High purity and recovery of native filamentous hemagglutinin (FHA) from Bordetella pertussis using affinity chromatography.

Filamentous hemagglutinin (FHA) is a critical adhesion molecule produced by Bordetella pertussis (BP), the causative agent of highly contagious respiratory infection known as whooping cough. FHA plays a pivotal role in the pathogenesis of whooping cough and is a key component of acellular pertussis vaccines (aPV). However, conventional purification methods for FHA often involve labor-intensive processes and result in low purity and recovery rates. Therefore, this study explores the use of monoclonal and polyclonal antibodies as specific tools to achieve highly pure and efficient FHA purificati…

Non-specific Effects of Live Attenuated Pertussis Vaccine Against Heterologous Infectious and Inflammatory Diseases.

Bordetella pertussis is the agent of pertussis, also referred to as whooping cough, a disease that remains an important public health issue. Vaccine-induced immunity to pertussis wanes over time. In industrialized countries, high vaccine coverage has not prevented infection and transmission of B. pertussis, leading to periodic outbreaks in people of all ages. The consequence is the formation of a large source for transmission to children, who show the highest susceptibility of developing severe whooping cough and mortality. With the aim of providing protection against both disease and infectio…

Symptoms and complications of pertussis in adults.

There is increasing evidence that pertussis occurs frequently in adults, but there is limited information on the clinical course of this disease beyond childhood. A household contact study on the efficacy of an acellular pertussis vaccine was used to study the symptoms of pertussis in adults. Among 257 patients with pertussis identified in 121 families during a two-year period in one study center with a low whole-cell pertussis-vaccine uptake, 79 (30.7%) were adults, aged 19-83 years (mean age: 36 years) with a 1:1.8 male to female ratio. Ninety-one percent of the adults suffered from coughing

[Immunization of 28 months old children in Geneva, Switzerland: trend over a 6-year period, 1995-2000].

OBJECTIVES: Trend in immunization coverage of 28 months old children in Geneva, 1995-2000. METHODS: Comprehensive analysis of routinely collected child immunization cards. Sample survey of non-responding parents. RESULTS: Among 25,288 children (response rate, 91%) immunization coverage (three doses) was 96% for diphtheria and tetanus, 94% for whooping-cough and poliomyelitis and 84% for Haemophilus influenzae. Immunization coverage was 88% for measles (one dose). There was no significant change over time. CONCLUSIONS: Immunization coverage can be monitored by routinely collecting data from

Transients and attractors in epidemics.

Historical records of childhood disease incidence reveal complex dynamics. For measles, a simple model has indicated that epidemic patterns represent attractors of a nonlinear dynamic system and that transitions between different attractors are driven by slow changes in birth rates and vaccination levels. The same analysis can explain the main features of chickenpox dynamics, but fails for rubella and whooping cough. We show that an additional (perturbative) analysis of the model, together with knowledge of the population size in question, can account for all the observed incidence patterns by…

Recombinant Bordetella pertussis pertactin (P69) from the yeast Pichia pastoris: high-level production and immunological properties.

Acellular whooping cough vaccines are based on pertussis toxoid but their effectiveness may be increased by the addition of other Bordetella pertussis antigens. We expressed the immunogenic outer membrane protein pertactin (P69) from B. pertussis to high levels in multi-copy transformants of the industrial yeast Pichia pastoris. In high-density fermentations, engineered P. pastoris yielded greater than 3 g of the protein per litre of culture. Purified recombinant pertactin was able to stimulate the incomplete protection afforded by toxoid to the level of the whole-cell vaccine, as shown by the…

Occupational hazards in hospitals: risk of infection.

In this review of the risk of infection to hospital staff, attention is drawn to the continuing risk presented by hepatitis B and pulmonary tuberculosis, which are more common than diseases such as typhoid fever, brucellosis, histoplasmosis, whooping cough, infectious gastroenteritis, measles, and parotiditis. Other items considered include the susceptibility of female hospital staff to rubella and the importance of their undergoing screening and vaccination; the risks currently presented by epidemic keratoconjunctivitis and by herpes viruses (herpes simplex, varicella zoster, and cytomegalovi…

Beyond the cough: acute myocarditis following pertussis in an adolescent.

Pertussis is a highly pathogenic respiratory infection that poses a particularly serious threat to unvaccinated children, often leading to severe disease and complications. In adults and adolescents, the condition frequently goes underdiagnosed because its presentation is typically non-specific and can mimic other, more common respiratory illnesses. Although complications affecting the respiratory system are well documented in the literature, cardiac involvement, especially in the form of acute myocarditis, remains a very rare and under-reported condition. Historically, efforts to establish a

[Persistent cough caused by Bordetella pertussis. Study of 6 cases].

