotherapy on the developing dentition: caries risk and developmental aspects.

The dental health of 45 children who had been placed on long term evaluation after chemotherapy treatment for malignancies was examined in this study. All children had received cytotoxic drugs during the period of tooth formation. It was found that they had more filled or diseased permanent teeth than control children. Their current caries activity as indicated by initial white spot lesions was also higher. Forty-three of the children showed evidence of disturbed amelogenesis. This had resulted in aesthetically displeasing grooves, pits and discoloration. Twenty-three of the children were coun…