BACKGROUND: Pertussis is currently an infrequent illness in Spain, since DTP vaccine was introduced. Immunity acquired after immunization is, nevertheless, lower than immunity acquired after the clinical disease, and decreases steadily. After the adolescence, serum levels of antibodies against B. pertussis may not be enough, leading to atypical infections in other wise immunized patients. METHODS: We inoculated, during 6 months, nasal swabs of patients with symptoms of pertussis and of patients with persistent cough on blood agar, MacConkey agar, chocolate agar, charcoal agar and Bordet Gengo

Comparison of pulsed field gel electrophoresis patterns of Bordetella pertussis isolates from unvaccinated children with severe or mild pertussis.

BACKGROUND: The clinical spectrum of pertussis ranges from mild cough illnesses of short duration to typical illness with paroxysmal cough, whooping and posttussive vomiting. We performed comparative genotyping of Bordetella pertussis isolates from children in Germany with severe or mild pertussis. METHODS: Pulsed field gel electrophoresis (PFGE) of genomic DNA, digested with the restriction endonucleases Xba I and Spe I, was used to determine the profiles of strains isolated from 31 unvaccinated children with severe symptoms of pertussis (with hospitalization and/or complications) compared w

[Principles of antibiotic prophylaxis].

Antibiotic therapy aims to reduce the risk of bacterial infection in a given patient. It can be useful after exposure to a pathogen (invasive meningococcal infections, whooping cough, etc.), before or during surgery, after an infection to prevent recurrence, or in patients who are at a high risk of bacterial infection (most particularly in hematology). The more the targeted pathogenic agent is unique (making use of a narrow-spectrum antibiotic possible), the more exposure is limited in time, the more the disease is potentially serious, and the more the antibiotic is easy to use and well tolera…

Interactions of Bordetella pertussis adenylyl cyclase toxin CyaA with calmodulin mutants and calmodulin antagonists: comparison with membranous adenylyl cyclase I.

The adenylyl cyclase (AC) toxin CyaA from Bordetella pertussis constitutes an important virulence factor for the pathogenesis of whooping cough. CyaA is activated by calmodulin (CaM) and compromises host defense by excessive cAMP production. Hence, pharmacological modulation of the CyaA/CaM interaction could constitute a promising approach to treat whooping cough, provided that interactions of endogenous effector proteins with CaM are not affected. As a first step toward this ambitious goal we examined the interactions of CyaA with wild-type CaM and four CaM mutants in which most methionine re…

[Estimating the coverage of systematic vaccinations among the Balearic Island infant population].

BACKGROUND: In order for vaccination programs to be carried out properly, it is essential for the vaccination coverage to be known. On the Balearic Islands, the reported coverage was much less than for the rest of Spain. The objective of this study is that of estimating the coverage of the vaccinations included on the recommended schedule up to 18 months of age (4 doses of oral polio, tetanus and diphtheria; 3 doses of whooping cough; 1 dose of measles, rubella and mumps). METHODS: A descriptive, cross-sectional study of the population base. The ideal-time frequency distribution estimators an…

Bordetella pertussis binds human C1 esterase inhibitor during the virulent phase, to evade complement-mediated killing.

C1 esterase inhibitor (C1inh) is a major inhibitor of several pathways of inflammation in humans. In this study, we show that virulent-phase cultures of Bordetella pertussis, the etiological agent for whooping cough, but not other Bordetella species specifically recruit C1inh from human serum. Using a spontaneous mutant of B. pertussis that was deficient in C1inh binding, we demonstrate that the ability of B. pertussis to acquire high levels of human C1inh and wild-type levels of serum resistance are well correlated, suggesting that, in addition to and independent of BrkA expression, acquisiti…

Bioengineering of Bordetella pertussis Adenylate Cyclase Toxin for Vaccine Development and Other Biotechnological Purposes.

The adenylate cyclase toxin, CyaA, is one of the key virulent factors produced by Bordetella pertussis, the causative agent of whooping cough. This toxin primarily targets innate immunity to facilitate bacterial colonization of the respiratory tract. CyaA exhibits several remarkable characteristics that have been exploited for various applications in vaccinology and other biotechnological purposes. CyaA has been engineered as a potent vaccine vehicle to deliver antigens into antigen-presenting cells, while the adenylate cyclase catalytic domain has been used to design a robust genetic assay fo…
